A large-scale bloom occurred from May to June in 2011 in sea area near Qinhuangdao of the Bohai Sea, leading to huge damage of the scallop culture industry. Similar blooms have been observed in this region for three years. The causative species of the bloom, which dominated the phytoplankton community with the maximum cell density around 109 cell/L, could not be identified with morphological features due to the small cell size (~2 mm). A pigment analytical method was then adopted to analyze the pigment profile of the phytoplankton samples collected from the blooming sea area. It was found that pico-sized (<2 mm), nano-sized (2-20 mm), and bulk phytoplankton samples had similar pigment profile, representing the pigment signature of the bloom-causative species. The major pigments detected included 19-butanoyloxyfucoxanthin (But-fuco), fucoxanthin (Fuco), diadinoxanthin (Diad) and chlorophyll a (Chl a), and high content of But-fuco was the most significant characteristics of the phytoplankton samples. Based on the pigment composition and content, the bloom-causative species could be tentatively identified as pelagophyte, "type 8" group of haptophyte, or silicoflagellate. Some unique features of the bloom, such as the extremely high cell density, small-sized and But-fuco containing cells, occurring in early summer, and the feeding-cessation effects on scallops, suggest it be a "brown tide" event similar to those reported in the east coast of the United States of America. The recurrent "brown tide" events and their dramatic impacts on the shellfish mariculture industry in Qinhuangdao need close attention in the coming years.  相似文献

An inter-laboratory comparison of the AOAC mouse bioassay for paralytic shellfish poisoning (PSP) toxicity in shellfish was carried out among 25 Chinese laboratories to examine the overall performance for PSP testing in China, and to analyze the main factors affecting the performance of this method. The toxic scallop Patinopecten yessoensis collected from coast of Bohai Sea, China, was used as a test sample in the comparison study. The results were reported and evaluated using robust statistical methods. The z scores showed that 80%, 8%, and 12% of laboratories reported satisfactory results, unsatisfactory results, and questionable results, respectively. This evaluation demonstrates that the PSP mouse bioassay is an appropriate method for screening and testing PSP toxicity in shellfish. However, it was found that the experience and skill of technicians, as well as the body weight and health status of mice being used significantly affected the accuracy of the method.  相似文献

A cruise was conducted from late August to early September 2004 with the intention of obtaining an interdisciplinary understanding of the Yangtze River Estuary including the biological, chemical and physical subjects. Water sample analysis indicated that total phytoplankton species richness was 137. Of them 81 were found in Bacillariophyta and 48 in Pyrrophyta, accounting for 59.1% and 35.0% respectively. The average cell abundance of surface water samples was 8.8×104 cells L-1, with the maximum, 102.9×104 cells L-1, encountered in the area (31.75°N, 122.33°E) and the minimum, 0.2×104 cells L-1, in (30.75°N, 122.17°E). The dominant species at most stations were Skeletonema costatum and Proboscia alata f. gracillima with the dominance of 0.35 and 0.27. Vertical distribution analysis indicated that obvious stratification of cell abundance and dominant species was found in the representative stations of 5, 18 and 33. Shannon-Wiener index and evenness of phytoplankton assemblage presented negative correlation with the cell abundance, with the optimum appearing in (30.75°N, 122.67°E). According to the PCA analysis of the environmental variables, elevated nutrients of nitrate, silicate and phosphate through river discharge were mainly responsible for the phytoplankton bloom in this area.  相似文献

