五株微囊藻毒素降解菌的分离与鉴定   总被引:9,自引:0,他引:9  
有毒蓝藻产生的蓝藻毒素导致地表水的污染已是全球普遍的现象,最常见的毒素是能导致肝癌的肝毒素微囊藻毒素(MC),迄今为止尚无适用的MC驱除技术,微生物降解MC可能是一条有效的途径.本研究以铜绿微囊藻(Microcystis aeruginosa PCC 7806)所产生的MC为底物,从多个形成微囊藻水华的水体中分离获得能显著降解MC的细菌18株,相对降解能力从18.4%到48.2%.对其中5株MC相对降解能力较强的菌,通过形态特征和生理生化特征分析,鉴定出5株菌分别为:不动细菌属(Acinetobacter)、肠杆菌属(Enterobacter)、微杆菌属(Microbacterium)、芽孢杆菌属(Bacillus)和弗拉特氏菌属(Frateuria),均不同于已报道的属种.  相似文献

蓝藻水华及其释放的蓝藻毒素给我国很多地区的饮用水安全带来较大威胁.尽管去除蓝藻和净化藻毒素的技术目前已有较多报道,但能同时对二者起作用的技术方法还较少,发展能够同时去除蓝藻和蓝藻毒素的技术显得十分必要.根据前期Fe(Ⅲ)化合物去除微囊藻毒素的工作基础,选取4种不同的土壤对其进行Fe(Ⅲ)改性,并应用于微囊藻毒素的去除.结果表明,黏土含量高低决定土壤本身对毒素的吸附能力强弱;土壤经过离子改性后,其毒素吸附能力与其阳离子交换量大小直接相关,且毒素去除能力较未改性前有显著提高,其中效率提升最高的一种土壤离子改性后其吸附能力增加了约148倍.离子改性土壤对微囊藻细胞同样具有絮凝沉降功能,沉降能力同样随土壤对Fe(Ⅲ)负载能力的提高而上升,使用剂量提高时微囊藻沉降平衡时间明显缩短.在野外蓝藻水华去除实验中,蓝藻细胞去除可以保持与室内实验相近的高去除率,但蓝藻毒素的去除效率有一定的下降,可能与天然水体中其它杂质的竞争作用有关.  相似文献

我国淡水水体蓝藻水华的频繁发生已成为我国目前和今后长时期内面临的重大水环境问题.微囊藻(Microcystis)因其产生对人类健康具有危害的微囊藻毒素而尤其受到重视.研究者对构成蓝藻水华的主要种类、微囊藻毒素的毒性、毒理等方面的认识逐渐明晰,但对微囊藻毒素生物学功能的了解还相对较少.本文对微囊藻毒素的产生,尤其是近年来随着技术的发展在微囊藻毒素生物学功能方面的研究开展讨论:总结了微囊藻毒素可能作为化感物质、参与光合作用、在微囊藻的越冬或复苏中可能发挥的作用以及可能参与微囊藻群体形成及维持等4个方面的生物学功能;分析了微囊藻毒素作为信号分子的研究现状;探讨了全球气候变化及实验室模拟条件下微囊藻毒素产毒与无毒株的竞争,并对微囊藻毒素未来的研究热点及组学技术可能在其研究中的应用进行了展望.  相似文献

2013年秋季,洱海再次出现大规模水华,周边几十万居民的饮用水安全引起当地政府的极大关注.为了解水华发生期间洱海水体微囊藻毒素的含量和浮游植物的种类及丰度,分别在2013年9月和10月两次在洱海采集水样,测定其中两种主要微囊藻毒素MC-LR和MC-RR含量,探讨微囊藻毒素含量对洱海周边水源地的安全影响.同时也对水华期间的浮游植物特别是蓝藻进行研究,从而分析微囊藻毒素与浮游植物种类和丰度的关系.结果表明,洱海水体中微囊藻毒素含量偏低,暂不会对人类健康产生威胁,但在洱海的水鸟体内仍然检测到微囊藻毒素的富集,显示洱海蓝藻水华对水生生物的潜在危害不容忽视.浮游植物的研究发现,洱海水华的优势种属为微囊藻(Microcystis)和暗丝藻(Psephonema aenigmaticus).9月份暗丝藻更占优势,10月份则是微囊藻更占优势.研究结果显示处于富营养化初期的洱海水体中湖湾微囊藻毒素的含量与浮游植物中产毒蓝藻的种类和丰度密切相关.  相似文献

