镉对中华绒螯蟹鳃组织及其亚显微结构的影响   总被引:16,自引:2,他引:16

于1986年10月—1987年5月以不同浓度的镉(6.25,12.5和25.0mg/L),对采自长江口的中华绒螯蟹进行了急性毒性试验,以光镜和电镜观察。结果表明,受镉作用后,整个鳃叶增厚,血腔增大;血细胞增多,上皮层破坏直至解体;鳃上皮细胞的亚显微结构因镉的作用顶膜微绒毛和底膜内折消失,线粒体膨胀直至解体。以统计分析检验了对照组和不同镉浓度组处理的蟹鳃之间的形态指标的差异,并分析了这些形态指标的变化同镉浓度的相关性。通过对鳃上皮细胞与鳃生理功能关系的分析,认为镉对鳃组织及细胞结构的损害影响了鳃组织的能量代谢、离子主动转运机制及渗透压调节能力。  相似文献

作者提出镉对中华绒螯蟹肝胰脏R—细胞亚显微结构和血清谷丙转氨酶活力影响。在亚显微结构水平上,R—细胞中,受影响变化最明显的是线粒体;线粒体体积和其内膜嵴的破坏程度与介质镉浓度成显著的正相关关系;随镉浓度的升高,细胞器减少,内质网膨胀,细胞质和线粒体基质变稀薄。SGPT活力也随镉浓度升高而显著增大,并与R—细胞亚显微结构变化程度一致。本文还就处于不同镉浓度下的标本,与生理机能相联系的R—细胞形态变化的意义、R-细胞线粒体亚显微结构变化与SGPT活力变化的关系及SGPT活性变化的特点等进行了初步探讨。  相似文献

用透射电镜观察,发现锯齿溪蟹心肌细胞属横纹肌。粗丝和细丝排列成明带和暗带相间的横纹,每一条粗丝周围有6条细丝组成一个规则的6边形图案,但粗丝是中空的,细丝则是实心结构,且肌原纤维在心肌细胞体积中所占比例较少,线粒体的嵴较短,线粒体排列疏松。心肌细胞之间没有形成象脊椎动物那样的闰盘结构,因此,这些好构就造成了锯齿溪蟹的心脏功能比较低等。  相似文献

提要采用人工染毒的方法,进行了镉污染暴露对大弹涂鱼血清转铁蛋白(STF)铁结合能力和血红细胞影响的研究。结果表明,自染毒第7天开始,血清铁浓度,血清铁饱和度稳定上升并与对照组比较有统计学上的极其显著性差异,且对镉敏感。而转铁蛋白的总铁结合力和不饱和铁结合力则无稳定的趋势。血红蛋白的A值与血细胞浓度均呈稳定下降趋势,并于染毒11天后开始出现显著性降低。揭示镉胁迫引起的大弹涂鱼机体损伤与它对动物机体的铁代谢影响紧密相关。大弹涂鱼血清的铁饱和度、血清铁浓度、血红细胞的吸光度和浓度可以作为长期海洋污染暴露的生物标记,其中血清的铁饱和度可靠度最高,而血红细胞的吸光度和浓度的测定最为简便。推测,其他鱼类在镉胁迫下也应当有相似的血液生理变化,在明确健康对照的相关指标变化范围后,可以便利地监控环境中的重金属污染。  相似文献

栉孔扇贝对镉的富集及释放规律的研究   总被引:1,自引:0,他引:1  
通过暴露染毒方法研究了栉孔扇贝对镉离子(Cd2+)的35d富集与30d释放规律。结果表明:栉孔扇贝对镉有很强的富集性,体内的镉含量与暴露溶液里的Cd2+浓度呈正相关;栉孔扇贝对镉的释放速率随着暴露水体中镉浓度的增加而减慢。应用双箱动力学模型,通过非线性拟合得到了栉孔扇贝对Cd2+的吸收速率常数k1随着外部水体中镉浓度的增大而减小,生物富集系数BCF则随着外部水体中镉浓度的增大而减少。当镉浓度为0.05mg/L时BCF仅为3 730,而当镉浓度为0.000 5mg/L时BCF达到1.79×1011。平衡状态下栉孔扇贝体内镉含量(CAmax)随着水体中镉浓度增加急剧下降,与水体中镉浓度呈负相关;排出速率常数k2、生物学半衰期B1/2与低浓度镉无明显相关性,随着镉浓度的增大k2显著降低而B1/2显著增大。验证了双箱动力学模型适用于栉孔扇贝对Cd2+的生物富集与释放规律研究。  相似文献

