应用ISSR分子标记技术分析二种体色拟穴青蟹群体的遗传多样性.10条ISSR引物共扩增出81条带,其中73条表现出多态性,占总条带数的90.12%.暗红色和深绿色拟穴青蟹群体的多态性位点比率分别为81.48%和77.78%,暗红色群体的Nei’S基因多样性指数(He=0.3189)和Shannon’S信息指数(,=0.4690)略高于深绿色群体(He:0.3054,,=0.4491).群体间的Nei’S遗传相似性(0.9484)和遗传距离(0.0529)表明,群体间遗传分化属于种内遗传分化.POPGENE和AMOVA分析表明,总的遗传变异主要来源于群体内个体间,群体间发生中等程度遗传分化,群体间存在强大的基因流.UPGMA聚类图可视化群体间和群体内的遗传变异,显示60个个体并没有按体色聚类.  相似文献

广东沿海杂色鲍养殖群体遗传多样性的微卫星分析   总被引:1,自引:0,他引:1  
实验选取的7个杂色鲍(Haliotis diversicolor)养殖群体分别来自广东汕头、汕尾、惠东、湛江和徐闻,深圳野生驯养,利用7对微卫星引物对上述群体进行了遗传多样性和群体遗传结构的分析。结果显示:7个位点产生的等位基因数从7~13个不等,共检测到68个等位基因,平均每个位点的等位基因数为9.71个。有效等位基因数的范围为3.8~8.1,平均观测杂合度范围为0.422~0.906,平均期望杂合度范围为0.748~0.917。7个群体的多态信息含量变化范围为0.657~0.883,PIC值均在0.500以上,表现为高度多态性。与两个野生驯养的养殖群体相比,其他5个养殖群体均存在不同程度的遗传多样性降低。AMOVA分析显示,4.79%的遗传变异来自于群体之间,95.21%的变异来自于群体内的个体之间。研究表明,广东沿岸的杂色鲍具有较高的遗传多样性水平,7个群体之间呈现一定的遗传分化。  相似文献

为确定三种胡子鲶(胡子鲶Clarias fuscus、两栖胡子鲶C.batrachus和革胡子鲶C.lazera)的遗传变异及系统分类关系,采用淀粉凝胶电泳技术比较了三种胡子鲶4个样本的30—31个基因座位。每个座位观测到的平均等位基因数(Na)为1.01—1.39;多态座位百分比P_(0.99)为3.33—16.13%,平均杂合度观测值(Ho)为0.004—0.072;期望值(He)为0.004—0.064。3个种的遗传变异均较小,其中以革胡子鲶的遗传变异较其它两种大。两栖胡子鲶因采样的限制和养殖群体的奠基者效应而引起多态基因座位减少。4个样本的遗传相似性(I)和遗传距离(D)的比较,表现了3个水平的进化趋异;1)胡子鲶的两个没有生殖隔离的群体间的比较,彼此具有高度的遗传相似性(I=0.999);2)胡子鲶和两栖胡子鲶这对姐妹种间的比较,尽管它们在形态上难以分辨,但在遗传上仍有11个基因座位各具种的特异性;3)外表区别明显的种间比较,彼此表现了很大的遗传分异(D=0.999和0.818)。可见胡子鲶与两栖胡子鲶之间的系统分类关系较它们与革胡子鲶间的关系更密切。  相似文献

The study investigated the global and regional phylogeography of the yellowtail kingfish Seriola lalandi by examining genetic diversity and population genetic structure of this species at inter-and intra-ocean level and on a regional scale. DNA fragments of two mitochondrial genes, cytochrome b (Cytb) and cytochrome c oxidase subunit I (COI) and one nuclear gene, recombination activating gene 1 (RAG1), were sequenced to investigate the global-scale phylogeography of this species. The population genetic structure within the South Pacific, as well as along the South African coastline, was examined further using six microsatellite markers. Three distinct clades were identified for S. lalandi, which correspond with previously described subspecies of the North-East Pacific, North-West Pacific and the Southern Hemisphere. Within the latter, additional divergence was observed between the South Pacific and the South-East Atlantic regions. Divergence estimates were indicative of a Pacific origin for S. lalandi populations, because of Pleistocene vicariant events. Microsatellite analyses revealed overall significant genetic differentiation between South African and South Pacific samples. This corroborates recent findings on the global phylogeography of the species. No population differentiation was observed within South Africa, indicating high levels of gene flow.  相似文献

