The sorption of AuCl4 ,AuCl2 and Au(S2O3)3- on δ-MnO2 was investigated at pH2–11.6, 0.01 mol/L and 0.1 mol/L NaNO3 solutions. At pH 4 in two electrolyte strength solutions, Au sorption densities on δ-MnO2 are 0.18–0.21 and 0.28μmoL/m2 for AuCl4 and Au(S2O3)2 3-, respectively, and the Au surface coverage is approximate to or lower than 1%. This adsorption of the two Au complexes decreases as the solution pH increases, which conforms to the sorption regularity of the anion on δ-MnO2. The Au sorption decreases in the sequence of Au(S2O3)2 3- >AuCl4 >AuC12 . The intrinsic equilibrium constants (logK int) of the three Au complexes are 1.17–2.7, much higher than those of Cu and Cd. The hydrolysis products of AuCl4 - are preferentially adsorbed by δ-MnO2 and the inner-sphere Au-surface complexes are formed on the surface. An enzyme assay was developed for studies on peroxidase activities in humic lake water. 3,4-Dimethoxybenzyl alcohol (veratryl alcohol, VeraOH) was used as tracer substrate, and peroxidase (EC 1.11.1.7) activity was measured by high-performance liquid chromatography. The chemical stability of VeraOH and its application as peroxidase substrate was tested under light and dark conditions, different hydrogen peroxide (H2O2) concentrations and humic matter contents. VeraOH was stable under low UV radiation at in situ conditions in lake water (<0.010...0.25 kJ m–2 d–1), laboratory conditions (<0.05...0.30 kJ m–2 d–1), and low (1...100 μM) H2O2 concentrations. However, peroxides oxidized VeraOH above 1...10 mM H2O2 concentration in sterile Millipore-Q and humic lake water. Dark incubations showed little VeraOH oxidation products. The developed peroxidase assay was tested in the growth medium of Phanerochaete chrysosporium and a bacteria isolate (P.M.D. 20.4.3.1) from mesohumic lake Pääjärvi. Peroxidase activities were also measured in natural microbial communities under standard laboratory and under in situ conditions in humic lake water. Incubation times of about 5 to 12 days were usually needed to record significant (P < 0.05) peroxidase activities, in lake waters. In situ peroxidase activities varied in pelagial surface water (0...0.5 m) on a seasonal scale between 74 nmol L–1 h–1 and 273 nmol L–1 (mean: 176 nmol L–1 h–1) and within the water column between 110 nmol L–1 h–1 and 800 nmol L–1 h–1 (mean: 500 nmol L–1 h–1) in polyhumic lake Mekkojärvi.  相似文献

以香鱼卵母细胞为研究对象,采用观察卵内油球、有关酶的活力、以及卵的受精率和孵化率等变化的方法,研究了香鱼卵母细胞长时效演化的相关机理,结果表明,随着香鱼卵在腹腔内保存时间的延长,卵内油球从0h的油球直径≤10μm,且均匀分布,逐渐融合增大,至108h增大到120μm,油球数仅有5—6个。同时,SOD、POD的活力及MDA含量均呈先升后降的变化趋势,最大峰值分别出现在60h、36h和24h。随着油球和酶活的变化,卵子质量逐渐下降,表现为受精率和孵化率的降低:排卵后24h、48h、72h、96h的受精率分别为94.37%、88.19%、50.20%、10.83%,孵化率分别为85.78%、68.14%、25.54%、0.53%。  相似文献

Ultrasound-assisted solvent extraction was confirmed as a novel,effective method for separating lipid from mackerel pro-tein,resulting in a degreasing rate (DR) of 95% and a nitrogen recovery (NR) of 88.6%.To obtain protein hydrolysates with high ni-trogen recovery and low bitter taste,enzymatic hydrolysis was performed using eight commercially available proteases.It turned out that the optimum enzyme was the ‘Mixed enzymes for animal proteolysis’.An enzyme dosage of 4%,a temperature of 50℃,and a hydrolysis time of 300 min were found to be the optimum conditions to obtain high NR (84.28%) and degree of hydrolysis (DH,16.18%) by orthogonal experiments.Glutamic acid was the most abundant amino acid of MDP (defatted mackerel protein) and MDPH (defatted mackerel protein hydrolysates).Compared with the FAO/WHO reference protein,the essential amino acid chemical scores (CS) were greater than 1.0 (1.0 1.7) in MDPH,which is reflective of high nutritional value.This,coupled with the light color and slight fishy odor,indicates that MDPH would potentially have a wide range of applications such as nutritional additives,functional ingredients,and so on.  相似文献

Pits in ocean basalt glass are often attributed to the activity of microorganisms, however, neither the mechanism of formation of pits in glass nor the involvement of microorganisms have been confirmed by experimentation. Experimental abiotic corrosion of basalt glass with 1% hydrofluoric acid (HF), a proxy for more slowly acting organic acids, produces pits that are similar in size, shape, and distribution to pits found in basalt glass collected from the ocean floor and basalt glass incubated with microorganisms. This pit formation by HF etching was demonstrated by comparing secondary electron images taken before and after the acid treatment. The formation of pits by abiotic corrosion of basalt glass is an alternate hypothesis for the origin of these features, and the interpretation of these features as biomarkers may require a resolution of these alternative hypotheses.  相似文献

研究了动物蛋白酶 1 0 58水解鱼是鱼蛋白时 ,酶量、温度、作用时间对鱼是鱼蛋白质水解率和苦味的影响。结果表明 :酶浓度的提高 ,水解时间的延长 ,均使鱼是鱼蛋白水解率明显提高。最适水解温度为 50℃ ,当温度高于 50℃时 ,水解率逐渐下降 ,蛋白酶开始失活。苦味值随水解率提高而增加。用正交试验的方法 ,经综合分析和试验得出 ,该蛋白酶水解鱼是鱼的最佳条件是酶量与底物之比为 1 .5∶ 1 0 0 0 ;温度为 50℃ ;水解时间为 4h。  相似文献
