The authors investigated the effect of Bacillus mucilaginosus on weathering of phosphorite. Analysis of different proteins was of significance in exploring the molecular biological mechanism in the bacterial weathering process. The concrete methods are described as follows: Mineral powder was put into liquid culture medium and B. mucilaginosus was incubated in the medium. The control (group) had no mineral powder in the medium. The treatments and controls were cultured simultaneously under the same condition. In a few days, the supernatant was filtrated, the main cations (Ca^2+, Mg^2+, Na^+, Mn^2+, Al^3+, Fe^3+, K^+) were measured by ICP-OES, and the contents of water soluble phosphorus (Pws) and silicon (Siws) were determined by colorimetry. The residual solid was weighed on the filter paper, followed by digestion with concentrated HNO3. The concentrations of the main cations and Pws, Siws in the digest liquid were measured by using the method mentioned above. After the supernatant was centrifuged, the precipitation was used to analyze the protein differences between the treatment groups and the control groups by 2-dimentional gel electrophoresis (2-DE). The experimental results showed that apatite and quartz were partially weathered, but kaolinite was dissolved completely. The population of bacteria increased when mineral powder was added in the liquid medium. Software analysis and comparison of the 2-DE pictures of bacterial proteins revealed 1134 visible protein spots in the treatment group, and 729 visible protein spots in the control group. To compare the bacterial protein expression contents of the treatment group with those of the control group, there were 496 different protein spots, including 214 protein spots which indicated that the protein contents increased, 75 protein spots were indicative of a decrease, and 207 proteins were newly synthesized. 从软体动物中的腹足纲、双壳纲以及节肢动物中的甲壳纲分别选取了3种中国沿海常见的品种——东风螺、弯竹蛏、斑节对虾,提取并分析它们的壳基质蛋白。利用液相色谱串联质谱(LC-MS/MS)技术对蛋白质组学进行分析发现特定的蛋白质具有明显的种属差异:①N66蛋白(一种碳酸酐酶)通常存在于弯竹蛏中,但不存在于东风螺和斑节对虾中,揭示N66蛋白质具有特异性,在未来的物种鉴定中可能具有重要的参考意义;②角质层蛋白AM1199和CP14.6存在于斑节对虾的表皮结构中,在东风螺和弯竹蛏中未检测到;③血蓝蛋白同时在3种动物外壳的基质蛋白中被鉴定出来,揭示了其在不同物种的外壳形成与发育过程中的重要作用。该研究为壳基质蛋白在软体动物或节肢动物外壳中的鉴定提供了基础,也为探索不同物种的生物矿化过程提供了重要参考。  相似文献

In March 2011, the Ecophysiology and Genetics Working Groups of the European Science Foundation COST Action ES 0906, entitled Seagrass Productivity: From Genes to Ecosystem Management, organized an exploratory workshop entitled “Linking Ecophysiology and Ecogenomics in Seagrass Systems”. The goal of the workshop was to discuss how to integrate comparative gene expression studies with physiological processes such as photosynthetic performance, carbon and nitrogen utilization and environmental adaptation. The main questions discussed for integrative research related to mechanisms of carbon utilization, light requirements, temperature effects and natural variation in pH and ocean acidification. It was concluded that the seagrass research community is still in the nascent stages of linking ecophysiology with genomic responses, as carbon and nitrogen metabolism of seagrasses have not been sufficiently well studied. Likewise, genomic approaches have only been able to assign meaningful interpretations to a handful of differentially expressed genes. Nevertheless, the way forward has been established.  相似文献

建立了西施舌(Coelomactra antiquata)外套膜蛋白质组的双向凝胶电泳体系,比较分析了不同pH梯度胶条、水化条件的优化及不同染色方法对双向电泳结果的影响.结果表明:用pH 4~7的非线性胶条,水化时加上除盐步骤,采用硝酸银染色,即可获得高分辨率、高重现性的双向电泳图谱,为进一步研究西施舌外套膜蛋白质的组成奠定了基础.  相似文献

The methods of proteomics, the study of the protein complement of the genome, are applicable to environmental testing. Sets of proteins specific to different stressors can be isolated using computer imaging software. Individual proteins can be identified by mass spectrometry. The Protein Expression Signatures (PES) obtained have potential in diagnosing adverse factors in the environment. The challenge is to demonstrate their feasibility in complex environments. We have shown that PES for three endocrine disrupting compounds in trout (Onchorhynchus mykiss), can be detected in mixed sewage effluent. Other studies support these results. As protein databases expand, identification becomes routine, and capture molecules specific to each protein are developed, the possibility of simple field tests for multiple stressors becomes real.  相似文献

