This paper reports the first photosynthetic study of marestail in Jiuzhaigou. In this work, we used PAM fluorometry to examine photosynthetic rates of submerged and emerged marestail in three lakes. Three lakes were studied across a gradient of water temperature, with low water temperature conditions in Grass Lake and Arrow Bamboo Lake, and higher water temperature in Five Colored Lake. In the field, electron transport rates (ETRmax) were measured as rapid light curves (RLCs) by in situ yield measurements. Submerged and emerged marestail showed higher photosynthetic activity in Five Colored Lake compared to the other lakes, a response consistent with the adaptation of marestail in Five Colored Lake to high water temperature. The optimal temperature for photosynthesis of submerged marestail in Jiuzhaigou was about 12 °C. Nonphotochemical quenching (NPQ) of submerged and emerged marestail increased with increasing water temperature. Maximum quantum yield (F v/F m) of submerged marestail in Five Colored Lake showed full recovery at 1700 h due to higher NPQ. Further, the chlorophyll a for submerged marestail was the highest in Grass Lake and the lowest in Five Colored Lake. These results indicate that in different lakes the function of these aquatic plants is associated with a diversity of place-dependent environmental conditions, especially water temperature that leads to pronounced differences in the plant’s ecophysiological reactions.  相似文献

Gracilaria lemaneiformis Bory is an economically important alga that is primarily used for agar production. Although tetraspores are ideal seeds for the cultivation of G. lemaneiformis, the most popular culture method is currently based on vegetative fragments, which is labor-intensive and time-consuming. In this study, we optimized the conditions for tetraspore release and evaluated the photosynthetic activities of different colonies formed from the branches of G. lemaneiformis using a PAM (pulse-amplitude-modulated) measuring system. The results showed that variations in temperature and salinityhad significant effects on tetraspore yield. However, variations in the photon flux density (from 15 μmol m−2 s−1 to 480 μmol m−2 s−1) had no apparent effect on tetraspore yield. Moreover, the PAM-parameters Y(I), Y(II), ETR(I), ETR(II) and F v/F m of colonies formed from different branches showed the same trend: parameter values of first generation branches>second generation branches>third generation branches. These results suggest that the photosynthetic activities of different colonies of branches changed with the same trend. Furthermore, photosynthesis in G. lemaneiformis was found to be involved in vegetative reproduction and tetraspore formation. Finally, the first generation branches grew slowly, but accumulated organic compounds to form large numbers of tetraspores. Taken together, these results showed that the first generation branches are ideal materials for the release of tetraspores.  相似文献

Fluorescence-based maximal photochemical efficiency, Fv/Fm, is widely used as an indicator to photosynthetic competence in marine systems. It has been considered a useful parameter diagnosing the nutrient stress on phytoplankton photosynthesis, but many studies argue its usefulness. In the present study, we try to find a temporal relationship between Fv/Fm and nitrogen concentration, and provide a possible explanation on the controversy. We continuously measured Fv/Fm and nitrogen concentration once every 10 days from September 2003 to March 2004 at two stations in Jiaozhou Bay, northern China. It was found that Fv/ffm did not significantly correlate to synchronous nitrogen concentration, but the variation (i.e. the change between two adjacent cruises) of nitrogen concentration of the previous cruise and the variation ofFv/Fm of the current cruise were strongly correlated. This result indicates that a time lag exists between the variation of nutrient status and the subsequent Fv/Fm response. Length of the time lag seems just matched the interval of our measurements (10 days). In the field, direct dependence of Fv/Fm on nitrogen concentration may not be found because of the lagged response of Fv/Fm to nitrogen concentration variations or physiological acclimation. Our results provide a possible way to explain the previously reported conflicting results on the relationship between Fv/Fm and nutrient status. To give a more-accurate estimate about the length of the time lag, an investigation that includes more frequent measurements is needed.  相似文献

