The genetic structure of populations of the sea cucumber Stichopus (Apostichopus) japonicus was investigated using 10 microsatellite markers. In all, 152 individuals from five natural populations were collected from Aomori, Japan (JA and JR), Yosu, South Korea (KY), Dalian, China (CD) and Vladivostock, Russia (RV). A total of 145 alleles were found at 10 loci. The number of alleles per locus ranged from 9 at PSC03 to 20 at SCZ06, with an average of 14.5. Average Ho and He ranged from 0.260 (JR) to 0.434 (JA) and from 0.654 (RV) to 0.778 (KY), respectively. No significant differences at A, Ho and He were found, indicating similar genetic diversity in the five populations. A single allele was found at the PSC05 locus in the RV population. Of the 50 loci, 42 significantly deviated from Hardy–Weinberg equilibrium, all showing heterozygote deficiency. The genetic distances were all relatively great, ranging from 0.497 (between JA and KY) to 1.029 (between KY and JR). This suggests the five populations are genetically distinct. Cluster analysis indicated that JA, KY and CD form one branch and RV and JR another in the UPGMA tree. A hypothesis is proposed for the evolution of the Japanese red sea cucumbers and the genetic relationship among the populations.  相似文献

A complementary DNA (cDNA) library was constructed from half-smooth tongue sole spleen. A long-read expressed sequence tag (EST) database was generated, containing 3100 cDNA clones, of which 220 clones were fully sequenced. A total of 1060 non-redundant simple sequence repeats (SSRs) were obtained from the cDNA library. An average of 5 kb sequence generates 1 SSR in the half-smooth tongue sole spleen cDNA library. The proportion of the SSR unit size was different in the cDNA library. The monomeric repeats (51.4%) are the most abundant class of SSR in the dataset. The dimeric, trimeric, tetrameric and hexameric repeats are represented in decreasing proportions of 27.2%, 16.0%, 2.8% and 1.9%, respectively. The frequency of pentameric repeats was observed the least (only 0.7%). Most of the monomeric and dimeric repeats are distributed in 3′ and 5′ un-translation region. If translation regions are considered merely, trimeric repeats are the highest, accounting for 57% of the total microsatellites.  相似文献

The golden cuttlefish, Sepia esculenta Hoyle, 1885 (Cephalopoda: Sepiidae) is a valuable and important fishery resource for China, Japan and Korea. This fishery has experienced severe population decline largely due to overexploitation in past decades. To provide guidelines for fisheries management, we estimated genetic diversity and population structure across four locations along the coast of Japan and one location in China (a total 281 individuals) using nine microsatellite DNA loci. Sepia esculenta showed high genetic variability with mean allele richness ranging from 15.8 to 21.4, and mean heterozygosity from 0.80 to 0.90. Weak but significant genetic differentiation was present and the FST value was 0.020 across all five populations. The useful information obtained in this study will offer insights into how to fine‐tune conservation and fishery management measures for this species and resource in the future.  相似文献

为分析湛江流沙湾海域优势渔种卵鳎的遗传多样性,应用微卫星标记技术,选用15对微卫星引物,以等位基因数、基因杂合度、多态信息含量、固定指数等遗传参数为指标,评估卵鳎群体内的遗传多态性。结果表明:共检测到90个等位基因,等位基因数从1~12不等,平均为6.0;有效等位基因数从1.0~8.4,平均为4.0,多态性位点比例为53%,显示其具有中等杂合子水平,其中8个多态位点的期望杂合度(He)为0.670~0.881,平均为0.800,观测杂合度(Ho)为0.353~1.000,平均为0.773,多态信息含量(PIC)值为0.616~0.870,平均为0.773,群体内固定指数F为-0.199~0.564,平均为0.046;流沙湾卵鳎群体具有高度遗传多样性。  相似文献

自坛紫菜(Porphyrahaitanensis)丝状体中提取的DNA,经Sau3AI限制性内切酶消化后,将300~900bp之间的DNA片段回收,并连接到经BamHI酶切并去磷酸化的pUC18载体上,最后转化至大肠杆菌JM109感受态细胞中,构建坛紫菜小片段DNA质粒文库。选用pUC18质粒的通用引物进行PCR反应,对文库进一步筛选并检测插入片段的大小,在384个阳性克隆中,有278个含有大小合适的插入片段。经测序及序列分析,在103个克隆中获得172个微卫星序列,其中完美型107个,占62.2%,非完美型53个,占30.8%。(GC)n与(CG)n在坛紫菜DNA中非常丰富,分别占25%及17%,但重复频率相对较低。  相似文献

