于2004年利用3个品系的雨生红球藻(H0、H2和H3)和以去除营养物质的藻液为实验对象,探讨了光强对细胞转化和虾青素积累的影响。结果表明,光照强度对红球藻虾青素积累和细胞转化有显著影响,并在品系间存在一定差异性。其中:品系H0在光强≤25000lx时虾青素积累量随光强的增加而增加,光强在20000~35000lx时虾青素积累量处于较高水平,而光照强度>35000lx时,虾青素积累量随光强的升高而下降;H2品系在光强≤25000lx时虾青素积累量随光强的增加而增加,光强在25000lx左右时虾青素积累量较高,当光强≥35000lx时,虾青素积累大幅度下降;H3品系在≤35000lx时虾青素积累量随光强的升高而增加,20000~45000lx为虾青素积累较适宜的光强,高于45000lx时虾青素积累量则明显下降。进一步分析得出,弱光导致红球藻虾青素含量较低是光照强度不足、细胞内虾青素含量少的结果,而强光下虾青素含量下降则是细胞受到损伤、细胞数量减少导致的。对比研究发现:在较低光强下(20000~25000lx)H2单个细胞内虾青素积累量略低于H0,但其积累虾青素总量却最高,这是由于此时H2不动细胞增长速率比其它品系高造成的,不过适宜H2积累虾青素的光照范围最窄,它对高光强的耐受性也最差;而H3在较低光强下虾青素积累总量及单个细胞内的积累速率均最低,但是它的适宜光照范围最宽,即对高光强的耐受性最强,在较大的光强波动范围内都可以保证较高的安全转化量,这就使得它在较高的光强下虽然单个细胞积累虾青素的速率仍然最低,积累总量反而显著高于其它两个品系。  相似文献

研究了光照强度2 500~20 000 lx对孔石莼(Ulva pertusa)生长和藻体生化组成的影响。研究结果表明,光照强度对孔石莼的生长率具有明显的影响(P0.05),孔石莼在光照强度为12 500 lx时有最大的生物量积累。光照强度低于12 500 lx时,孔石莼的生长率有随着光照强度的降低而减小的趋势;光照强度高于12 500 lx时,孔石莼的生长率随着光照强度的升高而降低(P0.05)。不同光照强度对孔石莼的叶绿素、可溶性糖和游离脯氨酸的含量具有明显的影响(P0.05)。随着光照强度的增加,孔石莼的叶绿素a和叶绿素b的含量逐渐变小;低光照条件也促进了孔石莼蛋白质的积累;当光照强度超过17 500 lx时,游离脯氨酸的含量明显增加。光照强度对孔石莼化学组成也具有明显的影响(P0.05)。随着光照强度的升高,藻体的N元素含量有所升高,但光强达到17 500 lx后,随着光强的增加,N元素含量反而降低;C元素的含量在光照强度达到17 500 lx后,含量也明显升高;H元素的含量在光照强度2 500~20 000 lx范围内,具有随着光照强度增加而增加的趋势。  相似文献

于1989年1月—1989年8月采用连续培养和半连续培养方法进行了伪矮海链藻细胞分裂、叶绿素α含量和活体荧光特性与光、营养盐关系的研究。结果表明,细胞分裂、活体荧光、叶绿素α均呈现光照期的增长速率明显高于黑暗期的增长速率的日变化规律,荧光增强比则在光照期开始后或黑暗期结束时出现最高值;光强和营养盐不仅影响各指标日变化的幅度,而且还可改变荧光增强比峰值出现时间。因此,在研究细胞分裂、叶绿素α和荧光特性的昼夜节律时,必须考虑光和营养盐这两个重要因素。  相似文献

