以小杂鱼、低值鱼为原料,采用溶剂萃取法研究了溶剂种类温度、时间和溶剂量对鱼蛋白提取过程中脱水、脱脂、脱色和脱腥的影响,筛选出提取浓缩鱼蛋白最佳工艺条件。结果表明:在该工艺条件下制得的浓缩鱼蛋白提取其蛋白质含量达92.28%,脂肪含量仅为0.092%。  相似文献

研究了木瓜蛋白酶的酶解温度、酶浓度、酶解时间、加水量、pH值对青鳞鱼可溶性蛋白得率的影响 ,探讨了α 氨态氮含量与青鳞鱼可溶性蛋白得率的关系。结果表明 :温度小于 5 0℃ ,随着酶解温度的提高、酶浓度的增加、酶解时间的延长青鳞鱼可溶性蛋白得率及α 氨态氮的含量均增加 ,α 氨态氮含量增加比可溶性蛋白得率的增加明显 ;加水量的增加 ,可溶性蛋白得率增加 ;pH值对水解效果及可溶性蛋白得率影响不大。综合考虑 ,最佳水解条件为 :温度 5 0℃ ,酶浓度为 1 5 0 0u/g ,酶解时间 80min ,m原料 :m水 =1∶2 ,pH 6 8。酶解所得可溶性蛋白经氨基酸分析仪检测 ,其氨基酸总量为 49 47mg/mL ,必需氨基酸占氨基酸总量的 41 0 % ,游离氨基酸含量为 6 3mg/mL ,肽含量为 87 2 %。  相似文献

【目的】研究无定型碳酸钙结合蛋白(ACCBP)基因的上游调控机制。【方法】以合浦珠母贝(Pinctada fucata)为研究对象,利用基因组步移技术克隆合浦珠母贝ACCBP基因(PfACCBP)的启动子。设计启动子截短实验,通过双荧光素酶标记法确定启动子与转录因子CREB的结合位点,定位启动子中的功能元件。克隆转录因子环腺苷效应元件结合蛋白(CREB),并通过共转染实验和凝胶迁移率分析检测启动子的相对转录活性。【结果】获得合浦珠母贝ACCBP基因的启动子序列,ACCBP启动子在HEK-293T细胞系中转录活性较高;该启动子上存在CRE元件,该元件可在转录过程中与CREB蛋白直接结合,大幅上调ACCBP启动子的转录效率。【结论】转录因子CREB可通过与ACCBP启动子上的CRE元件结合而增强转录活性的调控作用。  相似文献

Preparation of Fe^2+ chelate of fish protein hydrolysate (Fe-FPH) obtained from low value fish proteins was introduced and its bioactivity was studied by compound enzymolysis. The optimum conditions for hydrolysate chelating Fe^2+ are DH (degree of hydrolysis) at 5%, pH 7.0, 20℃ and 15 min chelating time for FM (material not being defatted). Four types of Fe-FPH including CA (deposit after chelating), CB (deposit in 50% of absolute ethanol solution), CC (suspended deposit in 80% of absolute ethanol solution), and CD (bottom deposit in 80% of absolute ethanol solution) were fractionated with absolute ethanol from FM. Structural analysis through infra-red spectrum revealed that Fe^2+ was combined strongly with amino-group and carboxyl-group in each chelate and each Fe^2+ could form two five-member ring structures. All of the four chelates were shown more significant antioxidative activity and can be used as natural hydrophobic and hydrophilic antioxidant. Among all the chelates, the CB possesses the most effective antioxidative activity at 92% as high as that of a-tocopherol. Among all Fe-FPHs, only CD showed the most effective antibacterial activity against Escherichia coli, Staphylococcus aureus, Salmonella typhi, and Bacillus subtilis and can be used as natural antibacterial. It provides a more effective way for utilization of low value fish proteins and key information of Fe-FPH as additive in food industry.  相似文献

【目的】探讨蛋白核小球藻采收后培养液是否能循环再用于小球藻的培养。【方法】采用小球藻分批培养完成采收细胞后所得培养废液来配制培养基,重新进入下一批次小球藻培养。以藻类BG11培养基配方为基准,按照配方全部或部分补充营养盐,通过测定藻液的光密度值间接反映藻生物量,比较小球藻在循环培养中的生长情况。【结果】在连续循环利用回收水至第4批次培养收获时培养液的光密度D(600 nm)为9.9,比生长速率为0.80 d-1,与第1批培养收获时培养液的光密度D(600 nm)和比生长速率相比,小球藻细胞产量和生长速率均未受影响。经过4个轮次的循环培养,用水量可减少约75%。在连续循环培养中仅补充N、P、S盐,至第3个循环小球藻收获时的生物量变化在10%以内。【结论】在加糖混养、保持连续光照、按照BG11配方补全营养盐的培养条件下,至少可对蛋白核小球藻进行4个周期的循环培养。按照BG11配方量补充相应的N、P、S盐,减少其他营养盐的投放,在2个循环周期内可以维持蛋白核小球藻的正常生长。利用培养废液实现小球藻循环经济培养和减少生产排放方法可行。  相似文献

