Freshwater shallow lakes typically exhibit two alternative stable states under certain nutrient loadings:macrophyte-dominated and phytoplankton-dominated water regimes.An ecosystem regime shift from macrophytes to phytoplankton blooming typically reduces the number of species of invertebrates and fishes and results in the homogenization of communities in freshwater lakes.We investigated how microbial biodiversity has responded to a shift of the ecosystem regime in Dianchi Lake,which was previously fully covered with submerged macrophytes but currently harbors both ecological states.We observed marked divergence in the diversity and community composition of bacterioplankton between the two regimes.Although species richness,estimated as the number of operational taxonomic units and phylogenetic diversity(PD),was higher in the phytoplankton dominated ecosystem after this shift,the dissimilarity of bacterioplankton community across space decreased.This decrease in beta diversity was accompanied by loss of planktonic bacteria unique to the macrophyte-dominated ecosystem.Mantel tests between bacterioplankton community distances and Euclidian distance of environmental parameters indicated that this reduced bacterial community differentiation primarily reflected the loss of environmental niches,particularly in the macrophyte regime.The loss of this small-scale heterogeneity in bacterial communities should be considered when assessing long-term biodiversity changes in response to ecosystem regime conversions in freshwater lakes.  相似文献

Harmful cyanobacterial blooms, especially Microcystis blooms, occur worldwide and draw widespread attention. The dynamics of microcystin-producing Microcystis and competition between microcystin-producing Microcystis and non-microcystin-producing Microcystis are key to predicting and treating Microcystis blooms. Multiplex qPCR is a useful tool to assess such issues. In this study, we developed multiplex qPCR methods with newly-designed probes and primers for the microcystin-synthesis related genes mcyA and mcyE. We used seven toxic Microcystis strains and four non-toxic Microcystis strains to compare the differences in the ratios of toxic and non-toxic Microcystis in mixed cultures, which were calculated using abundances of the genes mcyA, mcyB, mcyD, mcy E and phycocyanin( PC). We also compared traditional cell counting and multiplex qPCR. Hierarchical clustering and principal component analysis indicated that mcyD was the most suitable mcy gene for quantification in laboratory experiments. mcyB abundances were always higher; we suggest that the amount of toxic Microcystis measured using mcyB might overestimate the actual percentages.  相似文献

在脉冲电晕放电条件下,探讨了O2和H2O对NO分解反应过程的影响,并对复杂组分中NO的分解特性进行了实验研究。研究结果表明,脉冲电晕放电对NO分解反应具有一定的促进作用,可有效地分解模拟烟气中的NO,脱除效率随脉冲电压的增加而缓慢增加;受O2和H2O影响,NO分解转化率较N2氛围略有降低。  相似文献

In this study, molecular weight controllable degradation of algal Laminaria japonica polysaccharides(LPS) was investigated by ultrasound combined with hydrogen peroxide. Three main factors, i.e., ultrasonic power(A), ultrasonic time(B), and H_2O_2 concentration(C) were chosen for optimizing parameters by employing three-factors, three-levels BBD. The influence of degradation on structure change and antioxidant activities was also investigated. A second-order polynomial equation including molecular weight(Y) of Laminaria japonica polysaccharides and each variable parameter, i.e., ultrasonic power(A), ultrasonic time(B), and H_2O_2 concentration(C), was established: Y=20718.67-4273.13A-4000.38B-1438.75C+2333.25AB+1511.00AC+873.00BC+2838.29A~2 + 2490.79B~2+873.04C~2. The equation regression coefficient value(R~2 = 0.969) indicated that this equation was valid. The value of the adjusted determination coefficient(adjusted R~2 = 0.914) also confirmed that the model was highly significant. The results of selected experimental degradation conditions matched with the predicted value. FT-IR spectra revealed that the structures of LPS before and after degradation were not significantly changed. Antioxidant activities of LPS revealed that low Mws possessed stronger inhibitory than the original polysaccharides. The scavenging effects on superoxide radicals was the highest when IC50 of crude LPS was 4.92 mg mL~(-1) and IC50 of Mw 18.576 KDa was 1.02 mg mL~(-1), which was fourfold higher than initial polysaccharide.  相似文献