Eutrophication has become a serious concern in many lakes, resulting in cyanobacterial blooms. However, the mechanism and pathways of cyanobacteria decline are less understood. To identify and define the growth and decline of Microcystis blooms in Taihu Lake of China, and to illuminate the destination of surface floating blooms, we investigated the biomass distribution and variations in colony size, morphology, and floating velocity from October 2008 to September 2009. The results showed that the Microcystis bloom declined in response to biomass decrease, colony disaggregation, buoyancy reduction, and increased phytoplankton biodiversity, and these indicative parameters could be applied for recognition of the development phases of the bloom. Three major decline pathways were proposed to describe the bloom decline process, colony disaggregation (Pathway Ⅰ), colony settlement (Pathway Ⅱ), and cell lysis in colonies (Pathway Ⅲ). We proposed a strategy to define the occurrence and decline of Microcystis blooms, to evaluate the survival state under different stress conditions, and to indicate the efficiency of controlling countermeasures against algal blooms.  相似文献

Sulfotransferase (ST) is the first enzyme discovered in association with paralytic shellfish poisoning (PSP) toxin biosynthesis in toxic dinoflagellates. This study investigates the ST activity in crude enzyme extraction of a toxic dinoflagellate species, Alexandrium tamarense CI01. The results show that crude enzyme can transfer a sulfate group from 3’-phosphoadenosine 5’-phosphosulfate (PAPS) to N-21 in the carbamoyl group of gonyautoxin 2/3 (GTX2/3) to produce C1/C2, but is inactive toward STX to produce GTX5. The crude enzyme is optimally active at pH 6.0 and 15°C. The activity is enhanced by Co2 , Mg2 , Mn2 and Ca2 individually, but is inhibited by Cu2 . Moreover, the activity shows no difference when various sulfur compounds are used as sulfate donors. These results demonstrate that the ST specific to GTX2/3 is present in the cells of A. tamarense CI01 and is involved in PSP toxin biosynthesis. In addition, the ST from different dinoflagellates is species-specific, which explains well the various biosynthesis pathways of the PSP toxins in toxic dinoflagellates.  相似文献

Phytoplankton abundance was found to be positively correlated with seasonal changes of seawater temperature in Port Shelter and Lamma Channel, Hong Kong in 1998. Rising water temperature from around 20°C to 25°C coincided with an increase in phytoplankton abundance at both locations. Heavy rains from June to September reduced salinity from 30 to 20, but the decrease in salinity was not correlated with a decline in phytoplankton abundance. The dinoflagellate Dinophysis acuminata Claparede & Lachmann is a toxic alga that causes diarrhetic shellfish poisoning. No Dinophysis species were maintained in culture for a long period of time until 2006 when Park successfully established D. acuminata in culture using a three-step feeding protocol in which the cryptophyte, Geminigera cryophila, is fed to Myrionecta rubra (=Mesodinium rubrum), a ciliate that is in turn fed to D. acuminata. In this paper, we present the details of culturing D. acuminata from the Northeastern United States. The protocols described herein can be adopted for laboratory studies of this species. The effects of temperature on the growth and ingestion rates of D. acuminata were also examined. The results show that D. acuminata growth rate was 0.23 /d at 10°C and 0.11 /d at 4°C when fed M. rubra prey. The maximum prey ingestion rate was 2.80 Dinophysis cell/d at 10°C, although the rate decreased slightly at 4°C. In overall, temperature showed a greater influence on growth rate of D. acuminata than on the ingestion rate under the study conditions, and the quantity of available food was also an important regulator to D. acuminata growth.  相似文献