Phycoerythrin and phycocyanin were purified from Porphyra yezoensis Ueda with their bioactivity determined in this study. Continuous precipitation with ammonium sulfate at different concentrations (10%, 20%, 40%and 50%) increased the purity (A564:A280) of phycoerythrin to 1.49, 3.92 fold of the raw extract (0.38) and the purity (A615:A280) of phycocyanin to 0.70, 3.33 fold of the raw extract (0.21). Two more times of chromatography with hydroxylapatites finally made the purity of phycoerythrin and phy-cocyanin reach 5.50, 14.47 fold of the raw extract, and 5.10, 24.29 fold of the raw extract, respectviely. The yield of high purity phycoerythrin and phycocyanin were 0.21%and 0.09%of dried P. yezoensis blade, respectively. The photodynamic cytotoxic ex-periment showed that both phycoerythrin and phycocyanin inhibited the growth of liver tumor cells significantly. It was found that 250 mg L-1 purified phycoerythrin and phycocyanin inhibited the growth of hepatocellular carcinoma cells 24 h after laser-irradiation by 80%and 59%, respectively, and 100 mg L-1 purified phycoerythrin and phycocyanin induced the apoptosis of 31.54%and 32.54%of the cells, respectively, 8 h after photodynamic therapy. Oue findings demonstrated that P. yezoensis can serve as photosensitizer (phycoerythrin and phycocyanin) producer.  相似文献

Arthrospira platensis is an economically important cyanobacterium; and it has been used widely in food and pharmaceutical industries. The phycocyanin(PC) from A. platensis is extremely valuable in medicine and molecular biology due to its antioxidation and anti-tumoring activity and applicability as fluorescence protein tag. In present study, two recombinant plasmids, one contained the phycocyanobilin(PCB)-producing genes(hox1 and pcyA) while the other contained the phycobiliprotein gene(cpcB) and the lyase gene(either cpcS/U or cpcT), were constructed and synchronically transferred into E. coli in order to test the the activities of relevant lyases for catalysing PCB addition to CpcB during synthesizing fluorescent PC holo-β-subunit(β-PC) of A. platensis. As was evidenced by the fluorescence emitted at a peak specific for PC, CpcB was successfully synthesized in E. coli, to which co-expressed PCBs attached though at a relatively low efficiency. The results showed that the attachment of PCBs to CpcB were carried out mainly by co-expressed CpcS/U but CpcB also showed some autocatalytic activity. Currently, no CpcT activity was detected in this E. coli expression system. Further studies will be conducted to improve the efficiency of fluorescent PC synthesis in E. coli.  相似文献

研究了木瓜蛋白酶的酶解温度,酶浓度,酶解时间,加水量,pH值对青鳞鱼可溶性蛋白得率的影响,探讨了α-氨态氮含量与青鳞鱼与可溶性蛋白得率的关系。结果表明:温度小于50℃,随着酶解温度的提高,酶浓度的增加,酶解时间的延长青鳞鱼可溶性蛋白得率及α-氨态氮的含量均增加,α=氨态氮含量增加比可溶性蛋白得率的增加明显;加水量的增加,可溶性蛋白得率增加;pH值对水解效果及可溶性蛋白得率影响不大。综合考虑,最佳水解条件为:温度50℃,酶浓度为1500u/g,酶解时间80min,m原料:m水=1:2,pH6.8。酶解所得可溶性蛋白经氨基酸分析仪检测,其氨基酸总量为49.47mg/mL,必需氨基酸占氨基酸总量的41.0%,游离氨基酸含量为6.3mg/mL,肽含量为87.2%。  相似文献

An open reading frame (lcn61) of iymphocystis disease virus China (LCDV-cn), probably responsible for encoding putative zinc-finger proteins was amplified and inserted into pET24a (+) vector.Then it expressed in E. coli BL21 (DE3), and His-tag fusion protein of high yield was obtained. It was found that the fusion protein existed in E. coli mainly as inclusion bodies. The bioinformatics analysis indicates that LCN61 is C2H2 type zinc-finger protein containing four C2H2 zinc-finger motifs. This work provides a theory for functional research of lcn61 gene.  相似文献