比较中华绒螯蟹、锯缘青蟹、三疣梭子蟹和长江华溪蟹胃、肠、肝胰腺的五种消化酶(胃蛋白酶、类胰蛋白酶、脂肪酶、淀粉酶、纤维素酶)活力,结果表明四种蟹的类胰蛋白酶比活力是胃蛋白酶的7~17倍,淀粉酶是纤维素酶的30~183倍,脂肪酶活力较低。消化酶活力大小具有器官特异性和种间差异。  相似文献

二苯甲酮-3 (BP-3)、4-甲基苄亚基樟脑(4-MBC)和4-甲氧基肉桂酸-2-乙基己酯(EHMC)是三种常用的有机紫外吸收剂, 在水环境中被频繁检出, 对水生生态系统安全构成潜在威胁。为探究三种有机紫外吸收剂对菲律宾蛤仔(Ruditapes philippinarum)鳃组织抗氧化响应和相关细胞凋亡基因的影响, 将蛤仔分别暴露于环境相关浓度的三种紫外吸收剂溶液中, 检测鳃组织抗氧化酶活性和细胞凋亡相关基因转录水平, 并通过第二代整合生物标志物响应法(IBRv2)对三种紫外吸收剂的生物毒性进行比较分析。结果显示, 三种紫外吸收剂短期暴露会诱导抗氧化响应提高抗氧化能力, 而长期高浓度暴露会导致抗氧化能力的降低。BP-3、4-MBC和EHMC可能通过启动线粒体途径和fas介导的死亡受体途径诱导菲律宾蛤仔鳃组织产生细胞凋亡。通过IBRv2分析发现, 在环境常见浓度1 μg/L的暴露水平下, 短期(1 d, 7 d)暴露时, BP-3对菲律宾蛤仔鳃组织表现出的综合毒性效应最强, 而随着暴露时间的延长(28 d), 三种紫外吸收剂表现出的综合毒性效应相近。研究结果为水环境中有机紫外吸收剂的生态风险评估提供了参考数据。  相似文献

为了了解苯并[a]芘(BaP)对鱼类细胞色素P4501A1(CYP1A1)表达的影响,以褐菖鲉(Sebasticus marmoratus)为实验材料,采用体内实验,研究其在经过不同浓度(0.1、1、10、20、50mg/kg鱼体重量)的BaP诱导后,鱼体肝脏研究CYP1A1基因表达的情况,筛选出后续时间-效应实验中BaP注射的最佳浓度,研究BaP诱导6h、12h、1d、3d、7d后(质量浓度为20mg/kg鱼体重量)鱼体肝脏CYP1A1酶活性、基因表达和蛋白表达的情况。结果表明:剂量-效应实验中,20mg/kg鱼体重量为最佳浓度,此浓度下,基因表达在各组中变化最显著。时间-效应实验中,较空白对照组而言,染毒6h、12h和1d后,EROD酶活性显著增加。3d后开始下降,与对照组相比变化不大,7d后酶活性又发生上调。半定量RT-PCR结果表明,各染毒组与对照组相比,CYP1A1基因表达量都发生了上调,呈现先上升后下降的趋势。其中,6h和12h组相对表达量极显著增加,1d后开始下降且与3d和7d组相比变化不明显。Western blot结果表明,蛋白表达量在染毒12h后表现出显著的诱导效应,随着时间的延长略有回落,但与对照组相比仍有显著性差异。研究表明:BaP对褐菖鲉CYP1A1具有较强的诱导作用。一定质量浓度的BaP注射于褐菖鲉不同的时间后,能诱导褐菖鲉活体EROD酶活性、CYP1A1基因m RNA表达及蛋白表达,并随着时间的延长呈现先诱导后抑制的趋势。这说明BaP作为诱导剂对CYP1A1酶活性和蛋白表达的作用机制可能与调控CYP1A1的转录水平有关。  相似文献