In recent years, seasonal blooms of the dinoflagellate Noctiluca miliaris have appeared in the open-waters of the northern Arabian Sea (NAS). This study provides the first characterization of bacteria from a seasonal bloom of green Noctiluca of NAS (20°N?17°N and 64°E–70°E), during the spring-inter-monsoon cruise of Sagar Sampada 253, in March 2007. Bacterial growth as assessed by most-probable number (MPN) and plate counts, revealed ‘variable-physiotypes’ over a wide range of salinities (0%–25% w/v NaCl), pH levels (5–8.5), and organic nutrient strengths, in comparison to non-bloom waters. MPN indices of bacteria in surface waters of bloom stations *DWK and *PRB, corresponded to (3.08–4.41)×103 cells/mL at 3.5% NaCl (w/v), and (2.82–9.49)×102 cells/mL at 25% (w/v) NaCl in tryptone-yeast extract broth (TYE). Plate counts were (1.12–4)×106 CFU/mL at 0% (w/v) NaCl, (1.28–3.9)×106 CFU/mL at 3.5% (w/v) NaCl, and (0.4–7)×104 CFU/mL at 25% NaCl (w/v) on TYE. One-tenth-strength Zobell’s gave (0.6–3.74)×105 CFU/mL at pH 5 to (3.58–7.5)×105 CFU/mL at pH 8.5. These bacteria were identified to the genera Bacillus, Cellulomonas, Staphylococcus, Planococcus, Dietzia, Virgibacillus, Micrococcus, Sporosarcinae, Leucobacter, and Halomonas. The identity of three strains (GUFBSS253N2, GUFBSS253N30, and GUFBSS253N84) was confirmed through 16S rDNA sequence homology as Bacillus cohnii, Bacillus flexus, and Bacillus cereus. The ~2–3-fold higher plate counts of culturable bacteria from the open-waters of the NAS indicate that these bacteria could critically determine the biogeochemical dynamics of the bloom and its milieu. The role of these bacteria in sustaining/terminating the bloom is under evaluation.  相似文献

The 5.8S ribosomal DNA sequences (5.8S rDNA) and their flanking regions, internal transcribed spacer 1 and spacer 2 (ITS1 and ITS2) of three new isolates in genus Alexandrium (Alexandrium sp. qd1, Alexandrium sp. qd2, Alexandrium sp. gz) from China were amplified, sequenced, and subjected to phylogenetic analysis. Alexandrium sp. gz and Alexandrium sp. qd1 were grouped with high bootstrap values with four strains/species, i.e., A. catenella South Korea strain, A. catenella Japan strain, and two from China, Alexandrium sp. AC03 and Alexandrium sp. AN01 being proposed to be A. catenalla in a previous study. Then Alexandrium sp. gz and Alexandrium sp. qd1 were identified as Alexandrium catenella. As A. catenella was isolated from Qingdao and Guangzhou sea areas, it supposedly distributed at least in these two areas and was genetically different. Alexandrium sp. qd2 differed greatly from species in Alexandrium. It clustered with Symbiodinium californium, Symbiodinium sp. G15 and Gymnodinium sp. Zhao 01 with 100% bootstrap value; so Alexandrium sp. qd2 affiliates to genus Symbiodinium, and is probably a free-living Symbiodinium species.  相似文献

To explore the impact of environmental variables on macroalgal communities, their temporal and spatial distributions were examined along the Yantai coast, China between April 2010 and March 2011. Macroalgae sampling was conducted monthly at four sites along the coast: Jiahe River estuary, Zhifu Island, Fisherman's Wharf, and Yangma Island. The species composition and abundance, and their relationship with environmental variables were assessed. Along the Yantai coast, 35 macroalgae species were identified, including 24 Rhodophyta spp., 7 Chlorophyta, and 4 Phaeophyta spp. Highest species numbers were recorded in the summer at all sampling sites, except in the Jiahe River estuary. Macroalgae biomass was the greatest in the summer. Year-round, the highest species number and dry biomass recorded at Fisherman's Wharf and Yangma Island was attributed to the substrate type. In summer, Ulva pertusa Kjellman was the dominant species identified along the Yantai coast, which indicates a risk ofmacroalgae blooms. Our results show that seawater temperature and nutrients appear to significantly affect the temporal and spatial patterns of macroalgal abundance along the Yantai coast. The effects of environmental variables on the macroalgae on the Yantai coast need further study.  相似文献

