Quorum sensing, or auto induction, as a cell density dependent signaling mechanism in many microorganisms, is triggered via auto inducers which passively diffuse across the bacterial envelope and therefore intracellulaly accumulate only at higher bacterial densities to regulate specialized processes such as genetic competence, bioluminescence, virulence and sporulation. N-acyl homoserine lactones are the most common type of signal molecules. Aquaculture is one of the fastest-growing food-producing industries, but disease outbreaks caused by pathogenic bacteria are a significant constraint on the development of the sector worldwide. Many of these pathogens have been found to be controlled by their quorum sensing systems. As there is relevance between the pathogenic bacteria's virulence factor expression and their auto inducers, quorum quenching is a new effective anti-infective strategy to control infections caused by bacterial pathogens in aquaculture. The techniques used to do this mainly include the following: (1) the inhibition of signal molecule biosynthesis, (2) blocking signal transduction, and (3) chemical inactivation and biodegradation of signal molecules. To provide a basis for finding alternative means of controlling aquatic diseases by quorum quenching instead of treatment by antibiotics and disinfectants, we will discuss the examination, purification and identification of auto inducers in this paper.  相似文献

Quorum sensing(QS)disruption is considered as a potential alternative strategy to combat bacterial diseases in aquaculture.In this study,we isolated and identified bacteria degrading QS molecules from pond sediment and fish intestine.A total of 132 strains were obtained in the enrichment culture,of which two strains were identified as Enterobacter sp.f003 and Staphylococcus sp.sw120,being isolated from the fish intestine and pond sediment,respectively.We found that strains f003 and sw120 could degrade acyl-homoserine lactones(AHLs)and cause no hemolysis of sheep red blood cells.The AHL lactonase(aiiA)homologous gene in the two strains was detected in PCR amplification and the high-degrading activity to N-hexanoyl-L-homoserine lactone(C6-HSL)and AHLs secreted from pathogenic Aeromonas hydrophila was assessed.Meanwhile,the artificial infection of cyprinid Carassius auratus gibelio with intraperitoneal injection showed that the two strains were avirulent.Therefore,the obtained indigenous bacteria are candidate probiotics against pathogenic A.hydrophila in aquaculture.  相似文献

【目的】研究海洋土曲霉C23-3与不同类型海洋微生物共培养对其次生代谢产物的影响,挖掘土曲霉菌株C23-3的次生代谢潜力。【方法】将副溶血弧菌、枯草芽孢杆菌和胶红酵母的活菌或灭活菌体分别以不同时间与土曲霉C23-3共培养;采用DPPH自由基清除法和Ellman法分别评价各提取物的抗氧化和乙酰胆碱酯酶抑制活性,采用滤纸片法比较各提取物的抗菌活性,采用HPLC指纹图谱分析各提取物化学成分的差异。【结果与结论】发现不同类型的微生物与土曲霉共培养均可激活一些原本沉默的活性次级代谢产物;菌株及共培养物的化学拮抗能力、HPLC指纹图谱中的特征代谢产物变化揭示在活菌培养体系中土曲霉占竞争优势;而且灭活菌体中可能存在某些信号物质,在12 d时对土曲霉群体自感应信号分子丁内酯-I的产生具有比活菌更强的刺激作用。研究结果为深入挖掘海洋真菌代谢潜力及抗老年痴呆等活性先导化合物的发现提供基础。  相似文献

Cultured Apostichopusjaponicus in China suffers from a kind of skin ulceration disease that has caused severe economic loss in recent years. The disease, pathogens of which are supposed to be bacteria by most researchers, is highly infectious and can often cause all individuals in the same culture pool to die in a very short time. The 16S rRNA gene phylogenesis of the culturable bacteria from the lesions of diseased individuals was conducted to study the biodiversity of the bacterial communities in the lesions and to identify probable pathogen(s) associated with this kind of disease. S. japonica samples were selected from a hatchery located in the eastern part of Qingdao, China. Bacterial universal primers GM5F and DS907R were used to amplify the 16S rRNA gene of bacteria colonies, and touchdown PCR was performed to amplify the target sequences. The results suggest that γ- proteobacteria (Alteromonadales and Vibrionales) of CFB group, many strains of which have been also determined as pathogens in other marine species, are the predominant bacterial genera of the diseased Apostichopusjaponicus individuals.  相似文献