Cyanobacterial blooms in eutrophic freshwater systems are a worldwide problem, creating adverse effects for many aquatic organisms by producing toxic microcystins and deteriorating water quality. In this study, microcystins (MCs) in Microcystis aeruginosa, and Daphnia magna exposed to M. aeruginosa, were analyzed by HPLC-MS, and the effects of M. aeruginosa on D. magna were investigated. When D. magna was exposed to M. aeruginosa for more than 2 h, Microcystin-LR (MC-LR) was detected. When exposed to 1.5 × 106, 3 × 106, 0.75 × 107, and 1.5 × 107 cell/mL of M. aeruginosa for 96 h, average survival of D. magna for treatments were 23.33%, 33.33%, 13.33%, 16.67%, respectively, which were significantly lower than the average 100% survival in the control group (P < 0.05). The adverse effects of M. aeruginosa on body length, time for the first brood, brood numbers, gross fecundity, lifespan, and population growth of D. magna were density-dependent. These results suggest that the occurrence of M. aeruginosa blooms could strongly inhibit the population growth of D. magna through depression of survival, individual growth and gross fecundity. In the most serious situations, M. aeruginosa blooms could undermine the food web by eliminating filter-feeding zooplankton, which would destroy the ecological balance of aquaculture water bodies.  相似文献

In this study, the 454 pyrosequencing technology was used to analyze the DNA of the Microcystis aeruginosa symbiosis system from cyanobacterial algal blooms in Taihu Lake, China. We generated 183 228 reads with an average length of 248 bp. Running the 454 assembly algorithm over our sequences yielded 22 239 significant contigs. After excluding the M. aeruginosa sequences, we obtained 1 322 assembled contigs longer than 1 000 bp. Taxonomic analysis indicated that four kingdoms were represented in the community: Archaea (n = 9; 0.01%), Bacteria (n = 98 921; 99.6%), Eukaryota (n = 373; 3.7%), and Viruses (n = 18; 0.02%). The bacterial sequences were predominantly Alphaproteobacteria (n = 41 805; 83.3%), Betaproteobacteria (n = 5 254; 10.5%) and Gammaproteobacteria (n = 1 180; 2.4%). Gene annotations and assignment of COG (clusters of orthologous groups) functional categories indicate that a large number of the predicted genes are involved in metabolic, genetic, and environmental information processes. Our results demonstrate the extraordinary diversity of a microbial community in an ectosymbiotic system and further establish the tremendous utility of pyrosequencing.  相似文献

The response of zooplankton to the ecological environment in Daya Bay is unclear under the influence of both climate changes and anthropogenic activities on a seasonal to inter-annual scale. Based on monthly surveys and historical data, we found the zooplankton community had changed temporally and spatially. A total of 134 species was recorded during the study, and copepods dominated numerically in terms of diversity and abundance. Both copepods and cladocerans were the main contributors to zooplankton abundance. The community structure of zooplankton was temporally classified into the warm and cold groups, and spatially into the three groups located in the marine cage-culture area(MCCA), the outflow of nuclear power plants(ONPP) and unpolluted waters(UPW). The zooplankton was characterized by low biomass(dry weight), high diversity and abundance in the warm period in contrast to that in the cold period. Compared with the other two groups, the MCCA group of zooplankton showed high abundance, low diversity and biomass. Variations in dominant species were closely related to temperature, salinity and chlorophyll a concentration.Species diversity and dry weight decreased in comparison with 30 years ago, while zooplankton abundance increased. The seasonal variation in zooplankton was affected mainly by temperature that was controlled by monsoon, while the spatial difference in the community structure was probably due to eutrophication in the MCCA and thermal water discharge from ONPP. The zooplankton community is undergoing great changes with the tendency of miniaturization and gelatinization in recent 30 years in Daya Bay.  相似文献