近年来,随着全球变暖和水体富营养化程度的日趋严重,淡水湖泊藻类水华现象频发;由此引发的微囊藻毒素对水生生物的生态毒性效应备受关注.本实验探究了4个温度(16、20、24和28℃)和3个微囊藻毒素MC-LR浓度(2.0、4.0和6.0μg/ml)对4个品系萼花臂尾轮虫(Brachionus calyciflorus)形态特征的影响.通过2种DNA分类方法——自动条形码查询系统和Yule溯祖模型将萼花臂尾轮虫划分为3个进化种;4个品系轮虫的形态学参数总体随温度升高呈减小趋势.在16℃下,轮虫的形态特征在各品系间未见显著差异;20℃下的昆明品系KMC23形态参数最小,而西宁XN2品系最大;在24℃下,西宁XN2品系的形态参数显著增大;兰州LZB1和西宁XN2品系对温度变化的响应特征相似且在28℃下最小,这可能与两者属于同一进化种有关.4个品系轮虫的卵特征参数对温度变化的响应各有不同.可见,品系和隐种的不同是造成4个轮虫克隆形态学参数对温度差异响应的重要内源性因素.各品系轮虫中,兰州LZB1品系受温度、MC-LR及两者交互作用的影响最为显著,形态可塑性最强.在高温胁迫下,兰州LZB1品系的所有形态学参数均显著减小,但MC-LR浓度对该品系轮虫的形态学参数并未产生显著影响;经低温胁迫后,低浓度的MC-LR没有显著改变兰州LZB1品系轮虫的形态学参数,而高浓度的MC-LR显著降低了轮虫的大部分形态学参数.微囊藻毒素对轮虫形态可塑性的影响受到了内源性因素和外源性因素的综合制约.  相似文献

利用改性粘土治理蓝藻水华堆积的湖泊近岸区域以及发生水华的养殖水体成为应急治理蓝藻的重要措施,然而负载有毒藻体的改性粘土沉积水体可能引发的安全性风险尚缺乏研究.选取PAC改性粘土作为研究对象,通过模拟实验,研究改性粘土处理蓝藻水华后沉积水体对负载的藻细胞结构、水体营养盐与藻毒素的释放与削减以及对其他水体功能性细菌(硝化、反硝化细菌)的影响.结果表明,PAC改性粘土的施用对低水华水体蓝藻细胞的去除率为96.04%±0.99%,高水华水体与低水华水体施用的去除率之间差异不显著,PAC改性粘土的施用能够在较长的时间内有效地控制水体中的蓝藻浓度.透射电镜结果表明,PAC改性粘土沉降蓝藻第4 d后,蓝藻细胞膜出现了一定程度的不完整,细胞内规则的类囊体片层结构出现了实质性损伤.随着粘土负载蓝藻细胞的损伤,水体中的总可溶性氮浓度迅速上升,但总可溶性磷浓度仍可在较长一段时间内维持在较低水平.PAC改性粘土施用后,水体的总细菌数与空白对照组趋于一致,但硝化、反硝化细菌数却呈显著下降趋势.PAC改性粘土施用下的藻毒素释放风险主要集中在高水华水体.高水华水体中,PAC改性粘土施用导致藻毒素MC-LR和MC-RR加速释放,其最高峰值分别达到空白对照组的1.69±0.09和2.04±0.09倍,但水体MC-LR浓度达到安全限(1μg/L)的时间明显比空白对照组早8 d.此外,PAC改性粘土的施用并没有导致水体中Al~(3+)和Cl~-浓度超标.  相似文献

蓝藻暴发及其打捞后的随意堆放,会产生藻毒素、多环芳烃、臭味气体等有毒物质.通过实验室模拟巢湖蓝藻死亡衰败过程,检测其中的有机元素、沥青(A)、饱和烃、芳烃等含量变化,探讨了死亡衰败阶段蓝藻有害成分尤其是多环芳烃等有毒物质的形成及控制对策.结果表明,蓝藻死亡衰败过程中,经兼性工艺处理的蓝藻其芳烃类总含量最低,多环芳烃的种类和含量也最低.单纯厌氧作用使芳烃含量略有升高,而单纯的好氧作用使饱和烃含量增高,并随作用时间的延长而增加,在好氧处理3个月的蓝藻中达到最高值.因此,本文认为兼性工艺处理的蓝藻最适于堆肥等资源化利用,其发酵液通入农田用作肥料,气体收集用于燃烧.并且提出依靠蓝藻死亡衰败过程中的藻胆蛋白、叶绿素等的荧光强度来监测其死亡衰败进程,进而控制其有害成分形成的对策.  相似文献