Flavobacterium columnare, the etiological agent of columnaris disease, is one of the most important and widespread bacterial pathogens of freshwater fish. In this study, we constructed two artificial selectable markers (chloramphenicol and spectinomycin resistance) for gene transfer in F. columnare. These two new artificial selectable markers, which were created by placing the chloramphenicol or spectinomycin resistance gene under the control of the native acs regulatory region of F. columnare, were functional in both F. columnare and Escherichia coli. The integrative/conjugative plasmids constructed by using these markers were introduced into F. columnare G4 via electroporation or conjugation. The integrated plasmid DNA was confirmed by Southern blotting and PCR analysis. These two markers can be employed in future investigations into gene deletion and the pathogenicity of virulence factors in F. columnare.  相似文献

The pathogenic species of genus Vibrio cause vibriosis, one of the most prevalent diseases of maricultured animals and seafood consumers. Monitoring their kinetics in the chain of seafood production, processing and consumption is of great importance for food and mariculture safety. In order to enrich Vibrio-representing 16S ribosomal RNA gene (rDNA) fragments and identify these bacteria further real-timely and synchronously among bacterial flora in the chain, a pair of primers that selectively amplify Vibrio 16S rDNA fragments were designed with their specificities and coverage testified in the analysis of seawater Vibrio community. The specificities and coverage of two primers, VF169 and VR744, were determined theoretically among bacterial 16S rDNAs available in GenBank by using BLAST program and practically by amplifying Vibrio 16S rDNA fragments from seawater DNA. More than 88.3% of sequences in GenBank, which showed identical matches with VR744, belong to Vibrio genus. A total of 33 clones were randomly selected and sequenced. All of the sequences showed their highest similarities to and clustered around those of diverse known Vibrio species. The primers designed are capable of retrieving a wide range of Vibrio 16S rDNA fragments specifically among bacterial flora in seawater, the most important natural environment of seafood cultivation.  相似文献

Vibrio parahaemolyticus (VP) is one of the pathogenic vibrios endangering net-cage cultured Pseudosciaena crocea,Fennerpenaeus chinensis, and shellfish in coastal areas of China. Several types of hemolysins produced by Vp have been characterized as major virulence factors.They are thermostable direct hemolysin (TDH),TDH-related hemolysin (TRH) and thermolabile hemolysin (TLH). In this study, we cloned tdh, trh, and tlh genes from the genome DNA of VP by polymerase chain reaction (PCR).We ligated the three genes into prokaryotic expression vector pET-28a (+),and transformed the recombinant plasmids into Es-cherichia coli BL21 (DE3). The expression of recombinant proteins was induced by isopropyl-β-D-thiogalacto-pyranoside (IPTG). The recombinant proteins were expressed in a form of inclusion bodies and thus purified with Ni-NTA affinity chromatography. Western blotting results showed that recombinant proteins,TDH, TRH and TLH, could be recognized by rabbit anti-VP serum. The three purified proteins were renatured by gradient dialysis.The renatured proteins exhibited hemolytic activity except for TLH in the presence of phosphatidylcholine. These results not only are helpful for better understanding these genes’ functions under a single factor level, but also provide evidence for VP vaccine engineering.  相似文献

尺度效应是地球科学和定量遥感中的重要研究课题,目前的许多研究大多集中在估算尺度效应带来的误差,而对一些关键的植被结构参数是否存在尺度效应及其尺度转换方法尚存在诸多不同见解。本文针对真实和有效叶面积指数(Leaf Area Index, LAI和Effective LAI, LAIe)以及聚集指数(Clumping Index, CI)3个植被关键结构参数,从基本概念和获取方法上分析参数的尺度效应及其尺度转换方法。从定义上看,LAI并不存在尺度效应,而LAIe和CI则存在尺度效应,其中CI的尺度效应由LAIe引入(CI=LAIe/LAI)。在野外实测中,LAI破坏测量法没有尺度效应,但由孔隙率模型获取3个参数的方法均具有尺度效应。异速生长方程和遥感反演方法的尺度效应取决于方法本身的线性或非线性特征。目前全球主要的LAI、LAIe和CI遥感产品都基于非线性模型获取,其反演过程具有尺度效应。像元尺度的LAI本身并不具有尺度效应,而像元尺度的LAIe和CI虽然具有尺度效应,但在实践中常常被忽略。因此,实际工作中应注意区分参数概念本身、野外测量、遥感反演方法以及遥感产品等所展示的不同尺度效应。  相似文献

