In this study, marine microplankton were identified by combining standard light microscopy with Sanger 18S rRNA gene sequencing. The image-matching individual PCR technique was applied to identify the image collectable unicellular microplankton to genera. Instead of pure strain culture and morphological identification, microplankton individual cells were isolated and fixed with glutaraldehyde, frozen and stored for months. Finally, they were imaged under a microscope and molecularly identified via phylogenetic analysis of their 18S ribosomal RNA gene(18S rDNA). Microplankton cells were collected at 30 locations in South China Sea, and were assigned to 21 known and 4 unidentified genera(2 uncultured fungi and 2 uncultured stramenopiles) with phylogenetic analysis in parallel to the morphological identification.  相似文献

丽江2.4 m望远镜的圆顶采用方位随动和滑动式天窗的超半球结构,起初为改善因圆顶和望远镜终端电子设备引起的内外大气湍流,设计了大面积的多组机械式侧窗。但由于高平台手动操作危险、缓慢、不精确且易损坏,为实现侧窗稳定的多组自动控制,开发了基于STM32板的嵌入式圆顶侧窗自动控制系统,利用WiFi模块、串口模块和手柄实现侧窗的远程和多通道控制。同时结合气象数据、圆顶位置信息等使系统能根据气象阈值进行预警、自动开合,并尽量减小风对望远镜振动的影响。侧窗控制系统的设计可满足上层系统集成的需要。该系统稳定可靠,能满足侧窗的自主运行与人为控制。  相似文献

A three-dimensional general mathematical hydroelastic model dealing with the problem of wave interaction with a floating and a submerged flexible structure is developed based on small amplitude wave theory and linear structural response. The horizontal floating and submerged flexible structures are modelled with a thin plate theory. The linearized long wave equations based on shallow water approximations are derived and results are compared. Three-dimensional Green’s functions are derived using fundamental source potentials in water of finite and infinite depths. The expansion formulae associated with orthogonal mode-coupling relations are derived based on the application of Fourier transform in finite and infinite depths in case of finite width in three-dimensions. The usefulness of the expansion formula is demonstrated by analysing a physical problem of surface gravity wave interaction with a moored finite floating elastic plate in the presence of a finite submerged flexible membrane in three-dimensions. The numerical accuracy of the method is demonstrated by computing the complex values of reflected wave amplitudes for different modes of oscillation and mooring stiffness. Further, the effect of compressive force and modes of oscillations on a free oscillation hydroelastic waves in a closed channel of finite width and length for floating and submerged elastic plate system is analysed.  相似文献

羽毛粉是一种潜在的饲料蛋白源。在水产动物饲料中添加适量的羽毛粉,不仅可以降低饲料成本,而且有助于禽类废弃物的再利用。在动物消化、降解、吸收羽毛粉过程中,动物肠道中的微生物可能发挥着重要作用;添加羽毛粉的饲料对鱼类肠道微生物多样性与群落结构的影响目前还不清楚。基于细菌16SrRNA基因高通量测序,作者对比研究了以添加羽毛粉(包括水解、酸解、膨化处理或生羽毛粉实验组,添加量为5%)和不添加羽毛粉的饲料(即对照组,包括鱼粉和商业饲料两组)喂养黑鲷12周后,黑鲷肠道细菌多样性与群落结构的变化情况。统计分析结果表明:添加羽毛粉的实验组中肠道细菌的丰富度、辛普森指数和香农指数显著低于对照组(P0.05);不含酸解羽毛粉组的实验组,其细菌群落结构与对照组差异显著(P0.05)。总体来看,黑鲷肠道细菌主要由厚壁菌门(57.3%)和变形菌门(36.3%)组成;实验组细菌群落中拟杆菌纲、梭菌纲、δ-变形菌纲、草酸杆菌科和青枯菌属的相对丰度显著低于对照组。不同的羽毛粉处理组之间也呈现出群落结构上的差异:与水解羽毛粉组相比,生羽毛粉组的变形菌门、γ-变形菌门、海洋螺菌目、盐单胞菌科和盐单胞菌属的相对丰度显著下降;与鱼粉组相比,生羽毛粉组中芽孢杆菌目在群落中的占比显著上升,而拟杆菌门、草酸杆菌科和青枯菌属的占比则显著下降,而且生羽毛粉组中厚壁菌门与变形菌门相对丰度的比值(7.42)明显高于鱼粉组(0.97)和水解羽毛粉组(0.74)且均接近显著(P=0.057)。与鱼粉组相比,水解羽毛粉组中紫单胞菌科和回线属的相对丰度显著下降,而盐单胞菌属的相对丰度显著上升。  相似文献

ABSTRACT

The authors employ the global production network (GPN) approach to analyse the development of the renewable energy sector. Through a case study of the development of a Hywind floating offshore wind project (Hywind) across two oil and gas economies, namely Norway and Scotland, the paper sheds light on the key drivers and role of core GPN actors. Methodologically, the authors investigate the process from both ‘inside-out’ and ‘outside-in’ perspectives, referring to the efforts of firms expanding into overseas markets and the efforts of host countries to attract investment from outside their territories. The analysis shows how the configuration of extractive production networks is shaped by the interactions between the network development practices of firms and the market development strategies of host states. The authors conclude that the distinct materiality of floating wind power technology shapes the territorial configuration of the production network by enabling its spatial extension across a range of locations. By contrast, existing research on other extractive sectors has emphasized the spatially constraining effects of materiality (Bridge 2008).  相似文献