Anthropogenic nutrient discharge poses widespread threats to coastal ecosystems and has increased environmental gradients from coast to sea. Bacterioplankton play crucial roles in coastal biogeochemical cycling, and a variety of factors af fect bacterial community diversity and structure. We used 16 S r RNA gene pyrosequencing to investigate the spatial variation in bacterial community composition(BCC) across five sites on a coast-of fshore gradient in the East China Sea. Overall, bacterial alpha-diversity did not diff er across sites, except that richness and phylogenetic diversity were lower in the of fshore sites, and the highest alpha-diversity was found in the most landward site, with Chl-abeing the main factor. BCCs generally clustered into coastal and of fshore groups. Chl-a explained 12.3% of the variation in BCCs, more than that explained by either the physicochemical(5.7%) or spatial(8.5%) variables. Nutrients(particularly nitrate and phosphate), along with phytoplankton abundance, were more important than other physicochemical factors, co-explaining 20.0% of the variation in BCCs. Additionally, a series of discriminant families(primarily affiliated with Gammaproteobacteria and Alphaproteobacteria), whose relative abundances correlated with Chl-a, DIN, and phosphate concentrations, were identified, implying their potential to indicate phytoplankton blooms and nutrient enrichment in this marine ecosystem. This study provides insight into bacterioplankton response patterns along a coast-of fshore gradient, with phytoplankton abundance increasing in the of fshore sites. Time-series sampling across multiple transects should be performed to determine the seasonal and spatial patterns in bacterial diversity and community structure along this gradient.  相似文献

Cyanobacterial blooms occur frequently in lakes due to eutrophication. Although a number of models have been proposed to forecast algal blooms, a good and applicable method is still lacking. This study explored a simple and effective mathematical-ecological model to evaluate the growth status and predict the population dynamics of Microcystis blooms. In this study, phytoplankton were collected and identified from 8 sampling sites in Chaohu Lake every month from July to October, 2010. The niche breadth and niche overlap of common species were calculated using standard equations, and the potential relative growth rates of Microcystis were calculated as a weighted-value of niche overlap. In July, the potential relative growth rate was 2.79 (a.u., arbitrary units) but then rapidly declined in the following months to -3.99 a.u. in September. A significant correlation (R=0.998, P<0.01) was found in the model between the net-increase in biomass of Microcystis in the field and the predicted values calculated by the niche model, we concluded that the niche model is suitable for forecasting the dynamics of Microcystis blooms. Redundancy analysis indicated that decreases in water temperature, dissolved oxygen and total dissolved phosphorus might be major factors underlying bloom decline. Based on the theory of community succession being caused by resource competition, the growth and decline of blooms can be predicted from a community structure. This may provide a basis for early warning and control of algal blooms.  相似文献

We describe the phytoplankton dynamics and structure in Xiangxi Bay, Three Gorges Reservoir. Samples were collected monthly in the surface waters between August 2007 and July 2008. We identified 10 principle functional groups. C-strategists and S/R-strategists with a wide range of tolerance dominated the phytoplankton assemblage. Seasonal variation was related to water column stability because of changes in hydraulic operation in October, January, and May. Functional group C (Asterionella formosa) and P (Aulacoseria granulata) dominated in August and September, whereas group Lo (Peridiniopsis niei) was the most abundant between February and April, forming a dinoflagellate bloom. Group B (Stephanodiscus hantzschii), X2 (Komma acudata), and Y (Cryptomonas erosa) were present throughout most of the year but were most abundant in late spring. A cyanobacterial bloom occurred from June to July, during which group M (Microcystis aeruginosa, M. wesenbergii) and H1 (Anabaena flos-aquae) were dominant. Green algae, characterized by group G (Eudorina sp., Pandorina sp., Pyramidomonas sp.) and J (Pediastrum spp., Coelastrum spp., Scenedesums spp.), were abundant after the bloom degraded. This sequence was corroborated by canonical correspondence analysis (CCA). The summary sequence of functional groups resulting from CCA was: C/P → Lo → H1/M/J/G. The dynamics of the phytoplankton community may be explained by the stability of water column, irradiance, water temperature, and nutrient structure.  相似文献