The finfish and shellfish resources were assessed quantitatively and qualitatively in regard to their abundance in creek waters at three sites within a period of two years, from January 1999 to December 2000, in the western mangrove areas of Kachchh. The catch rate varied from 0.69 to 6.99 kg h−1. It was low during monsoon (July to October), which could be due to the freshwater-flow-induced salinity reduction in all the sites. Among 38 species recorded, 5 were shellfish and 33 were finfish. The spawning period of fishes was found to be during summer and early monsoon period (May to August). Surface water temperatures varied from 17 °C to 37 °C. Salinity values varied from 34 to 44 and the pH ranged between 7 and 8.9. Variation in dissolved oxygen content was from 3.42 to 5.85 mL L−1. The high fishery densities in these semi arid mangrove creek areas were recorded during monsoon and early winter season.  相似文献

The bloom forming dinoflagellate, Ceratium furca, has been linked with coastal eutrophication worldwide in tropical and subtropical locations. During the summer of 2007, an unusual 6-month long bloom of C. furca was observed in Pago Pago Harbor, Tutuila Island, American Samoa. Incidents of dinoflagellate blooms in this area have not been previously reported. The bloom was first reported in May and dissipated in November 2007. In February?CMarch 2009, a similar C. furca bloom was observed. During both blooms, no fish mortality events were reported. Maximum cell counts were observed on September 20, 2007 at 9 200 cell/mL. At this time, total nitrogen was measured at 1.2 mg/L while total phosphate was below detection limits. Changes in land use practices may have been the primary driver of these blooms. Intense fertilization of athletic fields coupled with ineffective management strategies is hypothesized to have a direct link to the increase in nutrients found in the Pago Pago Harbor and may have been the trigger for the initialization of these blooms. During 2008, the fields were not used due to an infestation of the fire ant, Solenopsis geminata. Once controlled, the fields were opened again in 2009 and fertilizers were applied in January, a month before the bloom was observed.  相似文献

From 1997 to 2000, four field surveys were conducted in the East China Sea (ECS) (23°30′–33°00′N, 118°30′–128°00′E). A field data yield density model was used to determine the optimal salinities for 19 dominant copepod species to establish the relationship between surface salinities and abundance of those species. In addition, ecological groups of the copepods were classified based on optimal salinity and geographical distribution. The results indicate that the yield density model is suitable for determining the relationship between salinity and abundance. Cosmocalanus darwini, Euchaeta rimana, Pleuromamma gracilis, Rhincalanus cornutus, Scolecithrix danae and Pareucalanus attenuatus were determined as oceanic species, with optimal salinities of >34.0. They were stenohaline and mainly distributed in waters influenced by the Kuroshio or Taiwan warm current. Temora discaudata, T. stylifera and Canthocalanus pauper were nearshore species with optimal salinities of <33.0 and most abundant in coastal waters. The remaining 10 species, including Undinula vulgaris and Subeucalanus subcrassus, were offshore species, with optimal salinity ranging from 33.0–34.0. They were widely distributed in nearshore, offshore and oceanic waters but mainly in the mixed water of the ECS.  相似文献

A pool of dormant stages of planktonic organisms in saline lakes is a substantial component in the plankton communities;we need to take it into account to understand plankton dynamics.Hypersaline water bodies in Crimea,the largest peninsula in the Black Sea,constitute a very characteristic and peculiar habitat type in the region.We examined the presence of crustacean resting stages in sediments of dried up sites of the Crimean hypersaline lakes.Sediment samples were taken in 9 different lakes.Experiments performed on the hatching of these resting stages showed the presence of Moina salina(Cladocera),parthenogenetic Artemia and Artemia urmiana(Anostraca),Eucypris mareotica( inflata)(Ostracoda),and Cletocamptus retrogressus(Harpacticoida).Comparing the experimental results obtained with clean dried brine shrimp cysts and those kept in sediment samples,it was noted that clean cysts hatched much faster than those from sediments did.Some components in bottom sediments slow down and desynchronize hatching from resting eggs in different groups of crustaceans.The sediments of different lakes inhibited the nauplii output from Artemia and ostracod resting eggs to different degrees.More data are needed before we can discuss the reasons of this inhibition.The nonsynchronous output of active stages from the bottom resting ones may be an adaptation that allows crustacean species to exist in extreme and unpredictably changing environments,avoiding the risk that all may emerge at once under unsuitable conditions.  相似文献