While the geopolitical legacies of the World War I peace negotiations are widely recognized, this article examines the often overlooked connection between the WWI Paris Peace Conference's spatial and geopolitical logics and contemporary refugee‐border dynamics. We argue that the spatial and geopolitical logics that framed the WWI Paris Peace Conference—the creation of new states, the propagation of the Western ideal of bounded sovereign states, the nationalist goals of self‐determination and homogeneous ethnic nations, and the establishment of a system of international governance—continue to impact refugee‐border dynamics and “crises” today. The categories, ideals, and practices of the international refugee regime that emerged over the last one‐hundred years stem in great part from these logics. In this paper, we urge critical contemplation about how these foundations—including the establishment of the post of High Commissioner for Refugees in 1921, the resultant Nansen Passports, the post‐WWI minority treaties, and lastly the 1933 Convention Relating to the International Status of Refugees—connect to contemporary human (im)mobility and border violence. We also introduce the articles in this special issue and highlight key themes and future directions for research in critical migration studies.  相似文献

We investigated the bacterioplankton abundance, community composition and the associated Vibrio clades of natural seawater in Bohai Sea coastal waters. Seawater samples (10 L in triplicate) were collected at 0.5, 3, and 5 m depths near the coastal aquaculture zone of the Bohai Sea on May 12, 2016. Real-time PCR and 16S rRNA gene amplicon high-throughput sequencing methods were employed by which 485 operational taxonomic units (OTUs) at a 97% sequence similarity level were generated. Gammaproteobacteria, Alphaproteobacteria, and Bacteroidetes were the most abundant groups, accounting for 49.5%, 23.5%, and 18.9% of the total assemblage, respectively. Obvious variations in Pseudoalteromonas, Vibrio, and Octadecabacter , which were the most abundant genera, could be observed among diff erent samples. Notably, the results of Vibrio -specifi c real-time PCR indicated that Vibrio had extremely high 16S rRNA gene copy numbers. The 16S rRNA gene sequencing results across all the samples also indicated that they occupied a large proportion of the total assemblage. Both the alpha diversity and major bacterioplankton group Pseudoalteromonas had significant correlations with the concentration of PO4^3-. Overall, studies on bacterioplankton communities with highly abundant Vibrio clades can provide interesting insight into the microbial function and health assessment of the Bohai Sea coastal ecosystem.  相似文献

辽河口沉积物中古菌和细菌群落结构分析   总被引:5,自引:1,他引:4  
本研究旨在了解辽河口表层沉积物中古菌和细菌的群落结构组成、多样性及其与环境因子之间的相关性。采用构建古菌和细菌16S rRNA基因文库的方法,应用Illumina Miseq测序技术进行序列分析。结果表明:辽河口表层沉积物中细菌多样性高于古菌多样性,近岸细菌和古菌多样性高于远岸,即河相区(0.8~7.04)细菌多样性高于混合区(13.1~20.7)和海相区(24.2~31.5);主要古菌群落为奇古菌门(Thaumarchaeota,72.73%)和广古菌门(Euryarchaeota,25.05%),其中泉古菌门(Crenarchaeota,0.001%)只在河相区站位被发现;细菌群落组成中变形菌门(Proteobacteria,61.94%)为该河口的优势菌群,其次为拟杆菌门(Bacteroidetes,11.21%)和酸杆菌门(Acidobacteria,5.59%),其他门类如蓝藻门(Cyanobacteria,3.03%)等比例较小。与环境因子的冗余度分析表明:影响表层沉积物中古菌群落分布的主要环境因子依次为氨盐、泥、酸碱度、盐度、电导率和砂,而细菌群落的分布主要受到溶氧、泥、砂、黏土和总磷的影响。由此可见,不同环境条件下微生物的群落结构存在空间异质性,不同微生物对同一环境条件的响应亦不同。  相似文献

海洋蓝洞作为一种独特的海洋地貌单元,洞内的生物群落结构与生态系统特征一直是国内外同行学者关注的热点问题之一。南海三沙永乐龙洞是世界上最深的海洋蓝洞(300m),其独特的生态特征更为引入瞩目。本研究以古菌16S rRNA基因序列为目标,采用Illumina高通量测序技术对西沙永乐龙洞内水体与沉积物中的古菌群落结构进行了研究。结果表明,本研究所获得的古菌类群可归为4门、21纲、29目、42科、45属,广古菌和奇古菌是丰度最高的两种古菌。龙洞内表层水体中古菌群落多样性与洞外水体相差不大,但随深度增加,洞内古菌群落多样性显著降低,深水层古菌群落结构与浅水层差异显著。龙洞内沉积物中的古菌生存环境与水体差异巨大,为大量特殊物种提供了生存空间。侧壁沉积物中的古菌群落多样性随深度增加显著降低,而150m平台处古菌群落多样性则远高于侧壁沉积物。水温对龙洞内水体中的古菌群落结构具有显著影响(P0.01)。研究结果显示永乐龙洞古菌群落有很高的多样性,其垂向分布与洞内环境多要素的变化密切相关。本研究对认识三沙永乐龙洞内的生态系统特征有显著意义。  相似文献

加卸载响应比(Load/Unload Response Ratio,LURR)是根据岩石介质本构关系的动态演化特征提出的地震预测方法。2020年6月26日新疆于田县发生6.4级地震,震前以Benioff应变为响应量观测到明显的LURR高值异常,异常出现于2019年初,持续1年后发生地震,意味着震源区介质的应力状态在2019年发生了明显改变。进一步,以地电场为响应量,计算了距震中280km的和田地电场长、短极距共6个测向的LURR异常时序曲线,结果表明:和田地电场长、短极距的NS向和NW向共4个测向在震前1~3个月出现了LURR异常逐渐增大的同步变化,这种现象可能反映了孕震区介质因微裂隙的产生和扩展导致流体在裂隙中的运移引起的动电效应。综合分析2种不同响应量的LURR异常演化特征,有助于更深入地认知地震孕育过程,更准确地评估地震活动趋势。  相似文献
