采用改良微孔板法建立哈维氏弧菌Vibrio harveyi TS-628菌株生物被膜体外模型,并进一步研究环境因子对哈维氏弧菌体外形成生物被膜的影响。结果发现,哈维氏弧菌TS-628菌株可以在聚苯乙烯板中形成生物被膜,并且该菌生物被膜的形成受起始菌液浓度、温度、pH等理化因子的显著影响。温度为30℃、NaCl质量分数为3%—6%、pH为7、孵育时间为36h是哈维氏弧菌形成生物被膜的最佳条件。此外哈维氏弧菌在分别经表皮黏液、鳃黏液、肠道黏液、肝脏提取液、脾脏提取液、肌肉提取液包被的基质上成膜效果显著,其中在经肝脏提取液包被的基质上形成的生物被膜量显著高于在其他生物基质上形成的生物被膜量(p<0.01)。  相似文献

The effects of elevated atmospheric carbon dioxide [CO2] on microbial communities in arid rhizosphere soils beneath Larrea tridentata were examined. Roots of Larrea were harvested from plots fumigated with elevated or ambient levels of [CO2] using Free-Air CO2 Enrichment (FACE) technology. Twelve bacterial and fungal rRNA gene libraries were constructed, sequenced and categorized into operational taxonomical units (OTUs). There was a significant decrease in OTUs within the Firmicutes (bacteria) in elevated [CO2], and increase in Basiomycota (fungi) in rhizosphere soils of plots exposed to ambient [CO2]. Phylogenetic analyses indicated that OTUs belonged to a wide range of bacterial and fungal taxa. To further study changes in bacterial communities, Quantitative Polymerase Chain Reaction (QPCR) was used to quantify populations of bacteria in rhizosphere soil. The concentration of total bacteria 16S rDNA was similar in conditions of enriched and ambient [CO2]. However, QPCR of Gram-positive microorganisms showed a 43% decrease in the population in elevated [CO2]. The decrease in representation of Gram positives and the similar values for total bacterial DNA suggest that the representation of other bacterial taxa was promoted by elevated [CO2]. These results indicate that elevated [CO2] changes structure and representation of microorganisms associated with roots of desert plants.  相似文献

Back-barrier tidal flat systems are characterized by basins and inlets through which water is exchanged with the coastal sea by tidal water movements. The hydrographic and morphometric properties at the inlets and in the basins vary considerably, but there is little information available how biogeochemical properties in the water column at these different sites respond to these differences. Therefore, we investigated tidal dynamics of suspended particulate matter (SPM), particulate and dissolved organic carbon (DOC), chlorophyll a, phaeopigments, numbers of particle-associated (PA) and free-living bacteria (FL), bacterial biomass production, and concentrations of dissolved manganese (Mn). Samples were taken at the surface, a mid-depth and 1 m above the bottom at a fixed station at the inlet and in the basin of the Spiekeroog back-barrier tidal flat system in the German Wadden Sea. Five tidal cycles representative for typical seasonal situations, January (winter), April and May (late spring bloom), July (summer), and November (late fall) were studied in 2005 and 2006. In July, processes related to phytoplankton dynamics and bacterial decomposition were much more enhanced in the basin, whereas in April, these processes were enhanced at the inlet but were particularly low within the basin itself. The low values within the basin were a result of the settled phytoplankton spring bloom and represent a rather short period at the decline of this bloom. In November and January, differences were much less pronounced than during the growing season and restricted mainly to SPM and PA bacteria, exhibiting higher values in the basin. FL bacteria, DOC, and dissolved Mn exhibited different patterns and much less differences between the two stations, indicating that biogeochemical processes in the dissolved phase were controlled by different factors than PA biogeochemical processes. These differences reflect the retentive properties of the basin for particles and PA biogeochemical processes, particularly during the growing season, and in general emphasize the high productivity of back-barrier tidal flat systems.  相似文献

The effects of shrimp farm effluents on bacterial communities in mangroves have been infrequently reported. Classic and molecular biology methods were used to survey bacterial communities from four mangroves systems. Water temperature, salinity, pH, total heterotrophic bacteria and maximum probable numbers of Vibrio spp. were investigated. Genetic profiles of bacterial communities were also characterized by polymerase chain reaction (PCR) amplification of eubacterial and Vibrio 16S rDNA using denaturing gradient gel electrophoresis (DGGE). Highest heterotrophic counts were registered in the mangrove not directly polluted by shrimp farming. The Enterobacteriaceae and Chryseomonas luteola dominated the heterotrophic isolates. Vibrio spp. pathogenic to humans and shrimps were identified. Eubacterial genetic profiles suggest a shared community structure independent of mangrove system. Vibrio genetic profiles were mangrove specific. Neither microbial counts nor genetic profiling revealed a significant decrease in species richness associated with shrimp farm effluent. The complex nature of mangrove ecosystems and their microbial communities is discussed.  相似文献