Seaweeds are known to exhibit various antimicrobial properties,since it harbours an enormous range of indigenous bioactive compounds.The emergence of drug resistant strains has directed to the identification of prospective metabolites from seaweed and its endophytes,thereby exploiting the properties in resisting bacterial diseases.The current study was aimed to assess the antimicrobial activity of extracts obtained from Ulva reticulate,for which metabolites of Ulva reticulata and its endophytes were extracted and assessed against human pathogens like Staphylococcus aureus,Escherichia coli,Pseudomonas aeruginosa,Salmonella typhi,and Bacillus subtilis.It was observed that the hexane extract of isolate VITDSJ2 was effective against all the tested pathogens but a significant inhibition was observed for Staphylococcus aureus and Escherichia coli.Further,Gas chromatography coupled with Mass spectroscopy(GC-MS) revealed the existence of phenol,3,5-bis(1,1-dimethylethyl) in the crude hexane extract which is well-known to possess antibacterial activity.The effective isolate VITDSJ2 was identified to be the closest neighbour of Pseudomonas stutzeri by phenotypic and genotypic methods.The crude extracts of the seaweed Ulva reticulata was also screened for antibacterial activity and the hexane extract was effective in showing inhibition against all the tested pathogens.The compound in the crude extract of Ulva reticulata was identified as hentriacontane using GC-MS.The extracts obtained from dichloromethane did not show significant activity in comparison with the hexane extracts.Hence the metabolites of Ulva reticulata and the bacterial secondary metabolites of the endophytes could be used in the treatment of bacterial infections.  相似文献

The purpose of this study was to investigate the antibacterial activity of extracts from Zostera marina against the pathogens of Apostichopus japonicus skin ulceration disease. When 95% ethanol (v/v) solvent was used to extract Zostera marina at 50°C, aqueous extract (ZA) showed obvious bacteriostatic effects on the tested bacterial strains (inhibition halo diameters between 8.23 mm and 13.62 mm), whereas the ethyl acetate extract (ZE) was almost inactive. The minimal inhibitory concentration (MIC) of ZA against four pathogens were homogeneous at 12.8 g L−1. ZA components were analyzed by thin layer chromatography (TLC) assay and six fractions were obtained. In another study, the six fractions showed inhibitory effects against the tested bacteria while their functions seemed to counteract the ZA activity.  相似文献

为了更好地消除混杂在变形序列中的噪声,利用完备经验模态分解(CEEMD)将形变信号自适应分解为不同尺度的振动模态。针对分解分量中信号和噪声区分标准不唯一的问题,构造一种CEEMD与自相关分析相结合的去噪算法,实现有效信号和随机信号的分离。将该算法应用在仿真实验和GNSS变形监测实测数据处理中,并与传统的小波去噪方法进行比较。结果表明,该算法避免了小波基选择带来的影响。  相似文献

Vibrio anguillarum is an important bacterial pathogen of aquatic organisms and a significant problem in aquatic farming.The rapid detection and identification of V.anguillarum,and other pathogens that infect marine organisms,is crucial to effective disease management.In this study,we developed a loop-mediated amplification (LAMP) assay to detect V.anguillarum in an hour in a single tube without the need for thermal cycling.Conserved regions of the metalloproteinase (empA) gene of V.anguillarum served as the targets for primer design.A fragment of the empA gene was amplified at 65°C in the presence of the primer mixture and Bst DNA polymerase.In the optimized LAMP assay,6.7 pg of V.anguillarum DNA could be detected.Six strains of V.anguillarum and 17 strains of non-V.anguillarum bacteria were used in this study to evaluate the species specificity of the primers.The six V.anguillarum strains gave a positive result in the LAMP assay.This method was also validated in V.anguillarum-infected fish.This LAMP method is more sensitive than PCR in the detection of V.anguillarum and shows good species specificity.The LAMP assay is therefore an effective method for the quick detection of V.anguillarum both in the laboratory and in the field.  相似文献

采用微生物分离培养技术和巢式PCR技术相结合方法对2011年采自广西北海南万和古城、钦州茅岭和钦州港、防城港企沙和公车的拟穴青蟹(Scylla paramamosain)携带寄生虫、细菌和病毒状况开展调查.寄生虫检测结果显示,仅钦州茅岭拟穴青蟹鳃组织中发现寄生虫茗荷(儿).细菌性病原体检测结果显示,共有11种病原性细菌存在于健康养殖拟穴青蟹体内,弧菌属细菌性病原出现频率最高,其中副溶血弧菌和溶藻弧菌在调查区域内均有分布,其他菌种则呈现一定的地域特异性.病毒性病原体检测结果显示,白斑综合症病毒仅在钦州茅岭拟穴青蟹检出,阳性率为4.94%;双顺反子病毒于北海古城和钦州茅岭拟穴青蟹检出阳性携带率为3.70%;呼肠孤病毒携带率则较高,北海、防城港以及钦州拟穴青蟹均有检出,阳性率达25.93%.由此可见,10-11月广西区内健康养殖拟穴青蟹携带弧菌和呼肠孤病毒的比例远远高于其他病原体,为拟穴青蟹携带的主要病原体.  相似文献