国内外关于好氧不产氧光合异养细菌(AAPB)和上升流之间关系的研究甚少。本文采用“基于蓝细菌校正的时序红外显微技术”研究了台湾海峡南部近岸上升流中心区AAPB对上升流变化的响应。研究结果发现,在上升流涌升的初始阶段,AAPB和总异养细菌丰度较低;随着上升流的发展,两者丰度均增加并在上升流的成熟期达到最高值;而当上升流衰退时,AAPB和总异养细菌丰度开始下降。在上升流发展过程中,AAPB丰度与叶绿素a浓度在一定范围内呈显著正相关,但同时受环境低磷浓度的限制,总异养细菌丰度与氮、磷、硅营养盐均有显著正相关,表明叶绿素a指示的浮游植物所释放的溶解有机碳和环境中的磷限制可能对AAPB起着更为直接和重要的作用,而营养盐则可能在总异养细菌对上升流的响应中起着重要作用。本研究有助于我们理解AAPB在碳及其他生源要素循环中的作用及其调控机制。  相似文献

海洋亚心形扁藻(Platymonas subcordiformis)在光照生长后,经过暗培养诱导其可逆产氢酶,然后转移到光照下可产生氢气。在300 mL产氢反应器中对海洋亚心形扁藻产氢情况的考察表明,不同的光强条件可导致扁藻产生不同的生长曲线,而在不同的培养阶段,扁藻的产氢能力也有很大的差别。其他培养条件相同,高光照强度(E=13000 lx)条件下,扁藻生长较快,而且可以获得更大的产氢能力,300 mL密度为3×106个/mL、叶绿素质量浓度为5.8 mg/L的藻液最大产氢浓度可达17.5%,比低光照强度(E=5000 lx)时提高了32%,叶绿素含量降低了61%。根据产氢前后以及暗诱导后扁藻生理状态的考察结果,可以推测出是不同生长阶段扁藻的生理状态差异导致了其产氢能力的变化。  相似文献

本文对卡德藻在自养、异养和兼养培养条件下的比生长速率、细胞密度、细胞色素组成及脂肪酸组成等方面进行了比较研究。兼养卡德藻的细胞密度大于自养条件和异养条件下细胞密度之和。兼养培养的卡德藻比生长速率是自养的 2倍 ,异养的 1.3倍。兼养和异养生长的对数期较自养的长。光合自养培养卡德藻最适光强为 10 0 0 0 lx,而兼养培养最适光强范围为 10 0 0~ 2 0 0 0 lx。在自养和兼养培养时亚油酸的含量很高 ,自养和兼养状态下分别为 16 .6 %和 17.4 9%。 EPA合成的量较低。异养藻的多不饱和脂肪酸的产量较自养藻和兼养藻要低。兼养藻细胞光合色素组成和含量与自养藻细胞的基本一致 ,但在异养条件下藻细胞色素组成发生明显改变。  相似文献

采用控制光照强度的方法,研究了不同光照强度对白色霞水母无性繁殖的影响。结果表明,400lx组螅状体的相对增长率、特定生长率和螅状体足囊、囊胞繁殖率均显著高于其它各试验组(P<0.05),呈现出400lx>800lx>1200lx>0lx的趋势。在设定的光照强度范围内,螅状体足囊、囊胞繁殖出现的时间随光照强度的减少而延迟。800lx组的足囊萌发率最高,但各试验组足囊萌发率之间无显著性差异(P>0.05)。试验期间始终是400lx组螅状体的横裂率最高,试验结束时,0lx组螅状体的横裂率最低。试验结果说明,适当的光照度对白色霞水母螅状体生长、螅状体足囊、囊胞繁殖和横裂生殖均有促进作用,强光条件或完全无光照条件不适宜白色霞水母螅状体的生长并可使横裂生殖延迟或受到抑制。  相似文献

