Techniques have been developed for the vegetative propagation of Tetraena mongolica,a rare and endangered species endemic to the western Ordos Plateau in the northcentral portion of China.The purpose of the study was to investigate the rooting characteris-tics of the plant species from the aspects of length and ages of cuttings,external hormone concentration,leaf retention,cutting orientations,and cutting positions.The results obtained from the experiments indicated that the rooting ratio of 2-year or 3-year old cuttings was larger than more than of 4-year old cuttings.The influence of external hormone concentration on rooting was ob-vious;the rooting ratio of cuttings was increased by external hormone treatment,especially when treated by ABT1(ABT No.1 Rooting Powder) 50 mg/L for 3 hours,in which case,the rooting ratio reached 91 percent.Leaf retention influenced rooting sig-nificantly;the rooting rate of cuttings became greater as the leaf retention increased.There was no significant difference in rooting capability between south and north orientations of the ortets.The rooting ratio of the cuttings obtained from the distal portions of shoots was significantly larger than those obtained from the middle and the basal portions of the shoots.On day three after treat-ment,calluses started appearing at the incisions of some cuttings dipped in 500 mg/L IAA(indole acetic acid).Most cuttings were healed or swelled,and 6 percent of cuttings rooted on day 6 after treatment.Higher rooting period occurred 7-10 days after the experiment was set up.  相似文献

The solution of second order elliptic partial differential equations typically arising for flow problems, e.g., in porous media, is frequently expressed as face fluxes of a flow field at hexahedral cells. In this letter, we show by example that the following properties are incompatible: (1) A local velocity reconstruction, (2) reproduction of uniform flow for general hexahedral cells, and (3) a flow field in H(div). This is in particular relevant to mixed finite element methods approximating H(div), and to other methods trying to reconstruct a flow field in a hexahedron using only local face fluxes, e.g., streamline tracing methods for reservoir simulation.  相似文献

热脉冲技术是生态生理学和森林水文学研究树木耗水最为常用的技术之一,该技术具有在树木自然生长状态基本不变的情况下实时测量树干木质部上升液流速度及流量,可以简捷准确地确定树冠蒸腾耗水量,同时具有经济可行,测定准确的优点.综述了热脉冲技术的发展历程,对热脉冲方法测定树干液流理论研究、树干液流时空变异性与环境因子的关系研究以及单株耗水向林分水平的尺度转换研究等方面作了较为详细的总结,据此,对热脉冲技术的发展做了展望.  相似文献

采用酸抽提法和Sephadex G-50凝胶层析从鲎血细胞中分离出含有一低分子量组分的提取物,SDS-PAGE显示一条区带。提取物可以抑制内毒素诱导的鲎试剂(TAL)反应,抑制金色葡萄菌和大肠杆菌的生长,并发现25mg/L的提取物对HL-60细胞的生长有强烈的抑制作用。  相似文献

通过控制暂养水体盐度,用盐度突变与盐度渐变2种方法测定长蛸的盐度耐受性.测定了长蛸血细胞密度、各类血细胞数量比例以及各种酶的活力几组数据,并分析其血细胞与体内酶活力的变化.结果表明:长蛸的生存盐度范围为7.0~30.3,适宜盐度范围为16.3~27.3,最适盐度范围为18.3~24.3,对盐度的适应范围较广,有利于长蛸大规模养殖的开展.在盐度胁迫下,长蛸血细胞密度,各种血细胞数量比例均发生显著改变,长蛸个体也随着胁迫加大变得越来越不适应,出现喷墨、休克、甚至死亡,这表明长蛸的免疫机能降低.无论在低盐度还是高盐度胁迫情况下,受渗透压的影响,长蛸肌体大量吸水和失水,呼吸作用减弱,体内供氧降低,导致长蛸体内有氧呼吸降低和无氧呼吸提高,于是催化有氧呼吸的LDH酶活力降低.同时,细胞内与免疫和消化相关的细胞器由于细胞吸水或失水作用功能受到影响,保护酶(SOD、POD、CAT)、磷酸酶(ACP、AKP)和消化酶(蛋白酶和脂肪酶)的活力均呈下降趋势.由此可见,盐度胁迫对长蛸的影响是显著的.  相似文献

通过组织学连续切片与扫描电镜观察,研究凡纳滨对虾的性别分化。结果显示:凡纳滨对虾外部形态的性别分化最早发生在仔虾后第37天,此时在扫描电镜下可观察到雄性附肢的芽基,仔虾后第45天,光镜下可观察到交接器;仔虾后第24天出现原始生殖细胞(PGCS),位于背大动脉的两侧及围心腔膜的腹面,细胞个体及细胞核明显比周围的细胞大,具有强嗜碱性,细胞核圆形或椭圆形,核内染色质清晰可见,随着生殖腺的发育,在第43天出现生殖管,第55天生殖腺可区分为精巢和卵巢。  相似文献