Serial data from soil–sawdust column experiments were used to develop a mathematical model to describe the biological sulfate reduction processes in porous media. The mathematical model numerically solves the equation of solute transport in one-dimensional saturated state. Solute transport is coupled to sulfate reducing bacteria sub model. Bacterial growth is assumed to follow double Monod kinetic equation. Two bacterial groups (X1 and X2) were described. Bacterial group X1 uses under aerobic conditions oxygen and under anaerobic conditions nitrate as electron acceptor. Under anaerobic conditions bacterial groups X2, use sulfate as an electron acceptor. Sulfate rich wastewater is usually deficient in electron donor and requires external addition of electron donors in order to achieve complete sulfate reduction. The organic carbon as electron donor is one of the most important factors that affect sulfate reduction bacterial activity. In this study the possible source of organic carbon is the solid organic carbon supplied to the system in the form of sawdust. The results of this study indicate that sawdust could be employed as low-cost materials to enhance the biological sulfate reduction processes in porous media. While the availability of organic carbon as electron donor is one of the most important factors that affect the sulfate reducing bacterial activity in porous media, this study demonstrates that using sawdust as a carbon source can improve the bacterial activity and increase the column permeability.  相似文献

Five years of monthly data of indicator bacteria from 1998 to 2002 were evaluated to find out the changes in water quality during the rehabilitation of the Golden Horn, an estuary severely polluted from industrial and domestic discharges since the 1950s. Surface fecal coliform was above 10(6) CFU/100 ml at the inner part in 1998. Following the achievement of healthy water circulation and control of most surface discharges, fecal coliform and fecal streptococci counts decreased below 10(3) CFU/100 ml in the summer of 2002. However, the decrease was interrupted by sudden shifts in rainy periods. Runoff, enhanced by domestic inputs during rainfall, has become the main factor influencing water quality in the estuary today. Increasing values of fecal coliform were observed during periods of low salinity, pH, dissolved oxygen and high ortho-phosphate, whilst decreasing values were detected during high salinity, pH and dissolved oxygen and low ortho-phosphate periods. Striking changes were observed within five years, promising that even an anoxic water body can turn into a recreational area with appropriate treatment.  相似文献

The early diagenetic chemical dissolution of skeletal carbonates has previously been documented as taking place within bioturbated, shallow water, tropical carbonate sediments. The diagenetic reactions operating within carbonate sediments that fall under the influence of iron‐rich (terrigenous) sediment input are less clearly understood. Such inputs should modify carbonate diagenetic reactions both by minimizing bacterial sulphate reduction in favour of bacterial iron reduction, and by the reaction of any pore‐water sulphide with iron oxides, thereby minimizing sulphide oxidation and associated acidity. To test this hypothesis sediment cores were taken from sites within Discovery Bay (north Jamaica), which exhibit varying levels of Fe‐rich bauxite sediment contamination. At non‐impacted sites sediments are dominated by CaCO3 (up to 99% by weight). Pore waters from the upper few centimetres of cores show evidence for active sulphate reduction (reduced SO4/Cl? ratios) and minor CaCO3 dissolution (increased Ca2+/Cl? ratios). Petrographic observations of carbonate grains (specifically Halimeda and Amphiroa) show clear morphological evidence for dissolution throughout the sediment column. In contrast, at bauxite‐impacted sites, the sediment is composed of up to 15% non‐carbonate and contains up to 6000 μg g?1 Fe. Pore waters show no evidence for sulphate reduction, but marked levels of Fe(II), suggesting that bacterial Fe(III) reduction is active. Carbonate grains show little evidence for dissolution, often exhibiting pristine surface morphologies. Samples from the deeper sections of these cores, which pre‐date bauxite influence, commonly exhibit morphological evidence for dissolution implying that this was a significant process prior to bauxite input. Previous studies have suggested that dissolution, driven by sulphate reduction and sulphide oxidation, can account for the loss of as much as 50% of primary carbonate production in localized platform environments. The finding that chemical dissolution is minor in a terrigenous‐impacted carbonate environment, therefore, has significant implications for carbonate budgets and cycling, and the preservation of carbonate grains in such sediment systems.  相似文献