Although biomass variations in Microcystis and microcystin have been widely reported,few studies have addressed whether different trophic states of natural lake water affect the spatial-temporal variations in abundances of microcystin-producing Microcystis in a given bloom.In this study,we used a harmful algal bloom in Chaohu Lake,eastern China,as an example to investigate the mutual relationship between different nutrient states and environmental factors,and the impact on Microcystis.Overall,cyanobacteria and Microcystis were more abundant in the middle and western parts of the lake under high nutrients levels,while in the eastern part,nutrient concentrations were low enough to limit biomass,and their fluctuations affected the contents of toxic Microcystis.Moreover,microcystin concentration was correlated positively to nutrient levels and Microcystis biomass during bloom developing in 2013 from June to August.Temporally,the cellular content of total microcystin was lowest when the bloom peaked in intensity.Our results suggest that lake eutrophication not only results in cyanobacterial blooms,but may also increase the proportion of toxic Microcystis species and their cell-bound MCs contents(i.e.microcystin cell quotas) under mild eutrophication.The present investigation provided molecular evidence for the selection of MC-producing and non-MC-producing genotypes.The current study provides new evidence advocating the monitoring of partitions of large lakes when studying cyanobacteria and toxin-contaminated freshwaters,which will be beneficial for both water agencies and water researchers.  相似文献

Community structure changes of macrobenthos in the South Yellow Sea   总被引:3,自引:0,他引:3  
The ecological environment in the Yellow Sea has changed greatly from the 1950s to 1990s and this has had significant impact on marine organisms. In this study, data on soft-sediment macrobenthos occurring in depths from 25 m to 81 m in the South Yellow Sea were used to compare changes in community structure. The agglomerative classification (CLUSTER) and multidimensional scaling (MDS) methods were applied. Five communities were recognized by cluster analysis: 1. The Yellow Sea Cold Water Mass community dominated by cold water species, which changed slightly in species composition since the 1950s; 2. The mixed community with the coexistence of cold water species and warm water species, as had been reported previously; 3. The polychaete-dominated eurythermal community in which the composition changed considerably as some dominant species disappeared or decreased; 4. The Changjiang (Yangtze) River Estuarine community, with some typical estuarine species; 5. The community affected by the Yellow Sea Warm Current. The greatest change occurred in the coastal area, which indicated that the change may be caused by human activities. Macrobenthos in the central region remained almost unchanged, particularly the cold water species shielded by the Yellow Sea Cold Water Mass. The depth, temperature and median grain size of sediments were important factors affecting the distributions of macrobenthos in the South Yellow Sea.  相似文献

We characterized variations in bacterioplankton community composition(BCC) in mesocosms subject to three different treatments. Two groups contained fish(group one: Cyprinus carpio; group two: Hypophthalmichthys molitrix); and group three, the untreated mesocosm, was the control. Samples were taken seven times over a 49-d period, and BCC was analyzed by PCR-denaturing gradient gel electrophoresis(DGGE) and real-time quantitative PCR(q PCR). Results revealed that introduction of C. carpio and H. molitrix had a remarkable impact on the composition of bacterioplankton communities, and the BCC was significantly diff erent between each treatment. Sequencing of DGGE bands revealed that the bacterioplankton community in the different treatment groups was consistent at a taxonomic level, but differed in its abundance. H. molitrix promoted the richness of Alphaproteobacteria and Actinobacteria, while more bands affiliated to Cyanobacteria were detected in C. carpio mesocosms. The redundancy analysis(RDA) result demonstrated that the BCC was closely related to the bottom-up(total phosphorus, chlorophyll a, phytoplankton biomass) and top-down forces(biomass of copepods and cladocera) in C. carpio and control mesocosms, respectively. We found no evidence for top-down regulation of BCC by zooplankton in H. molitrix mesocosms, while grazing by protozoa(heterotrophic nanoflagellates, ciliates) became the major way to regulate BCC. Total bacterioplankton abundances were significantly higher in C. carpio mesocosms because of high nutrient concentration and suspended solids. Our study provided insights into the relationship between fish and bacterioplankton at species level, leading to a deep understanding of the function of the microbial loop and the aquatic ecosystem.  相似文献