Nutrient enrichment experiments with nitrogen (N) and phosphorus (P) were conducted with samples from two stations in the coastal waters of Qingdao, China, during summer to identify limiting nutrients. In late July of 2009, low P concentrations and the maximum photochemical efficiency of photosystem II (F v /F m ) in the initial samples together with F v /F m and chlorophyll a (Chl a) responses to P addition indicated P limitation at the two stations. In early August, low P levels still limited phytoplankton growth at station A. F v /F m and Chl a were the highest in the NP treatments at station B, suggesting an N/P co-limitation. In mid-September, nutrient concentrations and F v /F m were elevated and phytoplankton communities were healthy. Greater F v /F m and Chl a in the treatments with added P than those without the addition suggested potential P limitation at station A. Lack of F v /F m and Chl a responses following nutrient additions indicated N and P repletion at station B. At the end of July 2010, neither N nor P was limited at station B. Additionally, F v /F m coupled with 24-h-long nutrient enrichment experiments can be used to detect P limitation and N/P co-limitation to natural populations. This method can be more accurate for assessing co-limitation than the use of criteria of nutrient concentrations and ratios as indicators, and can provide more rapid results than nutrient addition bioassays using chlorophyll response as an indicator, when a population is potentially limited. Compared with the two conventional methods, the results based on F v /F m can also provide more detailed information about physiological states of the populations.  相似文献

The antioxidative capacity of astaxanthin and enzyme activity of reactive oxygen eliminating enzymes such as superoxide dismutase (SOD), peroxidase (POD), catalase (CAT) and ascorbate peroxidase (APX) were studied in three cell types of Haematococcus pluvialis exposed to high concentrations of a superoxide anion radical (O2). The results show that defensive enzymes and astaxanthin-related mechanisms were both active in H. pluvialis during exposure to reactive oxygen species (ROS) such as O2. Astaxanthin reacted with ROS much faster than did the protective enzymes, and had the strongest antioxidative capacity to protect against lipid peroxidation. The defensive mechanisms varied significantly between the three cell types and were related to the level of astaxanthin that had accumulated in those cells. Astaxanthin-enriched red cells had the strongest antioxidative capacity, followed by brown cells, and astaxanthin-deficient green cells. Although there was no significant increase in expression of protective enzymes, the malondialdehyde (MDA) content in red cells was sustained at a low level because of the antioxidative effect of astaxanthin, which quenched O2 before the protective enzymes could act. In green cells, astaxanthin is very low or absent; therefore, scavenging of ROS is inevitably reliant on antioxidative enzymes. Accordingly, in green cells, these enzymes play the leading role in scavenging ROS, and the expression of these enzymes is rapidly increased to reduce excessive ROS. However, because ROS were constantly increased in this study, the enhance enzyme activity in the green cells was not able to repair the ROS damage, leading to elevated MDA content. Of the four defensive enzymes measured in astaxanthin-deficient green cells, SOD eliminates O2, POD eliminates H2O2, which is a by-product of SOD activity, and APX and CAT are then initiated to scavenge excessive ROS.  相似文献

We studied the effects of nitrogen (N) and phosphorus (P) supply on Fv/Fm (maximal quantum yield of photosystem II) in the algae Chaetoceros debilis, Dicrateria inornata, Platymonas subcordiformis and Heterosigma akashiwo to determine the sensitivity of Fv/Fm to P-limitation of the four species. Obvious decrease of Fv/Fm value was monitored in periods of P-depletion, the decrease showing different magnitudes among algal species. A more clear decrease of Fv/Fm in Platymonas subcordiformis and Het-erosigma akashiwo was observed, compared with that in Chaetoceros debilis and Dicrateria inornata. After the resupply of P, the index Fv/Fm of the four species all recovered quickly in 24 h. Cell division was maintained and chlorophyll content continued to increase until the end of the experiment in Platymonas subcordiformis and Heterosigma akashiwo, while in Chaetoceros debilis and Dicrateria inornata, the division stopped in the later period when the growth of organisms came into the stationary phase. The most obvious respond of chlorophyll content to P-additions was observed in Het-erosigma akashiwo culture in 24 h. The sensitivity of Fv/Fm to nutrient limitations is different among algal species, and much should be done to improve the application of this index.  相似文献