浮游蓝丝藻是水华蓝藻的常见种属,但对其生长和产毒特性认识不多本文首次对我国水体中的浮游蓝丝藻进行了研究.从广东省一重要供水水库中分离到一株丝状蓝藻,经形态特征鉴定为阿氏浮游蓝丝藻,并在不同的光照和温度下对该藻株进行了培养.结果表明,适合于该藻生长的光强和温度分别为20μmol/(m~2·s)和30℃;该藻株具有mcyE基因,能产生微囊藻毒素MCRR,产量为0.06μg/mg(DW).  相似文献

鄱阳湖不同湖区营养盐状态及藻类种群对比   总被引:4,自引:1,他引:3  
在平水期、丰水期和枯水期对鄱阳湖典型天然碟形湖、人控湖汊和主湖区进行了水质、藻类和蓝藻毒素等对比调查,结果表明鄱阳湖各个湖区的水质与藻类种群等差异较大,蓝藻毒素浓度和底泥中铁含量的分布具有一定关联性.在各水文季节蓝藻均为人控湖汊藻类的主要优势种之一.平水期鄱阳湖藻类生物量(叶绿素a浓度)与水体的pH呈正相关关系,与采样点的水深呈负相关关系,碟形湖区水体营养盐浓度和藻类细胞密度均较其他湖区水体低.丰水期各湖区的水质差异相对较小,碟形湖藻类细胞密度仍低于其他湖区,但蓝藻已成为各湖区的优势种,该时期藻类生物量与水体总磷浓度及浊度呈正相关关系.枯水季鄱阳湖各水体藻类生物量与水体总氮浓度、铵态氮浓度及电导率呈正相关关系,碟形湖与主湖区发生了完全分离,水体流动性差,暴发蓝藻水华的风险较高.高温丰枯季节鄱阳湖水体蓝藻毒素浓度与底泥铁含量呈现一定的相关分布关系,底泥铁含量高的地方,其水体蓝藻毒素浓度通常比较高,应警惕鄱阳湖流域富铁红壤流失带来的湖区蓝藻水华风险加剧后果.上述研究结果将为鄱阳湖水环境的预警和污染控制提供科学指导.  相似文献

The indiscriminate use of antimicrobials especially in developing countries has evoked serious bacterial resistance and led to the emergence of new and highly resistant strains of bacteria to commonly used antimicrobials. In Lebanon, pollution levels and bacterial infections are increasing at a high rate as a result of inadequate control measures to limit untreated effluent discharges into the sea or freshwater resources. The aim of this study was to isolate and molecularly characterize various Staphylococcus strains isolated from sea water, fresh water, sediments, and crab samples collected from representative communities along the coast of Lebanon. The results on the antimicrobial resistance indicated that the level of resistance of Staphylococcus aureus varied with various antimicrobials tested. The resistance patterns ranged between 45% in freshwater isolates and 54.8% in seawater ones. Fifty one percent of the tested isolates have shown resistance to at least one of the five tested antimicrobials; with seawater isolates exhibiting the highest rates of antimicrobial resistance.  相似文献

Analysis of trace amounts of various hepatotoxic microcystins in marine and freshwater samples is very important since these toxins, especially microcystin-LR, have been demonstrated to have tumour-promoting activity. In this study, instead of measuring the total amount of microcystins, we developed a capillary zone electrophoretic method for the separation and detection of individual toxin standards. No additives were used for enhancement of resolution. This technique is characterized by a high separation efficiency, short analysis time and small sample volume. In order to improve the detection sensitivity, a laser-induced fluorescence detector was used, and the labelling of microcystins was accomplished through a two-step procedure. First, the microcystin standards were converted into cysteine conjugates, followed by derivatization with Fluorescein 5-Isothiocyanate (FITC). After derivatization, the FITC-labelled microcystins were directly injected, separated and detected in 8 min. This method was shown to be a promising technique for sensitive and rapid analysis of individual microcystin toxins.  相似文献