The small yellow croaker (Larimichthys polyactis) belongs to the family Sciaenidae, which is an offshore warm fish species and widely distributed in the western Pacific. In this study, the variation of genetic diversity and genetic differentiation among L. polyactis populations was analyzed by mitochondrial DNA control region. A total of 110 polymorphic sites were checked, which defined 134 haplotypes. High level of haplotype diversity (h=0.993±0.002) was detected in the examined range. Population genetic structure analyse (analysis of molecular variance, Fst) showed there were high gene flow among L. polyactis populations. The result showed that there were relatively high genetic diversity and low genetic differentiation among the Yellow Sea and the East China Sea populations, which can be attributed to diverse habitats, wide distribution range and high mutation rate of control region. Using phylogenetic methods, coalescent analyses (neutrality tests, mismatch distribution analysis, Bayesian skyline analyses) and molecular dating interpreted in conjunction with paleoclimatic and physiographic evidence, we inferred that the genetic make-up of extant populations of L. polyactis was shaped by Pleistocene environmental impacts on the historical demography of this species. Besides, relatively constant genetic diversity and larger effective population size were detected in recent L. polyactis population. The result showed that the fishing policy certainly, such as the summer closed fishing, played a role in protecting resources of L. polyactis. This study can offer a wealth of biological novelties which indicates genetic structure of L. polyactis population and provides the foundation for resources protection and policy setting.  相似文献

为探究不同地理条件下斑节对虾天然群体的遗传多样性和遗传分化水平, 选用14对简单重复序列(simple sequence repeat, SSR)特异引物对广东湛江、海南三亚、海南琼海的斑节对虾天然群体共计90尾个体进行微卫星标记分析, 每对引物检测出的等位基因数为2~4不等, 平均2.7。3个群体(湛江、三亚、琼海)平均有效等位基因分别为1.9081、1.9715和2.0185, 平均多态信息含量分别为0.3864、0.3926和0.4078, 总的平均期望杂合度(0.4605 )明显高于观测杂合度(0.4046), 表明3个群体遗传多样性水平为中等, 且存在杂合度缺失的现象。种群分化指数和遗传距离分析表明, 在不同群体间已产生了遗传分化, 但是分化较小; 三亚群体和琼海群体之间的亲缘关系最近, 而与湛江群体的亲缘关系最远。分子方差分析表明, 群体间的遗传变异指数为8%, 群体内个体间的遗传变异指数为92%, 基因分化主要发生在群体内, 而不是群体之间。  相似文献

利用线粒体细胞色素b基因序列对中华鳖5个不同地理种群(太湖群体、日本群体、江西群体、台湾群体和乌鳖群体)的种群遗传结构进行分析.以特异引物进行PCR扩增,获得了408bp的基因片断序列.序列分析结果表明,在31个中华鳖个体中,共检测到17个变异位点,获得8种单倍型.日本群体的2种单倍型在其他4个群体中都未检测到,为该群体所特有,日本种群与其他4个群体的遗传距离最远.除日本群体缺乏主体单倍型外,其他4个群体共同拥有主体单倍型Hap.1,江西群体、台湾群体和乌鳖群体的单倍型类型都和其他群体共同享有,没有其特异的单倍型.MP法和NJ法构建的系统发生树显示日本群体的两种单倍型以很高的置信度聚成一支,这与其地理分布存在一定的相关性,其序列上的差异,可以成为种群鉴别的分子标记;另外4个群体的6种单倍型聚合成一大支,但同一个地理区域的群体并未聚成一类,未按照地理位置形成明显的地理格局.  相似文献