The detection of light intensity and subsequent phototaxis are essential for the fitness of sea urchins in intertidal and shallow subtidal waters, where light intensity varies in accordance with the depth and other hydrographic conditions. The molecular basis of photoresponse, however, remains largely unknown. We compared the expression of SiOpsin4, SiPax6, and SiOpsin5 among tube feet, coelomocytes, gonads, and gut of the sea urchin Strongylocentrotus intermedius. All three genes showed the significantly highest expression in tube feet. To test whether r-opsin genes (for example, Opsin4) and Pax6 in tube feet, rather than chaopsin genes (for example, Opsin5), are involved in the molecular responses to light intensity, we investigated the expression of Opsin4, Pax6, and Opsin5 among four light intensities. Both Opsin4 and Pax6 expressions were significantly higher in the sea urchins at 1,500 lx than at 100, 300, and 700 lx, indicating a co-upregulation of Opsin4 and Pax6 expressions in sea urchins exposed to the high light intensity (for example, 1,500 lx). However, there was no significant difference of SiOpsin5 expression among the four light intensity groups. Consistent with our hypothesis, there was no significant difference in either Opsin4 or Pax6 expression between the two light intensity environments (low light intensity environment: 50–100 lx; high light intensity environment: 330–1400 lx). This indicates that positive phototaxis and negative phototaxis probably regulate the expression of Opsin4 and Pax6 to a similar level when sea urchins are exposed to the high and low light intensities, respectively. The present study provides new insights into the molecular basis of light intensity detection and subsequent phototaxis of eyeless species.  相似文献

兼养对雨生红球藻细胞生长的促进作用及藻株差异性   总被引:1,自引:0,他引:1  
结合叶绿素荧光技术,对比研究了不同雨生红球藻藻株在兼养(光照+乙酸钠)培养过程中细胞生长和光合作用的共性与差异性反应。结果表明:兼养明显提高红球藻细胞密度和比生长速率;乙酸钠不仅为红球藻提供异养的有机碳源,而且还明显导致藻细胞光自养的光合综合性能指数发生改变,兼养对细胞生长促进作用的贡献在前期主要来自于其中的异养部分,而在后期却明显来自于自养部分;兼养对细胞生长的促进作用在藻株之间存在一定差异,不同藻株对乙酸钠的适应能力和最适质量浓度也存在明显差异,藻株H0可适应的乙酸钠质量浓度较高,而藻株H6对乙酸钠需求和适应性相对较低。上述结果意味着规模化培养红球藻过程中,针对不同藻株采用适宜乙酸钠兼养方式可有效缩短培养周期,提高生产效率。  相似文献

In this study, protoplasts were successfully isolated from Kappaphycus alvarezii using snail enzymes, abalone enzymes and cellulase. The optimum enzymic ratio was fixed to be 20% of abalone enzyme, 12% of cellulase and the osmotic stabilizer was 2.0 mol/L glucose. The optimum enzymic hydrolysis conditions were found to be dark enzymolysis at 30°C continuing for 4.0 h. The resultant density and yield of protoplasts achieved 32.60×104 mL-1, 65.20×104 g-1 tissue for Kappaphycus alvarezii. Finally, under the temperature of 20°C, light intensity of 1 500–2 000 lx and photoperiod of 12 h/d, two developmental pathways were investigated:(1) callus-like cell mass and regenerated plantlet occurred on protoplast;(2) young shoots and calluslike cell mass occurred in tissue blocks after enzymolysis.  相似文献

丝状藻体在紫菜养殖的人工苗种生产中非常重要,在利用于分离提取高价值的藻红蛋白方面也具有潜在的利用价值。本研究采集了成熟紫菜叶状体的果孢子释放的初始丝状藻体,基于3因子3水平的正交实验设计方案[采用具有分析交互作用的正交表L27(313)],确定了丝状藻体藻红蛋白含量对环境因子的响应以及优化的调控培养条件。结果表明,温度、光照强度和盐度因子对丝状藻体藻红蛋白含量均有显著影响(P≤0.048 1),并且温度和光照强度或盐度因子间存在交互作用(P ≤0.043 1),但光照强度和盐度因子间的交互作用不显著(P=0.469 8)。获得的优化调控培养条件为:温度18 ℃、光强1 500 lx、盐度20或25。优化调控培养条件下的丝状藻体藻红蛋白含量高达64.73 mg/g.dw(平均值±标准偏差SD为58.24±5.07 mg/g.dw)。  相似文献