为了探究化学诱变剂镉在海洋食物链中的传递规律及其与人类健康的关系,作者设计了从海洋浮游藻类——褐指藻(Phaeodactylum tricornutum Bohlin)~海湾扇贝(Argopecten irradians Lamarck)的实验室食物链,采用极谱分析方法研究了镉的传递,并对贝肉(软体组织)灰样进行了紫露草微核(Trad-MCN)测定,结果:(1)经过6d的传递实验,处理组与对照组贝肉中镉的浓度均未超过对应组褐指藻中镉的浓度;经过这条食物链的传递,镉的浓度减少。(2)通过t测验发现,两个高浓度处理组与对照组贝肉灰样之间的诱变性差异显著(P<0.001与P<0.005)。t值的大小与镉的浓度具有一致性关系,说明镉的浓度越大,诱变性越强。  相似文献

以大弹涂鱼为研究对象,应用单细胞凝胶电泳技术并结合外周血基因组DNA琼脂糖凝胶电泳,研究了镉胁迫对大弹涂鱼外周血细胞的遗传损伤。结果表明:(1)镉对大弹涂鱼外周血细胞是遗传毒性而非细胞毒性,并且产生的遗传损伤存在显著(P0.01或P0.05)的剂量效应,染毒1h的大弹涂鱼的全长和尾长与浓度的相关方程分别是y=29.592x-10.576(r2=0.9786),y=28.417x-14.859(r2=0.9857),染毒2h的大弹涂鱼的全长和尾长与浓度的相关方程分别是y=32.044x-5.0235(r2=0.941),y=29.911x-9.539(r2=0.9635)。(2)DNA的损伤程度和污染胁迫之间存在着时间效应。以上结果表明,DNA损伤可作为大弹涂鱼受镉胁迫时的生物指标。  相似文献

通过设置15℃、20℃、25℃、30℃4个干露温度梯度,检测不同发育阶段三疣梭子蟹幼体的露空时间、死亡率、含水量、失水率及体重消耗率等指标,分析干露温度、发育阶段与幼体死亡率的关系。结果表明,各发育阶段的幼体死亡率均随干露时间的增加而升高(P<0.05);不同温度条件下,Ⅰ期幼蟹(CⅠ)的含水量、失水率和体重消耗率均高于Ⅱ期幼蟹(CⅡ)(P<0.05);CⅡ20℃时的露空时间最长,为11h,半致死时间为7—8h;30℃时最短,为2h;CⅠ15℃时的露空时间最短,为0.5h;20℃时最长,为4h。相同温度下,幼体各发育阶段的耐干露能力为CⅡ>CⅠ>ZⅣ>ZⅢ=M。结果说明,温度≤15℃不利于幼体的存活,温度≥25℃干露耐受性降低。因此,20℃更有利于幼体获得长存活时间。  相似文献

A field study was conducted to assess the potential use of metallothionein (MT) and stress proteins as biomarkers of trace metal exposure. Shore crabs (Carcinus maenas) were collected from five sites: three located in the metal-contaminated Fal estuary (Southwest England) and two control sites located on the south coast of England. Two methods for determining MT levels in crab midgut gland were compared. A direct method in which MT was quantified by spectrophotometric measurement of SH content, and an indirect method in which the metal content (Cu and Zn) of partially purified MT was measured. Stress protein levels (stress-70) were quantified in crab gills using an improved western blotting technique. Elevated Cu and Cd concentrations compared to values in control crabs were found in the midgut glands of crabs from the Fal estuary. No significant variation was seen in Zn concentrations among the five sites. The two techniques used for MT analysis yielded similar results. Metallothionein concentrations were slightly higher in crabs from Restronguet Creek (the most metal-contaminated site) than in crabs from the other four sites, however this difference was not significant (p > 0.05). Stress protein levels did not reflect the metal gradient.  相似文献

The knowledge of the reproduction and growth background related to the shore crab Carcinus maenas promotes the use of this crab as a model crustacean to asses the potential for endocrine disruption in crustaceans. In addition, an enzyme linked inmunosorbent assay (ELISA), sensitive to the shore crab vitellogenin in serial hemolymph samples allows determination of the extent of disruption of the process of vitellogenesis in female crabs and its likely impact on reproductive output. Intermoult females Carcinus maenas were exposed to concentrations of Cd: 3 microgl(-1), Cu: 15 microgl(-1) and Zn: 700 microgl(-1) determined at the Guadalquivir estuary after the Aznalcóllar mining spill, during 21 days. Crab hemolymph samples, were taken every seven days, and analyzed through an ELISA for Carcinus maenas vitellogenin. Vitellogenin concentration along the time was fitted to a first order kinetic approach. Results showed a good correlation among experimental values and estimated ones. Metal exposure resulted in an increase in vitellogenin concentration in hemolymph, especially for cadmium.  相似文献