Heparin has been used as an anticoagulant drug for many years, but it has significant side ef fects. In the search for good substitutes, low molecular weight(MW) polysaccharides from S argassum fusiforme have been examined and confi rmed to possess biological activities. Here, S. fusiforme polysaccharides(SFP) were extracted and subjected to a hydrogen peroxide(H_2O_2) oxidation method for the preparation of low-MW SFP(LSFP). The effects of temperature, pH, and H_2O_2 concentration on the degradation process were also examined. Several LSFP of 36, 9, 5.7, and 2.7 kDa were obtained under different conditions, and their anticoagulant activities studied in vitro. The results showed that SFP and LSFP prolonged activated partial thromboplastin(APTT), prothrombin(PT) and thrombin times(TT) significantly, indicating that these low MW polysaccharides possessed anticoagulant activity in the intrinsic, extrinsic, and common coagulation pathways. As these ef fects were related to the MW of the polysaccharides in APTT and TT but not in PT, the contents of the monosaccharide fucose and sulfate and the polysaccharide MW could have exerted combined ef fects. The details of this mechanism require further verifi cation.  相似文献

Jellyfish blooms have increased worldwide,and the outbreaks of jellyfish population not only affect the food web structures via voracious predation but also play an important role in the dynamics of nutrients and oxygen in planktonic food webs.However,it remains unclear whether specific carbon compounds released through j ellyfish metabolic processes have the potential to shape bacterial community composition.Therefore,in this study,we aimed to investigate the compositional succession of the bacterioplankton community in response to the dissolved organic matter(DOM)released by the live Scyphomedusae Cyanea lamarcki'i and Chrysaora hysoscella collected from Helgoland Roads of the North Sea.The bacterial community was significantly stimulated by the DOM released form live jellyfish and different dominant phylotypes were observed for these two Scyphomedusae species.Furthermore,the bacterial community structures in the different DOM sources,jellyfish-incubated media,Kabeltonne seawater,and artificial seawater(DOM-free)were significantly different,as revealed by automated ribosomal intergenic spacer analy sis fingerprints.Catalyzed reporter depo sition fluorescence in situ hybridization(CARD-FISH)revealed a rapid species-specific shift in bacterial community composition.Gammaproteobacteria dominated the community instead of the Bacteroidetes community for C.lamarckii,whereas Gammaproteobacteria and Bacteroidetes dominated the community for C.hysoscella.The significant differences in the bacterial community composition and succession indicate that the components of the DOM released by jellyfish might differ with jellyfish species.  相似文献

对珠海凡纳滨对虾淡水养殖池塘浮游植物及其理化因子进行了调查和分析,结果表明:调查中共检出浮游植物143种,优势种主要有林氏藻、螺旋鱼腥藻、小颤藻、微囊藻和点状平裂藻等,物种多样性指数在0.24~2.77之间,优势度与多样性指数呈显著的负相关,相关系数r=-0.984;而均匀度与多样性指数呈显著的正相关,相关系数r=0.967,养殖后期优势度不高,浮游植物的密度及化学因子的浓度到后期都有所上升,藻类密度的剧烈变化会引起氨氮和硫化氢含量的变化;藻类密度的波动滞后于无机氮的波动,群落演替具有突发性、时间短、速度快等特点。  相似文献