The change in concentration of the disulfated polyether yessotoxin (YTX) produced by a culture of the marine dinoflagellate Protoceratium reticulatum was measured in laboratory experiments under light and dark conditions. Experimental cultures were inoculated and grew at a growth rate of 0.14 d(-1) until stationary phase was reached, after approximately 21 days. Cultures were maintained in the stationary phase until 31 days after inoculation. Cells of P. reticulatum contained a concentration of approximately 10-15 pg YTX cell(-1) during stationary phase but this was considerably lower (<5 pg cell-1) during the growth phase. Low amounts of 45-hydroxy-YTX were also detected. At day 32, P. reticulatum was killed by cooling to 1 degrees C (confirmed microscopically) and YTX concentrations were measured periodically under light and dark conditions. YTX concentrations decreased rapidly to approximately 10% of the initial concentration within the first 3 days and depleted to near zero within a week in the light treatment. In the dark environment, YTX persisted longer with approximately 10% of the initial YTX concentration still remaining after 18 days.  相似文献

近年来,海洋微藻脂肪酸组成的研究及其应用越来越受到国内外科学家的重视。高度不饱和脂肪酸(PUFA),特别是长链的n-3 PUFA,如二十碳五烯酸(EPA)和二十二碳六烯酸(DHA)对海洋动物和人类都具有营养学和医学上的价值。在海洋微藻的培养过程中,光照强度是海洋微藻生长的条件之一,光线的明与暗,光度的强与弱,不仅对微藻的生长速率、产量有影响,而且对其脂肪含量和脂肪酸的组成也有影响(Teshima et al.,1983;Thompson et al.,1990; Renaud et al., 1991)。 作者在以往研究的基础上,选择了3种有代表性的海洋微藻:(1)小球藻Chlorella sp-2(李荷芳等,1999),此藻脂肪酸中EPA含量高、且不含DHA;(2)球等鞭金藻(Isochrysis galbana),该藻DHA含量较高,但几乎不含EPA;(3)前沟藻(Amphidinium sp.)的EPA、DHA含量均高。将以上3种微藻作为原料,在不同的光照强度下进行培养,测定并分析藻体中的脂肪含量和脂肪酸组成的变化,从而了解光强对海洋微藻脂肪含量及其脂肪酸组成的影响。  相似文献

光照对真鲷(Pagrosomusmojor)仔、稚、幼鱼的摄食有重要影响。摄食的适宜光照度范围为100~102lx,最适光照度范围为101~102lx。103lx强光对仔鱼摄食的影响大于稚、幼鱼;100lx弱光对9d以前的仔鱼和稚、幼鱼的摄食影响较小,对9d以后的仔鱼影响较大。此结果和鱼的感觉器官发育及鱼所摄食的饵料种类有关。  相似文献

从海域沉积物中富集分离出以芘作为唯一碳源和能源的海洋微生物,以ST4富集培养的混合微生物作为研究对象;该海洋混合菌株能利用菲(Phe)、芘(Pyr)、荧蒽(Fla)等多种多环芳烃;在不同浓度的芘的降解中,当芘的浓度为50mg/dm^3时,其生长水平和降解速率最高;当芘的浓度为200mg/dm^3时,其生长受到抑制,芘几乎不能被降解。外加营养盐酵母浸出液和葡萄糖促进降解微生物的生长,提高降解速率。研究表明了海洋微生物在多环芳烃污染环境的生物修复应用前景。  相似文献