甲壳动物的卵巢发育及卵黄发生受到体内各种内分泌激素的调控。本文研究了两组不同饲料—脂类营养平衡组(A组,含有磷脂和HUFA)和不平衡组(B组,缺乏磷脂和HUFA)对中华绒螯蟹卵巢发育及卵黄发生的激素调控,运用放射免疫法和放射化学法分别测定了处于不同卵黄发生期的中华绒螯蟹的肝胰腺、卵巢、血淋巴中孕酮(PG)、雌二醇(E2)的含量及中华绒螯蟹大颚器(MO)合成与分泌甲基法尼酯(MF)的速率。结果表明:1)中华绒螯蟹体内三种组织(肝胰腺、卵巢和血淋巴)中均存在PG和E2,PG可能为E2的前体;2)早熟蟹的MO合成MF速率远远大于正常发育的幼蟹,MF的大量合成与分泌是促使早熟蟹产生的主要内分泌因素;3)B组饲料(含猪油)喂养的河蟹其MO合成MF的速率均高于A组饲料(含鱼油),且B组饲料的早熟蟹转化PG速度比B组饲料的早熟蟹快;4)PG和E2的脂类结合部位能结合外源性卵黄蛋白原(Vg),通过血淋巴的运输被卵泡细胞胞吞,最终与卵母细胞内的内源性Vg结合,共同形成卵黄体;5)多不饱和脂肪酸(如EPA和DHA)可能会抑制类固醇激素的产生。研究表明,MF是中华绒螯蟹的促性腺激素,具有促使PG合成和E2转化的作用,它们共同诱导河蟹卵巢发育及卵黄发生。  相似文献

为解析拟穴青蟹(Scylla paramamosain)血卵涡鞭虫病的流行病学特征,于2012年4—11月采用跟踪监测和实地调查的方法,结合病原分离鉴定及病蟹的病理学观察,统计分析了浙江省温州地区养殖拟穴青蟹血卵涡鞭虫病的流行情况。结果表明,患病拟穴青蟹的肝胰腺、肌肉、鳃等组织病理显示有大量血卵涡鞭虫侵染,并能用PCR方法快速检出。温州地区养殖拟穴青蟹血卵涡鞭虫病在每年5—11月均有发生,7—9月为集中发病时间,死亡高峰出现于8—9月之间,年份间(2010—2012)与发病率无相关性,而不同月份对病害的发生有极显著影响(P0.01)。不同发育阶段以脱壳期的发病率相对较高,发病蟹的规格主要以50g以下的幼蟹和250g以上的成蟹为主。拟穴青蟹血卵涡鞭虫病的发生与水温(r=0.756)和盐度(r=0.631)变化关系较密切,夏、秋季节受梅雨、台风影响而引起水温和盐度的剧烈变化可能是暴发血卵涡鞭虫病的诱发因素。  相似文献

采用FIASCO方法构建了中国鲎(Tachypleus tridentatus)(CA)_n微卫星富集文库,对214个阳性克隆进行测序,筛选获得60条含有微卫星的序列,其中可设计引物的序列共38条。38对微卫星引物在37个中国鲎个体中PCR检测发现,只有9个位点具有多态性,其等位基因数为5—14个,每个位点平均具有8.1个等位基因。9个位点之间不存在连锁。利用该9个具有多态性微卫星标记分析了中国沿海9个中国鲎地理群体的种群遗传多样性和遗传结构。结果表明:中国沿海的中国鲎种群遗传多态性水平仍较高, 9个地方群体间无遗传差异,它们之间未见显著分化,且有着较高的基因流;推测人为的迁移是造成遗传无分化的主要原因。我们建议中国鲎已绝迹的海域可从其他海域引入中国鲎个体来恢复地方群体资源。  相似文献