采用饲养对比试验方法,研究了谷氨酰胺二肽(Gin二肽)对日本对虾血清生化指标、肝胰腺细胞凋亡和肠绒毛的影响及对虾体健康的作用。结果表明,饲喂Gin二肽各组的PO、LSZ、ACP含量均显著(P〈0.05)高于对照组,添加0.5%和1.0%Gin二肽组的SOD和ALP含量也显著(P〈0.05)高于对照组:添加1.0%Gin二肽的TP含量显著(P〈0.05)高于其它各组。血清TP、PA、UN、CHO、PO、LSZ、SOD、ALP、ACP和LDH等各项指标均随Gin二肽添加量的提高而呈现增加趋势。试验建立了流式细胞术检测对虾肝胰腺细胞凋亡率的方法;肝胰腺细胞凋亡率随着日粮Gin二肽添加量的增加而显著降低(P〈0.05)。肠道切片显微观测结果表明,添加1.0%的Gin二肽能显著增加日本对虾肠绒毛高度(P〈0.05)。饲料中添加Gin二肽可显著提高日本对虾血清中的溶菌酶、抗氧化酶及磷酸酶活性,降低肝胰腺细胞凋亡率,增加肠绒毛高度,改善虾体健康状况。  相似文献

We extracted marine low-temperature lysozyme (MLTL), a novel lysozyme, from a marine microorganism through fermentation. Our previous study suggested that a low molecular weight (16 kDa) may exert anti-tumor activity through antiangiogenesis. In this study, we extracted a high weight (39 kDa) and investigated its antiangiogenic activity in vivo and in vitro. Using zebrafish embryos as an in vivo study model, we found that treatment with MLTL significantly inhibited the growth of subintestinal vessels (SIVs) in a dose-dependent manner and that 400 μg/ml MLTL was sufficient to block the growth of SIVs. An in vitro study conducted using human umbilical vein endothelial cells (HUVECs) revealed that MLTL suppressed the proliferation, migration and tube formation of HUVECs in a dose-dependent manner. Interestingly, assays by flow cytometry and DNA electrophoresis indicated that MLTL was able to induce apoptosis of HUVECs. Moreover, further study demonstrated that the disruption of intracellular Ca2+ homeostasis may play an important role in MLTL induced apoptosis of HUVECs. Taken together, the results of this study demonstrate for the first time that MLTL inhibits angiogenesis through its pleiotropic effects on vascular endothelial cells and induces apoptosis through regulation of cellular Ca2+ levels. The results of this study also revealed a possible mechanism underlying the antiangiogenic effect of MLTL and suggested that MLTL may be a promising new antiangiogenic agent for use in cancer therapy.  相似文献

为了探明姬松茸提取物的体外抗病毒感染活性及其作用方式,进而为水产养殖鱼类高效抗淋巴囊肿病毒(lymphocystis disease virus,LCDV)感染活性物质的开发和鲆蝶类淋巴囊肿病的防治奠定基础,利用热水浸提和酒精沉淀法得到了5种姬松茸提取物组分(E1~5),并利用MTT、细胞病变程度观察等方法研究了5种组分对LCDV感染体外培养大菱鲆鳍细胞(turbot fin cells,TF细胞)的影响作用.细胞毒性实验结果显示,5种组分对体外培养TF细胞均无毒性.细胞病变程度结果表明,本文所得5种姬松茸提取物组分尤其是E5组分具有显著的抗LCDV感染TF细胞的活性.不同方式感染的实验结果进一步显示,5种姬松茸提取物组分抗LCDV感染TF细胞的作用可能主要是通过直接灭活病毒和/或阻断病毒吸附细胞来实现的.  相似文献

Timing of vegetal-endodermal cell determination in amphioxus embryos remains uncertain. We tentatively tested effects of A23187, the calcium ionophore, on the development of vegetal blastomeres isolated at the 16-cell stage. It was found that when vegetal blastomeres committed to endoderm were treated with A23187 prior to gastrulation, they were transformed into ectodermal cells as evidenced by the cell morphology and function characteristic of epidermis. However, the developmental fate of the same blastomeres untreated or treated with DMSO at the same stage or of those treated with A23187 after gastrulation remained unchanged. Thus, vegetal-endodermal cells in amphioxus embryos are not irreversibly determined before the gastrula stage, and artificial increase in intracelluar Ca2+ concentration can induce transdetermination of the predetermined endodermal cells into ectodermal cells.