The dinoflagellate Alexandrium minutum is often associated with harmful algal blooms (HABs). This species consists of many strains that differ in their ability to produce toxins but have similar morphology, making identification difficult. In this study, species-specific rRNA probes were designed for whole-cell fluorescence in situ hybridization (FISH) to distinguish A. minutum from two phylogenetic clades. We acquired the complete SSU to LSU rDNA sequences (GenBank accession numbers JF906989-JF906999) of 11 Alexandrium strains and used these to design rRNA targeted oligonucleotide probes. Three ribotype-specific probes, M-GC-1, M-PC-2, and M-PC-3, were designed. The former is specific for the GC clade ("Global clade") of A. minutum, the majority of which have been found non-toxic, and the latter two are specific for the PSP (paralytic shellfish poisoning)-producing PC clade ("Pacific clade"). The specificity of these three probes was confirmed by FISH. All cells in observed fields of view were fluorescently labeled when probes and target species were incubated under optimized FISH conditions. However, the accessibility of rRNA molecules in ribosomes varied among the probe binding positions. Thus, there was variation in the distribution of positive signals in labeled cells within nucleolus and cytosol (M-GC-1, M-PC-3), or just nucleolus (M-PC-2). Our results provide a methodological basis for studying the biogeography and population dynamics of A. minutum, and providing an early warning of toxic HABs.  相似文献

Green tides are caused by the proliferation of chlorophytes under suitable hydrographic conditions.These blooms lead to environmental degradation and negatively impact the waters and seagrass beds,as well as fishing and other recreational activities in the bay.A comprehensive ecological understanding of the bloom dynamics,including the origin and persistence,is needed to foster management decisions.The algae in the great majority of green tide blooms usually belong to two genera of Ulvophyceae,Ulva and Enteromorpha.Ulva has been observed more often in recent years.In China,green tides occurred for the first time in the middle area of the Yellow Sea in 2007,and a large-scale algae blooming broke out in the middle and southern areas of the Yellow Sea in late May 2008.We identified them as Ulva prolifera by comparative analysis of the rDNA internal transcribed spacer 1 (ITS1),5.8S and ITS2 sequences in combination with microscopic observation.Morphological differences were found between the free-floating algae and the attached thalli.Various reproduction patterns of the free-floating algae include sexual,asexual and vegetative propagations,which played important roles in the long-term green tide persistence in China.The ITS sequences of the blooming algae were identical to those of the samples from the Lianyungang sea area but were different from the attached samples from the Qingdao sea area.The results infer that the blooms are originated from other sea areas rather than from the local attached populations.  相似文献

A bloom of the dinoflagellate Scrippsiella trochoidea was detected for the first time in inner Tolo Harbor, Hong Kong in 2 000. Water samples were collected at eight stations along a transect passing through a red tide patch for microscopic analysis of phytoplankton composition and high-performance liquid chromatography (HPLC) analysis of phytoplankton pigments. During the bloom, the density of dinoflagellates was 1.1×106 cells L−1 within the patch and 8.6×105 cells L−1 outside the patch where the phytoplankton community was dominated by diatoms. After the bloom the S. trochoidea began to decrease in density and was replaced by diatoms as the dominating bloom-causing organisms at all stations, and the density of dinoflagellates at most stations was less than 1.0 × 106 cells L−1. The status of S. trochoidea as the causative species of the bloom was indicated by the presence of peridinin, the marker pigment for dinoflagellates. The shift from dinoflagellates to diatoms was marked by the decline of peridinin and the prevalence of fucoxanthin. Phytoplankton pigment markers also revealed the presence of other minor phytoplankton assemblages such as cryptomonads and blue-green algal.  相似文献