In this paper, we investigated the effects of temperature, oxygen, antioxidants, and corn germ oil on the stability of astaxanthin from Haematococcus pluvialis under different storage conditions, and changes in the composition of astaxanthin esters during storage using high performance liquid chromatography and spectrophotometry. Oxygen and high temperatures (22-25~C) significantly reduced the stability of astaxanthin esters. Corn germ oil and antioxidants (ascorbic acid and vitamin E) failed to protect astaxanthin from oxidation, and actually significantly increased the instability of astaxanthin. A change in the relative composition of astaxanthin esters was observed after 96 weeks of long-term storage. During storage, the relative amounts of free astaxanthin and astaxanthin monoesters declined, while the relative amount of astaxanthin diesters increased. Thus, the ratio of astaxanthin diester to monoester increased, and this ratio could be used to indicate if astaxanthin esters have been properly preserved. If the ratio is greater than 0.2, it suggests that the decrease in astaxanthin content could be higher than 20%. Our results show that storing algal powder from H. pluvialis or other natural astaxanthin products under vacuum and in the dark below 4~C is the most economical and applicable storage method for the large-scale production of astaxanthin from H. pluvialis. This storage method can produce an astaxanthin preservation rate of at least 80% after 96 weeks of storage.  相似文献

To increase the cell concentration and the accumulation of astaxanthin in the cultivation of Haematococcus pluvialis, effects of different iron electrovalencies (Fe2+-EDTA and Fe3+-EDTA) and species (Fe-EDTA, Fe(OH)x32x and FeC6H5O7) addition on cell growth and accumulation of astaxanthin were studied. Results show that different iron electrovalencies have various effects on cell growth and astaxanthin accumulation of H. pluvialis. Compared with Fe3+-EDTA, Fe2+-EDTA stimulate more effectively the formation of astaxanthin. The maximum astaxanthin content (30.70 mg/g biomass cell) was obtained under conditions of 18 μmol/L Fe2+-EDTA, despite the lower cell density (2.3×105 cell/ml) in such condition. Fe3+-EDTA is more effective than Fe2+-EDTA in improving the cell growth. Especially, the maximal steady-state cell density, 2.9×105 cell/ml was obtained at 18 μmol/L Fe3+-EDTA addition. On the other hand, all the various species of iron (EDTA-Fe, Fe(OH)x32x, FeC6H5O7) are capable to improve the growth of the algae and astaxanthin production. Among the three iron species, FeC6H5O7 performed the best. Under the condition of a higher concentration (36 μmol/L) of FeC6H5O7, the cell density and astaxanthin production is 2 and 7 times higher than those of iron-limited group, respectively. The present study demonstrates that the effects of the stimulation with different iron species increased in the order of FeC6H5O7, Fe(OH)x32x and EDTA-Fe. Two strains H2-410 and H2-419 were obtained from the chemically mutated survivors of wild Haematococcus pluvialis 2 by using ethyl methanesulphonate (EMS). Strains H2-410 and H2-419 showed a fast cell growth with 13% and 20% increase in biomass compared to wild type, respectively. Then H2-419-4, a fast cell growth and high astaxanthin accumulation strain, was obtained by exposing the strain H2-419 to ultraviolet radiation (UV) further. The total biomass, the astaxanthin content per cell, astaxanthin production of H2-419-4 showed 68%, 28%, and 120% increase compared to wild H. pluvialis 2, respectively. HPLC (High Performance Liquid Chromatography) data showed also an obvious proportional variation of different carotenoid compositions in the extracts of H2-419-4 and the wild type, although no peak of carotenoids appeared or disappeared. Therefore, the main compositions in strain H2-419-4, like its wild one, were free of astaxanthin, monoester, and diester of astaxanthin. The asexual reproduction in survivors after exposed to UV was not synchronous, and different from the normal synchronous asexual reproduction as the mother cells were motile instead of non-motile. Interestingly, some survivors from UV irradiation produced many mini-spores (or gamete?), the spores moved away from the mother cell gradually 4 or 5 days later. This is quite similar to sexual reproduction described by Elliot in 1934. However, whether this was sexual reproduction remains questionable, as no mating process has been observed.  相似文献

