Sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) was applied in the detection of the end point temperature (EPT) of thermal denatured protein in fish and meat in this study. It was also used in studying the thermal denatured temperature range of proteins in salmon and chicken meat. The molecular distribution of dissolved proteins in seawater from coastal marine environments in Uranouchi Bay, Kochi Prefecture, is first reported in this article. Occurrence of bacteria-derived dissolved proteins and their source bacteria were examined using a probe of the antibody (anti-Omp35La) against a porin outer membrane protein (Omp35La) of the fish pathogenic bacterium Vibrio (Listonella) anguillarum. The electrophoretograms of dissolved proteins from coastal seawater showed a large number of discrete and individual proteins overlapped each other over a wide range of molecular masses indicating active processes in coastal environments in transferring proteins from organisms to the inanimate dissolved protein pool. Among the dissolved proteins, 37 kDa- and 18 kDa-proteins reacted with the Omp35La. In order to isolate the source bacteria of such dissolved proteins, bacteria from seawater and diseased fish were screened by colony Western blotting with anti-Omp35La. The reactive strains were further examined in sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE)/Western blotting to verify the presence of Omp35La homologues among the outer membrane proteins of such strains. Outer membrane proteins reacting with anti-Omp35La were detected in only 4 strains of the 129 strains that were positive in the colony Western blotting. The level of possible source bacteria of 37 kDa- and 18 kDa-dissolved proteins was suggested to be 5–6 orders of magnitude lower than the total bacterial count. The present study leads us to hypothesize that a minor portion of the bacterial assemblage is responsible for the dissolved proteins in the coastal waters.  相似文献

不同保存方法对厚壳贻贝样品蛋白提取效果的比较   总被引:1,自引:0,他引:1  
为了解厚壳贻贝(Mytilus coruscus)稚贝样品在冷冻保存过程中的蛋白变化情况,作者采用直接冷冻法、添加海水冷冻法、蛋白裂解液冷冻法等分别对保存1周至6个月的厚壳贻贝样品蛋白提取的不同效果进行了比较。SDS-PAGE胶结果显示,3种冷冻保存方法均出现4个有规律条带(a、b、c、d),其相对分子量依次约为55、46、38和26kDa。SDS-PAGE蛋白条带灰度值的检测结果表明,1个月内蛋白裂解液法保存的样品最佳;直接冷冻法和添加海水冷冻法的研究发现保存过程中的蛋白均出现不同程度的降解现象,因而可能不适宜用于该种稚贝样品的保存。  相似文献

采集采自山东某养殖场的栉孔扇贝(Chlamys farreri)健康贝和病贝的血淋巴,测定其血浆蛋白浓度、血浆蛋白图谱和相对分子质量,以及血浆抗菌活性、凝集素活性等几种重要的体液免疫因素,研究环境胁迫对血浆蛋白及其抗菌活性的影响。结果表明:栉孔扇贝(C.farreri)血浆蛋白质量浓度约1 260~1 650μg/mL;血浆蛋白SDS-PAGE图谱显示3条蛋白带,相对分子质量分别为34 000、25 000和14 000;健康贝血浆抗菌活性指数BI为0.34,病贝BI为0.21;凝集素活力水平为1.602~2.204。氨氮胁迫下血浆蛋白浓度下降约780μg/mL,凝集素活力下降为1.301;高温胁迫下血浆蛋白质量浓度下降约为900μg/mL,凝集素活力为1.903;高盐胁迫下血浆蛋白浓度下降约为990μg/mL,凝集素活力为2.204。  相似文献

<正>目前,针对传统抗生素治疗细菌感染中出现的耐药性及药物残留等问题,亟需寻找一种新型的替代药物。噬菌体对治疗病原菌感染的多种优越性,成为抗菌新药研究开发的新热点和理想材料。对噬菌体的溶菌机制[1-3]以及实践应用[4-6]等已有较多报道。笔者主要对副溶血弧菌噬菌体衣壳蛋白的组成进行了初步研究,并用SDS-PAGE电泳法分离纯化了其主要组分,为制备该蛋白质的抗体,建立副溶血弧菌噬菌体的免疫学检测方法奠定基础。  相似文献