采用12个多态性微卫星标记对方斑东风螺(Babylonia areolata)泰国选育系TF4~TF6连续3代选育群体的遗传多样性与遗传结构进行了分析。3个群体的多态信息含量分别为0.730、0.717和0.708,均高于0.5,表现出较高的多态性。平均等位基因数(A)、有效等位基因数(Ae)、观测杂合度(Ho)、期望杂合度(He)和Shannon’s信息指数(I)的变化范围分别为11.667~12.583、6.334~6.915、0.658~0.672、0.737~0.787和1.837~2.003,整体呈降低趋势,但差异并不显著(P>0.05)。分子方差分析结果显示,群体间的遗传变异仅占总变异的0.95%;群体间的遗传分化系数(Fst)介于0.00569~0.01324,处于低等分化水平(Fst<0.05)。主坐标分析和STRUCTURE分析结果显示,3个群体呈现出相似的遗传背景和遗传结构。以上研究结果表明,经过连续多代的人工选育,方斑东风螺选育群体保持着较高的遗传多样性水平,遗传变异和遗传分化水平较低,遗传结构趋于稳定,仍具有较高的遗传选育潜力。  相似文献

选取8个高多态性的微卫星位点, 对240只刺参(Apostichopus japonicus)幼体和14只疑似亲本进行了亲缘关系的鉴定。通过检测, 8个微卫星位点共得到53个等位基因, 平均等位基因数为6.63; 观察杂合度在0.730—0.902; 各位点的多态信息含量为0.696—0.836。通过亲缘关系分析软件CERVUS 3.0分析, 微卫星位点累积排除概率分别为0.9934、0.9997、0.99999。基于排除法个体基因型判定, 有37个子代有1个候选亲本, 194个子代有2个及以上候选亲本; 基于似然法判定, 置信度为80%时, 有198个子代个体找到最似亲本。综合排除法和似然法的亲权鉴定, 成功为229个子代个体找到它们的亲本, 鉴定成功率达95%以上。本研究结果可为刺参育种工作中亲本识别及家系分析提供技术支持和理论依据。  相似文献

Movements of humpback whales Megaptera novaeangliae among breeding regions within the southwestern Indian Ocean are poorly understood. Understanding the relationships among breeding regions is critical for effective conservation and management strategies. Through systematic comparisons of molecular genotypes and both systematic and non-systematic comparisons of individual identification photographs collected between 1996 and 2006, we have thus far identified nine whales (six males and three females) utilising two breeding areas within this region: the northern Mozambique Channel and eastern Madagascar. Four of the nine whales were recaptured using only photographic data, two whales were independently recaptured using both photographic and genetic data, and three whales were recaptured exclusively using molecular methods. The discovery of these nine individuals provides much-needed data to guide the formulation and future revision of stock boundaries.  相似文献

从GenBank上选取40对尼罗罗非鱼(Oreochromis niloticus)的微卫星引物,分别在奥利亚罗非鱼(Oreo-chromis aureus)(83系)、红色奥利亚罗非鱼、奥利亚罗非鱼(02系)和尼罗罗非鱼的基因组上进行扩增。40对引物中有37对(92.5%)可进行有效扩增,32对引物(80%)可检测到个体间等位基因的多态性。其中引物UNH168与奥利亚罗非鱼的性别相关,在雌性个体中可扩增出二条大小不同的特异带(分别为135和171 bp),在雄性个体中则只有一条(171 bp)。将特异条带回收、克隆并测序,结果显示雌雄个体中171 bp条带的序列完全相同,包括103bp的侧翼序列和34个CA重复,在雌性个体中获得的135 bp条带则只有16个CA重复。引物UNH846、UNH860和UNH995可鉴别奥利亚罗非鱼和尼罗罗非鱼,UNH890可鉴别出红色奥利亚罗非鱼。可见,大部分的尼罗罗非鱼微卫星位点存在于奥利亚罗非鱼中。  相似文献