Saccharina japonica is a common macroalga in sublittoral communities of cold seawater environments,and consequently may have highly effi cient ribulose-1,5-bisphosphate carboxylase/oxygenase(Rubisco)activity for carbon assimilation.In our study,we cloned the full-length Rubisco gene from S.japonica(SJ-rbc).It contained an open reading frame for a large subunit gene(SJ-rbcL)of 1 467 bp,a small subunit gene(SJ-rbcS)of 420 bp,and a SJ-rbcL /S intergenic spacer of 269 bp.The deduced peptides of SJ-rbcL and SJ-rbcS were 488 and 139 amino acids with theoretical molecular weights and isoelectric points of 53.97 kDa,5.81 and 15.84 kDa,4.71,respectively.After induction with 1 mmol/L isopropyl-β-Dthiogalactopyranoside for 5 h and purifi cation by Ni 2+ affi nity chromatography,electrophoresis and western blot detection demonstrated successful expression of the 55 kDa SJ-rbcL protein.Real-time quantitative PCR showed that the mRNA levels of SJ-rbcL in gametophytes increased when transferred into normal growth conditions and exhibited diurnal variations: increased expression during the day but suppressed expression at night.This observation implied that Rubisco played a role in normal gametophytic growth and development.In juvenile sporophytes,mRNA levels of SJ-rbcL,carbonic anhydrase,Calvin-BensonBassham cycle-related enzyme,and chloroplast light-harvesting protein were remarkably increased under continuous light irradiance.Similarly,expression of these genes was up-regulated under blue light irradiance at 350 μmol/(m 2·s).Our results indicate that long-term white light and short-term blue light irradiance enhances juvenile sporophytic growth by synergistic effects of various photosynthetic elements.  相似文献

We investigated the effects of dried macroalga Gracilaria lemaneiform (Rhodophyta) on photosynthesis of the bloom-forming microalga Chaetoceros curvisetus. C. curvisetus was cultured with different amounts of dried G. lemaneiformis under controlled laboratory conditions. We measured the photosynthetic oxygen evolution rate and established the chlorophyll a fluorescence transient (OJIP) curve coupled with its specific parameters. We observed concentration-dependent and time-dependent relationships between dried G. lemaneiformis and inhibition of photosynthesis in C. curvisetus. Co-culture with dried G. lemaneiformis also resulted in a decrease in the light-saturated maximum photosynthetic oxygen evolution rate (P~ax) in C. curvisetus, and a decrease in the OJIP curve along with its specific parameters; the maximum photochemical efficiency of PSII (FJFm), the amount of active PSII reaction centers per excited cross section at t=0 and t=--tFM (RC/CS0 and RC/CSm, respectively), the absorption flux per excited cross section at t=0 (ABS/ CS0), and the efficiency with which a trapped exciton moves an electron into the electron transport chain (~u0). The dark respiration rate (Rd) increased in C. curvisetus co-cultured with dried G. lemaneiformis. The JIP-test and the oxygen evolution results indicated that dried G. lemaneiJbrmis decreased the number of active reaction centers, blocked the electron transport chain, and damaged the oxygen-evolving complex of C. curvisetus. This result indicated that dried fragments of G. lemaneiformis could effectively inhibit photosynthesis of C. curvisetus, and thus, could serve as a functional product to control and mitigate C. curvisetus blooms.  相似文献