Properties of the light saturation curve of photosynthesis and ribulose-1,5-bisphosphate carboxylase (RuBPC) activity are shown to change qualitatively in a natural population of marine phytoplankton during a spring bloom. Evidence is presented to show that these changes constitute photoadapative responses to increasing irradiance. As irradiance increased during the bloom, both the level of light-saturated photosynthesis (Pm) and the initial slope of the light saturation curve (α = photosynthetic efficiency) increased whether those parameters were normalized to chlorophyll a concentration (Pmb, αb) or to cell numbers (Pmc, αc). The magnitudes of these changes were such that Ik (= Pm/α, the photoadaptation parameter) did not change, but Im, the light intensity at which photosynthesis becomes saturated, increased. RuBPC activity, both chlorophyll a (RuBPCb) and cell number normalized (RuBPCc), also increased during the bloom. We suggest that these adaptations were achieved by simultaneously increasing the number of photosynthetic units, proportionately decreasing the photosynthetic unit size, and increasing both the concentrations of the enzymes of the dark reactions and possibly also of photosynthetic electron transport components.We also observed diminished levels of photoinhibition in the high light adapted cells late in the bloom and have suggested that this was a consequence of the same suite of physiological changes.In situ carbon fixation per cell increased during the bloom whereas no change occurred in this parameter when normalized to chlorophyll a concentration. Although these photoadaptive responses thus permitted carbon to be fixed in situ more rapidly per cell, at a constant efficiency with respect to investment of energy in the photosynthetic apparatus, they did not result in a change in growth rate. Based on consideratios of the role of time scale in physiological adaptation, however, it is suggested that the observed alterations in photosynthesis with increasing irradiance might permit a cell to more rapidly fill an energy quota for division, possibly an advantage in a mixing environment in which energy is patchily distributed, both spatially and temporalyy.Phosphoenolpyruvate carboxylase activity when normalized to chlorophyll a (PEPCb) did not change during the bloom while chlorophyll a normalized dark carbon fixation decreased sharply and was quantitatively small compared to PEPCb. On this basis and considering that RuBPCb increased during the bloom, it is suggested that, although PEPC may be involved in dark carbon fixation, its most important quantitative role is probably an indirect one in light dependent photosynthesis.We have also considered the relevance of laboratory results on photoadaptation to interpretations of field studies and have suggested that batch culture studies must be treated with caution but that turbidistat and semi-continuous methods provide reasonable simulations of natural conditions.  相似文献

从大西洋深海沉积物中分离得到1株产黑色素细菌b Ip-2,16S rRNA基因序列分析表明该菌隶属于假单胞菌属(Pseudomonas),与施氏假单胞菌(P.stutzeri)有最高的16S rRNA基因序列相似性(99%).进一步对该菌的生长及产黑色素条件进行了初步研究,结果表明b Ip-2在稳定生长期后期才开始大量积累黑色素;b Ip-2可在4~37℃的温度范围下生长,最适生长温度为37℃,在28℃具有最高的黑色素产量;b Ip-2生长的pH范围偏碱性,在p H=7的最适生长条件下黑色素产量最高,同时碱性环境有利于黑色素的快速形成;b Ip-2可以在1%~9%的NaCl浓度范围中生长,在5%的NaCl浓度下具有最高的黑色素产量;低浓度的Fe^2+(0.05 mmol/dm^3)可以提高黑色素的产量,Fe^2+浓度≥0.20 mmol/dm^3时明显抑制黑色素的生成,而Fe^2+浓度为1.00 mmol/dm^3时可完全抑制菌株的生长;低浓度的Cu^2+(0.1 mmol/dm^3)对黑色素的产量无明显影响,Cu^2+浓度≥0.5mmol/dm^3时可抑制黑色素的生成,而Cu^2+浓度超过1.0 mmol/dm^3时可完全抑制菌株的生长.通过PCR扩增获得菌株b Ip-2的对羟苯基丙酮酸双加氧酶(hydroxyphenylpyruvic acid dioxygenase,HPPD)全长基因(1087 bp),分析发现其编码的HPPD蛋白序列(含361氨基酸)与来自菌株Pseudomonas stutzeri RCH2的HPPD蛋白序列具有最高的同源性(序列相似性为97%).这些结果为深入研究该细菌所产黑色素的合成,应用及生态作用奠定了基础.  相似文献

经紫外诱变,筛选出在碳酸氢铵浓度为400 mg/L条件下可以生存的突变株。通过培养基中氮、磷、铁、温度、光照及pH值对雨生红球藻诱变株生长的影响进行比较,对以上3种营养盐及3种环境因子分别进行三因素三水平正交实验,以优化培养条件。进而对雨生红球藻野生型及诱变株的生长状况及色素含量进行比较。实验结果表明,雨生红球藻抗铵品系在培养基中NaNO30.1 g/L,KH2PO40.01 g/L,FeCl3.6H2O 1.0 mg/L,在pH值为8.0,光照强度100μmol.m-2.s-1,温度为18℃的培养条件下,雨生红球藻的生物量得到有效提高。  相似文献