To improve the expression efficiency of exogenous genes in Chlamydomonas reinhardtii, a high efficient expression vector was constructed. Green fluorescent protein (GFP) was expressed in C. reinhardtii under the control of promoters: RBCS2 and HSP70A-RBCS2. Efficiency of transformation and expression were compared between two transgenic algae: RBCS2 mediated strain Tran-Ⅰ and HSP70A-RBCS2 mediated strain Tran-Ⅱ. Results show that HSP70A-RBCS2 could improve greatly the transformation efficiency by approximately eightfold of RBCS2, and the expression efficiency of GFP in Tran-Ⅱ was at least double of that in Tran-Ⅰ. In addition, a threefold increase of GFP in Tran-Ⅱ was induced by heat shock at 40℃. All of the results demonstrated that HSP70A-RBCS2 was more efficient than RBCS2 in expressing exogenous gene in C. reinhardtii.  相似文献

To investigate the antitumor effect of bromophenol derivatives in vitro and Leathesia nana extract in vivo, six bromophenol derivatives 6-(2,3-dibromo-4,5-dihydroxybenzyl)-2,3-dibromo-4,5-dihydroxy benzyl methyl ether (1), (+)-3-(2,3-dibromo-4,5-dihydroxyphenyl)-4-bromo-5,6-dihydroxy-1,3-dihydroisobenzofuran (2), 3-bromo-4-(2,3-dibromo-4,5-dihydroxybenzyl)-5-methoxymethyl-pyrocatechol (3), 2,2′,3,3′-tetrabromo-4,4′,5,5′-tetrahydroxy-diphenylmethane (4), bis(2,3-dibromo-4,5-dihydroxybenzyl) ether (5), 2,2′,3-tribromo-3′,4,4′,5-tetrahydroxy-6′-ethyloxymethyldiphenylmethane (6) were isolated from brown alga Leathesia nana, and their cytotoxicity were tested by MTT assays in human cancer cell lines A549, BGC-823, MCF-7, B16-BL6, HT-1080, A2780, Bel7402 and HCT-8. Their inhibitory activity against protein tyrosine kinase (PTK) with over-expression of c-kit was analyzed also by ELISA. The antitumor activity of ethanolic extraction of Leathesia nana (EELN) was evaluated on S180-bearing mice. All compounds showed very potent cytotoxicity against all of the eight cancer cell lines with IC50 below 10 μg/mL. In PTK inhibition study, all bromophenol derivatives showed moderate inhibitory activity and compounds 2, 5 and 6 showed significant bioactivity with the inhibition ratio of 77.5%, 80.1% and 71.4%, respectively. Pharmacological studies reveal that EELN could inhibit the growth of Sarcoma 180 tumor and increase the indices of thymus and spleen to improve the immune system remarkably in vivo. Results indicated that the bromophenol derivatives and EELN can be used as potent antitumor agents for PTK over-expression of c-kit and considered in a new therapeutic strategy for treatment of cancer. In aquatic ecosystems, macrophytes and phytoplankton are main primary producers, in which macrophyte plays an important role in maintaining clear water state, while phytoplankton often dominates in turbid waterbodies. In the present study, the growth and photosynthetic activity of the submerged aquatic plant Ceratophyllum oryzetorum Kom. in different cell densities of cyanobacterial bloom are studied. The results show that the plant length and fresh mass of C. oryzetorum are promoted by low cyanobacterial cell densities. Medium and high cyanobacterial cell densities, on the contrary, act as inhibitory. Furthermore, the photosynthetic activity of C. oryzetorum is strongly inhibited by high cyanobacterial cell densities. To a certain extent, the growth of cyanobacteria is inhibited by C. oryzetorum, but no significant effect is found in this study. Six steroids have been isolated from ethanolic extract of green alga Chaetomorpha basiretorsa Setchell by a combination of repeated normal phase silica gel and Sephadex LH-20 gel column chromatography as well as recrystallization. Using spectroscopic methods including MS and NMR, their structures were determined as β-1awsaritol (1), saringosterol (2), 24-hydroperoxy-24-vinyl- cholesterol (3), β-stigmasterol (4), stigmast-4-en-3α, 6β-diol (5), 29-hydroxystigmasta-5, 24 (28)-dien-3β-ol (6). All these compounds were obtained from this genus for the first time and they were inactive (IC50〉10μg/ml) against KB, Bel-7402, PC-3M, Ketr 3 and MCF-7 cell lines.  相似文献