Most studies on Haematococcus pluvialis have been focused on cell growth and astaxanthin accumulation;far less attention has been paid to cell cycles and proliferation patterns.The purpose of this study was to clarify cell cycles and proliferation patterns in H.pluvialis microscopically using a camera and video recorder system.The complicated life history of H.pluvial i s can be divided into two stages:the motile stage and the non-motile stage.All the cells can be classifi ed into forms as follows:motile cell,nonmotile cell,zoospore and aplanospore.The main cell proliferation,both in the motile phase and non-motile phase in H.pluvialis,is by asexual reproduction.Under normal growth conditions,a motile cell usually produces two,sometimes four,and exceptionally eight zoospores.Under unfavorable conditions,the motile cell loses its fl agella and transforms into a non-motile cell,and the non-motile cell usually produces 2,4 or 8 aplanospores,and occasionally 20–32 aplanospores,which further develop into non-motile cells.Under suitable conditions,the non-motile cell is also able to release zoospores.The larger non-motile cells produce more than 16 zoospores,and the smaller ones produce 4 or 8 zoospores.Vegetative reproduction is by direct cell division in the motile phase and by occasional cell budding in the non-motile phase.There is,as yet,no convincing direct evidence for sexual reproduction.  相似文献

The effects of different phosphorus (P) concentrations (0.36, 3.6, and 36 μmol/L corresponding to low-, middle-, and high-P concentration groups, respectively) and nitrogen (N)/P ratios on the growth and photosynthetic characteristics of Skeletonema costatum and Prorocentrum donghaiense were studied. For both species, the high-P (HP) concentration group showed the greatest algal density and highest specific growth rate. Changes in the maximum efficiency of photosystem II (F v /F m ) were monitored under the various P and N/P conditions. The largest decrease in F v /F m was in the low-P (LP) group in S. costatum and in the HP group in P. donghaiense. There were high rapid light curves and photochemical quantum yields (Φ PSII) for S. costatum in the HP group, while the actual photosynthetic capacity was higher in P. donghaiense than in S. costatum in the MP group. Under eutrophic but relatively P-restricted conditions, P. donghaiense had higher photosynthetic activity and potential, which could cause this dinoflagellate to increasingly dominate the phytoplankton community in these conditions. Under the same P concentration and N/P ratio, P. donghaiense had a larger relative maximum rate of electron transport and higher Φ PS II values than those of S. costatum. These differences between P. donghaiense and S. costatum may explain the interaction and succession patterns of these two species in the Changjiang (Yangtze) River estuary from a photosynthesis perspective.  相似文献

This study on dynamic changes of culture color,astaxanthin and chlorophylls,inorganic N including N-NO3^-,N-NO2^- and N-NH4^ in batch culture of Haematococcus pluvialis exposed to different additive nitrate concentration showed(1)ast/chl ratio was over 0.8 for brown and red algae,but was usually less than 0.5 for green and yellow algae;(2)N-NO3^-,in general,was unstable and decreased,except for a small unexpected increase in nitrate enriched treatment groups;(3)measurable amounts of N-NO2^- and N-NH4^ were observed respectively with three change modes although no extemal nitrite and ammonia were added into the culture;(4)a non-linear correlation between ast/chl ratio(or color)changes and the levels of N-NO3^-,N-NO2^-,N-NH4^ in H.pluvialis culture;(5)up and down variation of the ast/chl ratio occurred simultaneously with a perceptible color change from yellow to brown(or red)when N-NO3^-,N-NO2^- and N-NH4^ fluctuated around 30,5,5μmol/L respectively;(6)existence of three dynamic modes of N-NO3^-,N-NO2^- and N-NH4^ changes,obviously associated with initial extemal nitrate;(7)the key level of total inorganic N concentration regulating the above physiological changes during indoor cultivation was about 50 μmol/L;and(8)0.5-10mmol/L of nitrate was theoretically conducive to cell growth in batch culture.  相似文献