目的 探索Sysmex UF-1000i尿沉渣分析仪定量检测结果中细菌和白细胞计数在诊断尿路感染中的价值,阐明标本采集送检的不同对UF-1000i诊断尿路感染的影响。方法 选取笔者医院2015年7月~2015年8月466例怀疑尿路感染的入院患者同时做细菌培养及尿常规的检验结果(A组标本),收集2016年3月下旬~4月上旬148例尿细菌培养清洁中段尿标本,接种之后立即送检尿常规检查(B组标本)。取两组标本的细菌及白细胞计数结果,以细菌培养为金标准用SPSS 18.0统计软件分别绘制ROC曲线,求出尿沉渣细菌和白细胞计数诊断尿路感染的阈值,并计算其敏感度、特异性、阳/阴性预测值、假阳性/假阴性率和准确性,比较两组标本的各项指标。结果 尿沉渣细菌与白细胞定量检测最佳阈值,A组标本分别为101.7/μl与18.8/μl,B组标本分别为98.7/μl与11.1/μl。尿沉渣细菌与白细胞ROC曲线下面积,A组标本分别为0.604与0.661,B组标本分别为0.941与0.848。对B组标本而言,细菌与白细胞联合诊断尿路感染最佳敏感度为82.4%,特异性为92.1%,阳性预测值为77.8%,阴性预测值为93.8%,假阳性率为7.9%,假阴性率为17.6%,准确度为89.9%,并可将细菌培养降低70.9%。结论 UF-1000i尿液沉渣分析仪在诊断尿路感染中有早期筛选,辅助诊断的价值。标本达到无菌采集、中段尿并在2h内送检的要求才能使UF-1000i发挥最大价值。  相似文献

In the eastern North Water, most of the estimated annual new and net production of carbon (C) occurred during the main diatom bloom in 1998. During the bloom, at least 30% of total and new phytoplankton production occurred as dissolved organic carbon (DOC) and was unavailable for short-term assimilation into the herbivorous food web or sinking export. Based on particle interceptor traps and 234Th deficits, 27% of the particulate primary production (PP) sank out of the upper 50 m, with only 7% and 1% of PP reaching the benthos at shallow (≈200 m) and deep (≈500 m) sites, respectively. Mass balance calculations and grazing estimates agree that ≈79% of PP was ingested by pelagic consumers between April and July. During this period, the vertical flux of biogenic silica (BioSi) at 50 m was equivalent to the total BioSi produced, indicating that all of the diatom production was removed from the euphotic zone as intact cells (direct sinking) or empty frustules (grazing or lysis). The estimated flux of empty frustules was consistent with rates of herbivory by the large, dominant copepods and appendicularians during incubations. Since the carbon demand of the dominant planktivorous bird, Alle alle, amounted to ≈2% of the biomass synthesized by its main prey, the large copepod Calanus hyperboreus, most of the secondary carbon production was available to pelagic carnivores. Stable isotopes indicated that the biomass of predatory amphipods, polar cod and marine mammals was derived from these herbivores, but corresponding carbon fluxes were not quantified. Our analysis shows that a large fraction of PP in the eastern North Water was ingested by consumers in the upper 50 m, leading to substantial carbon respiration and DOC accumulation in surface waters. An increasingly early and prolonged opening of the Artic Ocean is likely to promote the productivity of the herbivorous food web, but not the short-term efficiency of the particulate, biological CO2 pump.  相似文献

新一代喹诺酮药物——6—F—喹啉—3—羧酸(商品名齐复康Ⅱ号)对中国对虾幼体的毒性,蚤状幼体、糠虾幼体、仔虾24hLC_(50)分别为16.3、21.3、45.8(×10~(-6));48hLC_(50)分别为8.1、12.5、30.0(×10~(-6));安全浓度(SC)分别为0.6、1.3、3.8(×10~(-6))。试验表明此药对防治对虾幼体细菌性疾病效果明显。0.5×10~(-6)。每天施药一次,连续三天,育苗池水中细菌总数由30750ind./ml减少到6850ind./ml,其中在TCBS培养基上能生长的细菌由22400ind./ml减少到1090ind./ml。  相似文献