于1998年3月在青岛市太平角采集裙带菜孢子体,通过破碎、离心制备类囊体膜,用去污剂SDS、DOC、DMG和DIG增溶裙带菜类囊体膜,经PAGE分离色素-蛋白质复合物,并测定其光谱特性,结果表明,电泳分离效果与所用的去污剂的种类有关。去污剂不同,分离到的同一类型复合物的吸收光谱有差异,捕光色素-蛋白质复合物的数目、种类及吸收光谱明显不同,电泳中产生的游离色素的多少也随去污剂种类而异,在4种去污剂中,以光谱明显不同,电泳中产生的游离色素的多少也随去污剂种类而异,在4种去污剂中,以DMG和DIG增溶的效果最好,经SDS-PAGE分出6种色素-蛋白质复合物,游离色素较少,DMG和DIG增溶的效果最好,经SDS-PAGE分出6种色素-蛋白质复合物,游离色素较少。  相似文献

Two-dimensional electrophoresis(2-DE) of protein extracted and purified from Alexandrium sp. LC3 was conducted. In the SDS-PAGE study, the relative molecular weights of the proteins were mainly in the range of 14 kDa?31 kDa and 43 kDa?66 kDa, and more proteins were detected between 14 kDa and 31 kDa. With the improved protein preparation, the two-dimensional electrophoresis patterns indicated that the relative molecular weights of the proteins were between 14 kDa and 100 kDa, and most of them ranged from 14 kDa to 31 kDa. This was consistent with the result of the SDS-PAGE analysis. The isoelectric points were found to lie between 3.0 and 8.0, and most of them were in the range of 3.0?6.0. Better separation effect was acquired with pre-prepared immobilized gradient (IPG) strip (pH 3?5.6), and about 320 protein spots could be visualized on the 2-DE map by staining. Within pH 3?10 and pH 3?5.6 strips, the protein samples of Alexandrium sp. LC3 could be separated well.  相似文献

采用5 mg/kg 17β-雌二醇对大菱鲆(Scophthalmus maximus)进行肌肉注射,一周后尾静脉取血,离心分离获得血浆,经Sephacryl S-300(高分辨)分子筛纯化卵黄原蛋白,对卵黄原蛋白进行常规聚丙烯酰胺凝胶电泳,考马斯亮蓝染色和糖、磷、脂的特征性基团染色.结果表明,17β-雌二醇能诱导大菱鲆产生卵黄原蛋白,以N-ative-PAGE方法计算得到大菱鲆卵黄原蛋白的分子质量为538 ku,以SDS-PAGE方法得出其两种亚基的分子质量分别为100 ku和82 ku.分子筛凝胶过滤能较好地纯化大菱鲆血浆中的卵黄原蛋白.  相似文献

采用SDS聚丙烯酰胺凝胶电泳方法,对锯缘青蟹肌肉中可溶性蛋白质进行分离,以过敏患者阳性血清和正常人阴性血清作对照,通过Western blot鉴定出分子量为14.3kDa的蛋白为锯缘青蟹的特异性过敏原.然后用Sephadex G75凝胶过滤层析和DEAE-52离子交换层析纯化出该过敏原,再经MALDI-TOF/TOF-MS检测,利用Blastz在线分析.结果表明,锯缘青蟹特异性过敏原为血蓝蛋白亚基,经Clustal W分析该蛋白的多肽片段序列与可口美青蟹、邓杰内斯蟹、加州龙虾、艾氏真蟹的血蓝蛋白亚基序列有很高的同源性,经Prosite数据库检索后发现肽段1和肽段2有酪蛋白激酶Ⅱ磷酸化位点和蛋白激酶C磷酸化位点,是过敏原参与机体反应的重要活性位点.  相似文献