The genus Cylindrotheca consists of a small group of marine diatoms with a few species described. Eleven isolates of diatoms identified as Cylindrotheca closterium morphologically were obtained from Jiaozhou Bay with their nuclear-encoded small-subunit ribosomal RNA (SSU rDNA) and chloroplast-encoded rbcL gene sequences determined in this study. Interestingly, very high sequence divergences of SSU rDNA and rbcL gene were found among these isolates, and numerous nucleotide variation of rbcL gene caused relatively few variation of deduced amino acid sequence. Phylogenetic analyses based on SSU rDNA and rbcL gene, respectively, grouped the isolates into 6 clades. Phylogenetic tree of SSU rDNA placed all the Cylindrotheca isolates together, separating them into two lineages clearly. LineageⅠ was composed of the eleven C. closterium isolates obtained in this study together with another C. closterium isolate, but some clades were not well supported. LineageⅠwas contained two C. closterium isolates and one C. fusiformis isolate. Phylogenetic analysis of rbcL gene also separated the Cylindrotheca isolates into two well-defined lineages. The eleven C. closterium isolates formed a lineage and all clades were supported strongly. Statistical comparisons of SSU rDNA indicated that the average distance within lineageⅠwas significantly higher than that of other microalgae species (P 〈0.01). These results suggested the existence of cryptic species within C. closterium.  相似文献

Field investigations and aerial photography after the earthquake of May 12,2008 show a large number of geo-hazards in the zone of extreme earthquake effects.In particular,landslides and debris flows,the geo-hazards that most threaten post-disaster reconstruction,are widely distributed.We describe the characteristics of these geo-hazards in Beichuan County using high-resolution remote sensing of landslide distribution,and the relationships between the area and volume of landslides and the peak-discharges of debris flows both pre-and post-earthquake.The results show:1) The concentration(defined as the number of landslide sources per unit area:Lc) of earthquaketriggered landslides is inversely correlated with distance from the earthquake(DF) fault.The relationship is described by the following equation:Lc = 3.2264exp(-0.0831DF)(R2 = 0.9246);2) 87 % of the earthquake-triggered landslides were less than 15×104 m2 in area,and these accounted only for 50% of the total area;84% of the landslide volumes were less than 60×104 m3,and these accounted only for 50% of the total volume.The probability densities of the area and volume distributions are correlated:landslide abundance increases with landslide area and volume up to maximum values of 5 × 104 m2 and 30 × 104 m3,respectively,and then decreases exponentially.3) The area(AL) and volume(VL) of earthquake-triggered landslides are correlated as described with the following equation:VL=6.5138AL1.0227(R2 = 0.9131);4) Characteristics of the debris flows changed after the earthquake because of the large amount of landslide material deposited in the gullies.Consequently,debris flow peak-discharge increased following the earthquake as described with the following equation:Vpost = 0.8421Vpre1.0972(R2 = 0.9821)(Vpre is the peak discharge of pre-earthquake flows and the Vpost is the peak discharge of post-earthquake flows).We obtained the distribution of the landslides based on the above analyses,as well as the magnitude of both the landslides and the post-earthquake debris flows.The results can be useful for guiding post-disaster reconstruction and recovery efforts,and for the future mitigation of these geo-hazards.However,the equations presented are not recommended for use in site-specific designs.Rather,we recommend their use for mapping regional seismic landslide hazards or for the preliminary,rapid screening of sites.  相似文献