Non-specific immune response of bullfrog Rana catesbeiana to pathogenic Aeromonas hydrophila was studied to 60 individuals in two groups. Each bullfrog in bacterium-injected group was injected intraperitoneally (i.p.) with 0.2 ml bacterial suspension at a density of 5.2 × 10^6 CFU/ml, while each one in control group injected i.p. with 0.2 ml sterile saline solution (0.85%, w/v). Three bullfrogs in both groups were sampled at 0, 1, 3, 7, 11, 15 and 20 days post-injection (dpi) for the evaluation of non-specific immune parameters. It was observed that intraperitoneal injection of A. hydrophila significantly increased the number of leucocytes and that of NBT-positive cells in peripheral blood. Significant increases in serum bactericidal activity and serum acid phosphatase activity were also observed in the bacterium-injected frogs when compared with those in the control group. However, a significant reduction was detected in vitro in phagocytosis activity of peripheral blood phagocytes. No significant difference in changes in the number of peripheral erythrocytes, serum superoxide dismutase (SOD) activity, and lysozyme activity was detected between the two groups. It is suggested that bullfrogs may produce a series of non-specific immune reactions in response to the A. hydrophila infection.  相似文献

A full length amphioxus cDNA, encoding a novel phosducin-like protein (Amphi-PhLP), was identified for the first time from the gut cDNA library of Branchiostoma belcheri. It is comprised of 1 550 bp and an open reading frame (ORF) of 241 amino acids, with a predicted molecular mass of approximately 28 kDa. In situ hybridization histochemistry revealed a tissue-specific expression pattern of Amphi-PhLP with the high levels in the ovary, and at a lower level in the hind gut and testis, hepatic caecum, gill, endostyle, and epipharyngeal groove, while it was absent in the muscle, neural tube and notochord. In the Chinese Hamster Ovary (CHO) cells transfected with the expression plasmid pEGFP-N1/Amphi-PhLP, the fusion protein was targeted in the cytoplasm of CHO cells, suggesting that Amphi-PhLP is a cytosolic protein. This work may provide a framework for further understanding of the physiological function of Amphi-PhLP in B. belcheri. In the present study, genetic polymorphism and diversity in unicellular clones of Chlorella vulgaris Beijerinck and Chlorella pyrenoidosa Chick were studied with Inter Simple Sequence Repeats PCR (ISSR PCR). Samples including four clones of C. vulgaris and three clones of C. pyrenoidosa were purified by single-clone-choice method. For four C. vulgaris unicellular clones, the total number of the bands scored for 18 primers was 298; and the number of the polymorphic bands was 118, of which 39.6% were polymorphic. The size of PCR products ranged from 200 to 2 500 bp. The total number of bands scored for 18 primers, the number of polymorphic bands and the percentage of three C. pyrenoidosa unicellular clones was 194.83 and 30.8%, respectively. POPGENE analysis show that the average Nei genetic diversity (h^*) and Shannon index of diversity (I^*) in the four C. vulgaris unicellular clones was 0.2181 and 0.3208, respectively, which is slightly higher than those of the three C. pyrenoidosa unicellular clones (0.190 3 and 0.274 8), which agreed with the percentage of polymorphic bands in the mixed samples of the two species. The results suggest that ISSR is a useful method to Chlorella for intra-species genetic analysis.  相似文献

For the investigation of anti-infection immune response of Pseudosciaena crocea, 160 healthy fish samples were categorized into infected and control groups. Each individual fish in the infected group was injected intraperitoneally (i.p.) with 0.2 ml bacterial suspension of Vibrio alginolyticus in density of 2×107 CFU/ml, while each individual in the control group was injected i.p. with 0.2 ml sterile saline solution (0.85%). It was observed that the artificial injection of V. alginolyticus significantly increased the number of erythrocytes, leucocytes, lymphocytes in peripheral blood as well as peripheral serum antibacterial activity and antibody titer of large yellow croaker, and significantly reduced the number of peripheral blood granulocytes as compared with those in the control group. No significant difference in acid phosphytase and superoxide dismutase activity of serum was detected between the two groups. It is suggested that non-specific immune factors including leucocytes and anti-bacteria substance in peripheral blood played important role at the initial stage of infection, and specific immune factors such as antibody then played important role in response to anti-infection at the latter stage. The hybrid between olive flounder Paralichthys olivaceus and stone flounder Kareius bicoloratus was produced by artificial insemination of olive flounder eggs with stone flounder sperm. Sinistral and dextral are two types of hybrid progeny after metamorphosis. Karyotypes of both hybrid flounders are the same as those of the two parental species. Of the 22 loci examined from 12 allozymes, 12 confirmed hybridization of the paternal and maternal loci in hybrids and no difference was found in allozyme patterns of sinistral and dextral hybrid fishes. RAPD patterns of these specimens were also studied with 38 primers selected from 104 tested. Among them, the PCR products of 30 primers showed hybridization of the paternal and maternal bands. Genetic variation between hybrids and their parental stocks was analyzed by RAPD using 10 of the above 38 primers. The average heterozygosity and genetic distance were calculated. The results suggested that the filial generation could inherit a little more genetic materials from paternal fish than that from maternal fish.