Lamellidens marginalis (Molluska Bivalvia Eulamellibranchiata) is an important member of the freshwater ecosystem of India, which is sustained by filtering phytoplankton, bacteria and other particulate organic matter from the available water. Hemocytes, the circulating blood cells of bivalves, function as immunological effector cells under exposure to toxins and/or parasites. The hemocytes of Lamellidens marginalis have been identified morphologically and enumerated as being qualified for use as a biomarker to analyze the threat of freshwater contamination by sublethal concentrations of sodium arsenite, a natural pollutant of freshwater ecosystems. Diverse subpopulations of hemocytes were identified as blast‐like cells, granulocytes, agranulocytes, hyalinocytes and asterocytes. Supression in the total count of hemocytes and blast‐like cells was recorded under the exposure of all the concentrations of sodium arsenite tested. Sodium arsenite elevated the relative density of granulocytes, hyalinocytes, and asterocytes. Partial restoration of the total count of the hemocytes was recorded after the post‐treated animals were maintained in arsenic‐free water for periods of 15 and 30 days. The data predict a shift in the immunological parameters of this bivalve in arsenic contaminated environments. The present study is aimed at quantifying the arsenic induced stress in Lamellidens marginalis and establishing the hemocyte density as a biomarker of aquatic pollution in selected geographical regions of India.  相似文献

Microcystins are cyclic heptapeptide toxins produced by a range of cyanobacterial genera. These cyanobacteria occur naturally in drinking water reservoirs subject to eutrophication, and in rivers and natural lakes. Because of the diversity of organisms, the toxins occur, from oligo‐mesotrophic lakes in North Temperate latitudes, to hypertrophic tropical ponds. The toxins are responsible for numerous cases of injury and death of domestic animals, and human poisoning from drinking water. The initial poisoning includes hepatic cell death. This leads to secondary effects from liver deficiency, including jaundice and photosensitisation. The toxic effects are largely due to inhibition of phosphatase enzymes, acting to regulate protein phosphorylation. The consequences include structural damage, apoptosis and, at lower concentrations, cell cycle effects and tumour promotion. As there is no clear evidence for direct carcinogenesis by microcystins, they are classed as non‐carcinogenic toxins in drinking water. Guideline Values for safe drinking water are derived from data for subchronic rodent toxicity, using the No Observed Adverse Effect Level (the highest dose giving no toxicity). To this dose are applied uncertainty factors, to calculate a Tolerable Daily Intake. On the basis of a standard bodyweight and water consumption the Guideline Value is determined for drinking water. For microcystin‐LR the WHO have set a provisional Guideline Value of 1 μg/L for drinking water.  相似文献

Purple clams (Hiatula rostrata Lighttoot) accumulate paralytic shellfish poisoning (PSP) toxins produced by a toxic strain of the dinoflagellate Alexandriun minutum Halim. The results confirm the data of our previous study concerning the muscle and siphon that were not showing a gradual rise in toxicity when shellfish accumulated more A. minutum. However, muscle and siphon are intermittently toxic both in exposure and depuration period in laboratory cultured purple clams. PSP toxins were detected in outdoor cultured purple clams, whereas no A. minutum were found in the culture pond during most of the survey time. The outdoor cultured purple clams need longer time to decrease toxicity to allowable levels than laboratory cultured purple clams. It was shown that laboratory data may not predict times over which pond-cultured purple clams may prove toxic to consumers.  相似文献

The Bahama Islands contain many abandoned dissolution caves at elevations between two and seven metres above current sea level. The development of dissolution caves in tropical carbonate islands is dependent on the position and nature of the freshwater lens. Lens position is controlled by sea level, which in stable carbonate platforms like the Bahamas is a function of glacioeustatic sea level still stands. Caves in the Bahamas that are currently subaerial must have developed during past higher sea levels. During the Late Quaternary, sea levels higher than present have been relatively short-lived, and that limits the amount of time that a freshwater lens could be situated at the elevation required for the cave formation. The Bahama Islands are low-lying landforms where only aeolian ridges extend to elevations higher than six metres above current sea level. Past high sea level events greatly reduced the exposed land area of the Bahama Islands, thus also limiting both the catchment for and size of freshwater lenses. Caves must be younger than the rock in which they are developed; most subaerial Bahamian caves are found in limestones that are less than 150000 years old. Development of large dissolution caves under these limitations of time and lens size requires a powerful dissolutional mechanism. The mixing of discharging freshwater with tide-pulsed incoming marine water under the flanks of emergent dune ridges may have produced the conditions necessary. Bahamian caves formed by this process are phreatic chambers with complex interconnections and blind tubes. Their presence demonstrates that significant dissolution can occur rapidly as a result of the mixing of fresh and marine waters beneath small carbonate islands.  相似文献