The adsorption/desorption of phosphorus across the sediment-water interface is the main mechanism that controls the buffering capacity of DRP in coastal and estuarine water. Determination by three processes revealed that: 1) DRP flux from early diagenesis in surface sediment isF p Dia =31.9μg/(m2·d), which is only 1/1600 of the ammonia released; 2) at normal temperature and in the long term, the TDP released by adsorption/desorption across the sediment-water interface is a function of the major environmental factors; and 3) based on the determined SODR and assumed average water depthH(m) and residence timet R (d), the maximum flux of DRP can be estimated. Grateloupia ramosa Wang & Luan sp. nov. (Halymeniaceae, Rhodophyta) is newly described from Hainan Province, southern China. The organism has the following morphological features: (1) purplish red, cartilaginous and lubricous thalli 5–10 cm in height; (2) compressed percurrent axes bearing abundant branches with opposite arrangement; (3) claw-like apices on top, constricted to 2–4 cm at the base; (4) cortex consisting of 3–6 layers of elliptical or anomalous cells and a medulla covered by compact medullary filaments; (5) reproductive structures distributed throughout the thallus, especially centralized at the bottom of the end portion of the branches; and (6) 4-celled Carpogonial branches and 3-celled auxiliary-cell branches, both of the Grateloupia-type. The morphological diff erences were supported by molecular phylogenetics based on ribulose-1, 5-bisphosphate carboxylase/oxygenase (rbcL) gene sequence analysis. There was only a 1 bp divergence between specimens collected from Wenchang and Lingshui of Hainan province. The new species was embedded in the large Grateloupia clade of the Halymeniaceae. The pairwise distances between G. ramosa and other species within Grateloupia ranged from 26 to 105 bp, within pairwise distances of 13–111 bp between species of the large genus Grateloupia in Halymeniaceae. Thus, we propose this new species as G. ramosa Wang & Luan sp. nov.  相似文献

Chlorophyta species are common in the southern and northern coastal areas of China. In recent years, frequent green tide incidents in Chinese coastal waters have raised concerns and attracted the attention of scientists. In this paper, we sequenced the 18S rDNA genes, the internal transcribed spacer (ITS) regions and the rbcL genes in seven organisms and obtained 536-566 bp long ITS sequences, 1 377-1 407 bp long rbcL sequences and 1 718-1 761 bp long partial 18S rDNA sequences. The GC base pair content was highest in the ITS regions and lowest in the rbcL genes. The sequencing results showed that the three Ulva prolifera (or U. pertusa) gene sequences from Qingdao and Nan’ao Island were identical. The ITS, 18S rDNA and rbcL genes in U. prolifera and U. pertusa from different sea areas in China were unchanged by geographic distance. U. flexuosa had the least evolutionary distance from U. californica in both the ITS regions (0.009) and the 18S rDNA (0.002). These data verified that Ulva and Enteromorpha are not separate genera.  相似文献

Based on DNA extraction and optimization of random amplified reaction (RAPD) to the gametophytes and sporophytes of Kelp “901” strain, genetic study on variation was conducted to its parents and offsprings of F6, F7, F8, and F9 generation. RAPD results have shown that among 30 selected primers for gametophytes, 297 loci ranging from 200 to 3 000 bp were obtained in the average of 9.9 loci for each primer. This indicated a high polymorphic rate with RAPD detection. UPGMA (unweighted pair-group method arithmetic average) analysis showed that each male and female gametophyte of a generation could be clustered into one pair separately. The genetic distances of the Kelp 901 generation were 0.3212–0.4767, and the maximum was between F7 and F8 (0.4767). Identity analysis showed that F6 generation was more close to the female parent (0.6593), and F7 generation was more close to the male parent (0.578 8). To the sporophytes study in 24 selected primers for RAPD amplification, 191 loci ranging from 230–2800 bp were obtained, in the average to each primer of 8.0 loci. The heterozygosity to six populations were male parent (0.2239), female parent (0.1072), F6 (0.2164), F7(0.2286), F8(0.2296) and F9(0.3172). The nearest genetic distance was 0.083 5(F8, F9). Total heterozygosity (HT) of F6, F7, F8 and F9 generations was 0.3186, the average heterozygosity (HS) for F6, F7, F8 and F9 generations was 0.2480, and deduced coefficient of population differentiation (Gst) was 22.2%. Six sequence characterized amplified regions (SCAR) were preliminary screened through RAPD analysis. It needed to be verified in detail as they are significant for molecular marker assistance in breeding and selectingLaminaria.  相似文献