The blue crab, Callinectes sapidus, is distributed along the east coast of the United States from Cape Cod, Mass., through the Gulf of Mexico, including both relatively unpolluted coastal areas and estuaries contaminated with trace metals. Cadmium is of particular concern because it is concentrated in the digestive glands of blue crabs and can be passed on to consumer organisms. Tissue concentrations and partitioning of trace metals from crabs exposed in the laboratory to 10 ppb dissolved cadmium for 40 days were compared with blue crabs collected from two locations on the Hudson River, NY, Foundry Cove and Haverstraw Bay, Foundry Cove and Haverstraw Bay, both of which have elevated trace metal levels relative to estuarine areas near Beaufort, NC. Crab digestive glands, gills and muscle were removed and analyzed for total cadmium, copper, zinc and nickel concentrations using acid digestion and atomic absorption spectrophotometry, and metal-binding (metallothionein-like) proteins were determined by gel-filtration chromatography. In crabs exposed to cadmium in the laboratory, the cytosolic partitioning was similar to previous investigations at our laboratory where higher levels of cadmium (100 ppb) and shorter exposure times (14 days) were used. The similarity in cadmium partitioning from these two separate experiments indicates dose independence. In crabs from polluted environments the digestive glands contained the highest concentrations of trace metals. Chromatograms of the cytosol from the digestive glands and gills from both field and laboratory exposed crabs showed similar distributions of cadmium, copper and zinc. The gills of both field and laboratory-exposed crabs had metal-binding proteins that contained mostly cadmium, and the digestive glands had metallothionein-like proteins that contained cadmium, copper and zinc. Estimated molecular weights for these proteins were similar to the metallothioneins found in other crustaceans and mammals.  相似文献

The ability of benzo[a]pyrene, Aroclor 1254, 2-3-7-8-tetrachlorodibenzo-p-dioxin and beta-naphthoflavone to induce DNA strand breaks (SB) and apoptosis in erythrocytes of the European eel (Anguilla anguilla) was investigated following by in vivo exposure. DNA damage was evaluated by the Comet assay, while the diffusion assay was used to investigate the induction of apoptosis 7 days after a single intraperitoneal administration. 2-3-7-8-Tetrachlorodibenzo-p-dioxin induced the highest genotoxic effect, followed by benzo[a]pyrene, while the other two substances had limited effects. A significant induction of apoptosis was observed at the highest doses after exposure to benzo[a]pyrene, when DNA damage was also elevated. The occurrence of apoptotic cells after exposure to Aroclor, 2-3-7-8-tetrachlorodibenzo-p-dioxin and beta-naphthoflavone was quite variable and did not show clear dose-related responses. The role of oxidative stress in mediating DNA damage was also discussed.  相似文献

The effects of temperature on cellular and biochemical parameters of the crab Carcinus aestuarii were evaluated. Crabs were kept for 7 days at 4, 17 (reference value) and 30 °C (salinity of 35 psu), and total haemocyte count (THC), haemocyte volume, haemocyte proliferation, phenoloxidase (PO) activity in both haemocyte lysate (HL) and cell-free haemolymph (CFH), CFH total protein and glucose levels, superoxide dismutase (SOD) and catalase (CAT) activities in both gills and digestive gland were evaluated. The lowest and the highest temperature significantly decreased THC, whereas haemocyte volume and haemolymph glucose concentration did not differ significantly among experimental conditions. Haemolymph protein concentration significantly reduced in crabs maintained at 30 °C, when compared with that of animals kept at 4 and 17 °C. Haemocyte proliferation increased significantly in crabs kept at 4 and 30 °C, when compared with that of crabs held at 17 °C. Likewise, a significantly higher PO activity was recorded in CFH from crabs kept at 4 and 30 °C, than in control crabs. Conversely, PO activity did not vary significantly in HL. With regard to antioxidant enzyme activities, a significant decrease in CAT activity was observed in gills from crabs kept at 4 °C, when compared to that of crabs kept at 17 °C and 30 °C. Results obtained demonstrated that the highest and lowest temperature tested influenced crab biological responses, and indicated that C. aestuarii modulated its cellular and biochemical parameters (mainly haemocyte proliferation, CFH protein concentrations and CFH PO activity) in order to cope with temperature.  相似文献