Isolates of Microcystis blooms collected from Dianchi Lake of South-western China in different seasons were tested by mouse bioassays. The LD50 of the vcdcrocystis isolates corre-sponding M. wesenbergii, M.viridis and M. aeruginosa were 221 mg · kg -1, 188 mg · kg -1, and 198 mg · kg -1 respectively. The cells of the three samples contained hepatotoxins or microcystins. The HPLC (high performance liquid chromatography) analysis showed that all the three Microcystis species contained microcystin LR. In addition, M. wesenbergii contained microcystin RR, M. viridis contained microcystinYR, M. aeruginosa contained both microcystin RR and YR. The results suggested that in different seasons the toxins which were produced by the dominating Microcystis species were fluctuent with regard to the composition and content of the toxins and consen-quently were obvious factors that aroused the water quality to decrease and change.  相似文献

The Ischia hydrothermal system was analysed through hydrogeological and microbial community investigations. Mesophilic communities were detected in two cold springs, suggesting a negligible influence of thermal circuits in freshwater sub‐systems which are mainly or only fed by local precipitations. Thermophilic and extremely thermophilic bacteria were detected in two wells, according to higher water temperatures (61 and 85 °C), even if the two communities show significant differences. In one well, thermophilic and extremely thermophilic bacteria are associated with strains belonging to ε‐Proteobacteria isolated in different sulphur‐rich carbonate environments. This association suggests a greater influence on ascending hot fluids that interact with the carbonate basement of volcanic rocks. In the other well, thermophilic and extremely thermophilic bacteria are associated with strains isolated in cold hypersaline environments or in aquatic habitats where terrestrial and marine components are coupled. This association supports the fact that seawater intrusion can affect this part of Ischia, according to results of hydrogeological and geochemical surveys. Differences in groundwater temperature and bacterial communities are probably mainly due to differences in permeability between volcanic rocks and differences in hydrogeologic behaviour between faults in the upper carbonate basement, above the deep magma chamber, that influence relationships between ascending hot fluids and local recharge. This study contributes to discussion of the reliability of the actual behaviour models of the Ischia system, based on the results of geochemical and isotopic investigations, and, in a wider context, it shows that microbial community investigations may be a valuable supplementary tool for analysing hydrothermal system behaviour. Reduced biological diversity in freshwater habitats situated in urban areas has been discussed in numerous studies. Certain municipal areas, however, can help save animal diversity of freshwater invertebrates. In the present study animals were collected or observed alive in 13 freshwater environments localized in Warsaw - the second largest city of Central Europe - in a densely populated, urban building complex close to the city, and also in suburban areas. Leech assemblages in all the environments under observation were numerically dominated by a few common species, but on the whole 19 species were collected or observed. The populations of six rare leech species inhabit both flowing and standing waters in Warsaw. Five of these species are on the Polish Red List of Species and one is strictly protected. The shallow Lake Powsinkowskie is the richest freshwater environment in the studied area in terms of species richness and rarity and also one of the richest lakes in Poland. Taxonomic diversity in the environments under study seems not to be directly related to the size of the water body or the level of degradation but rather to the habitat complexity, especially the diversity of the bottom in littoral zone. Certain freshwater habitats located inside this great urban complex still create good conditions for rare, highly specialized species.  相似文献

River floods influence sedimentary environments and ecosystems from the terrestrial to the deep-marine. This study documents the occurrence conditions of hyperpycnal flows generated by river floods and related organic-matter sedimentation for Holocene sediments of the Niigata Plain, Central Japan, based on detailed sedimentary facies, total sulfur and total organic carbon content, diatom assemblages and organic-matter composition. Holocene sediments of the Niigata Plain consist of sand, mud and gravel that were deposited in estuarine and fluvial systems during a sea-level rise (15 000–6800 years BP) and stillstand (after 6800 years BP) following the Last Glacial Maximum. Hyperpycnites are present in the upper part of the estuarine lagoon sediments. The depositional age is considered to be about 5000 years BP. The hyperpycnites comprise two successions of a top fining-up unit and a basal coarsening-up unit, and include abundant terrigenous organic matter and freshwater diatoms. A large volume of freshwater is inferred to have flowed into the lagoon during deposition of the upper part of the lagoon sediments. In consequence, hyperpycnal flows may have readily formed in the lagoon, because the halocline was weak. The hyperpycnal flows also produced a layer of concentrated terrigenous organic matter in the uppermost part of the hyperpycnites. The abundant organic matter on the estuarine floor is inferred to have produced anoxic bottom conditions owing to oxidative decomposition by benthic bacteria.  相似文献