海带(Saccharinajaponica)是我国的主要经济褐藻,从海带中制备的褐藻多糖硫酸酯具有免疫调节等多种生物活性,但鲜有基于组学的研究报道。本文采用RNA高通量测序(RNA-sequencing,RNA-seq)转录组学技术以及平行反应监测(ParallelReactionMonitoring,PRM)靶向定量蛋白组学方法,探究海带褐藻多糖硫酸酯作用于小鼠巨噬细胞RAW 264.7后对巨噬细胞功能和通路的影响。结果表明,褐藻多糖硫酸酯(100μg/mL)作用于RAW 264.7细胞后,差异表达基因主要富集在免疫反应、先天性免疫应答等生物过程以及TNF、NF-κB、Toll样受体等信号通路,也可促进RAW 264.7细胞膜蛋白A类清道夫受体(SR-A)中SCARA3和Toll样受体中TLR4的表达。采用流式细胞仪对褐藻多糖硫酸酯作用后SR-A和TLR4的表达量进行蛋白水平的验证, SR-A和TLR4的表达量均有升高,证实了组学结果的可靠性。本研究结果为利用褐藻多糖硫酸酯开发具有免疫调节作用的功能食品或药物提供依据。  相似文献

光温敏核不育水稻幼穗育性转换期蛋白质组的初步分析   总被引:2,自引:0,他引:2  
为找寻光温敏核不育水稻幼穗与育性调控有关的蛋白质,采用固相pH梯度-SDS聚丙烯酰胺双向凝胶电泳对光温敏核不育水稻(培矮64S)育性转换期幼穗总蛋白进行了分离,获取凝胶图像并进行重复性分析,通过微量测序-Edman降解方法和原位酶解及MALDI-TOF质谱的方法鉴定了60个蛋白质点。结果表明:本方法可得取分辨率和重复性较好的凝胶图谱。蛋白质点在2D胶上的重复性为:沿等电聚焦方向偏差为1.49±0.22 mm,沿SDS-PAGE方向偏差为1.24±0.18 mm。37个蛋白质点得到了准确的鉴定结果,其中16个是差异蛋白质点。在不育变化为可育的过程中,明显表达上调的蛋白质点包括几丁质酶,丙酮酸脱氢酶等;明显下调的蛋白质包括超氧化物歧化酶,硝酸还原酶脱辅基酶蛋白等;金属硫蛋白RicMT只在可育中存在。  相似文献

Man-made polybrominated diphenyl ethers (PBDEs) used as flame retardants in various consumer products may be harmful to marine organisms. Larvae of some marine invertebrates, especially invasive species, can develop resistance to PBDEs through altered protein expression patterns or proteome plasticity. This is the first report of a proteomics approach to study BDE-47 induced molecular changes in the invasive limpet Crepidula onyx. Larvae of C. onyx were cultured for 5 days (hatching to metamorphosis) in the presence of BDE-47 (1 μg L−1). Using a 2-DE proteomics approach with triple quadrupole and high-resolution TOF-MS, we showed that BDE-47 altered the proteome structure but not the growth or metamorphosis of C. onyx larvae. We found eight significant differentially expressed proteins in response to BDE-47, deemed the protein expression signature, consisting of cytoskeletal, stress tolerance, metabolism and energy production related proteins. Our data suggest C. onyx larvae have adequate proteome plasticity to tolerate BDE-47 toxicity.  相似文献

比较了裂解液法和直接三氯乙酸(TCA)-丙酮沉淀法对中肋骨条藻Skeletonema costatum 蛋白双向电泳的提取效果并优化了提取条件,结果表明:裂解液-TCA-丙酮沉淀法在去除胞内干扰物质方面,较裂解液-超滤管法和裂解液-试剂盒法的效果都好。 裂解液-TCA-丙酮沉淀法和直接TCA-丙酮沉淀法都能取得背景干净、蛋白点清晰的双向电泳图谱,但后者的双向电泳图谱聚焦更完全,在进一步优化条件后(即蛋白沉淀12 h并增加超声波洗涤过程),可作为提取中肋骨条藻蛋白的最适方法。  相似文献