A wild and a cultured greenshell mussel (Perna canaliculus) population were compared for biochemical genetic variation at seven polymorphic and four monomorphic allozyme loci. Significant heterozygote deficiencies were observed for all polymorphic loci (La—1, La—2, Lap, Lgg—1, Lgg— 2, Pgi, and Pgm) for both populations (except the Pgi locus of the wild mussel population). Genotypic disequilibrium was calculated for both populations: genotypic frequencies were significantly non‐random at three pairs of loci among the wild mussels, and significantly non‐random at three different pairs of loci among the cultured mussels. All six pairs of loci which exhibited significant genotypic disequilibrium involved amino‐peptidases, suggesting that these loci form a linkage group, and that neither the Pgi nor the Pgm loci are associated with this group. Exact tests for population differentiation based upon population‐specific allele distributions indicated that four of the polymorphic loci were significantly heterogeneous among the two populations, whereas the remaining three polymorphic loci were not. Based upon the private allele system, the number of migrants (N m) between the populations was estimated to be 2.009, which, according to the private allele system, represents a high level of gene flow. These findings are discussed with regard to the population biology and genetic structure of this species.  相似文献

采用聚丙烯酰胺梯度凝胶垂直平板电泳检测和比较了广东电白、南三和广西北海三个文蛤(Meretrix meretrix)野生群体9种同工酶24个基因座位的生化遗传变异。结果表明,三个群体的平均杂合度观察值分别为0.291,0.251和0.290,多态位点比例(P0.95)分别为37.5%,37.5%和41.6%,位点平均有效等位基因数分别为1.476,1.419和1.479,各群体在多个位点上都存在杂合子缺失现象。比较了三个群体之间的遗传相似度(I)和遗传距离(D),其中电白与南三群体间的遗传分歧极微,它们与北海群体间的遗传分歧稍明显,三群体基因分化系数为0.035,预示三地间有着较强的基因流。  相似文献

Heterogeneity in genetic composition among recruits, mostly due to a large variance in reproductive success mediated by oceanographic processes, has been reported for marine species but is less understood in coastal lagoons' organisms. Temporal genetic variation in natural populations of the Mediterranean shore crab Carcinus aestuarii was quantified over a multi-year sample. A total of 486 adult crabs were collected at eight different sites of the Venice Lagoon during the period 2005–2007 and screened for genetic variation using 11 microsatellite loci. Two additional samples (N = 115) from neighbouring sites, located approximately 100 km North and South to the Venice Lagoon, were included for the sake of comparison. Our results show significant differences in allelic frequencies at the micro-geographic scale of the Venice Lagoon, observed between sites of collection, typologies of habitat, and areas with different class of ecological risk or pattern of hemocyanin expression. However, this pattern was not constant between years, with significant differences observed mainly in 2005 and 2006, but not in 2007. Our results indicate significant temporal differences suggesting the existence of dynamic processes that act on the genetic pool of this species. Although natural selection and gene flow might play a role, we suggest that genetic drift linked to large variation in the reproductive success of individuals is the most probable scenario to explain the local genetic patterns of differentiation in the Mediterranean shore crab. Our study, by providing the first evidence for the existence of genetic differences in this species at the micro-geographic scale, suggests that a better comprehension of the link between reproduction, recruitment and oceanography is critical to understand how colonization and maintenance of genetic variation is achieved in ephemeral and vulnerable environments such as coastal lagoons.  相似文献

An increasing number of investigations revealed that responses of populations to environmental stressors may be genetically dependent. Occurrence of genetic adaptation to high concentrations of heavy metals in Hediste diversicolor local ecotypes has also been hypothesised. Analyses of allozymic patterns are a promising tool for the genetic monitoring of environmental stressors. The aim of this study was to investigate whether the allozymic patterns of H. diversicolor could be affected by levels of contamination. Five sites were investigated: four in a polluted brackish habitat, the Pialassa lagoons, and a reference site in the Lamone estuary. Total mercury was used as a tracer of the overall level of contamination. Concentrations of mercury were quantified in both sediments and H. diversicolor tissues by means of cold vapour atomic fluorescence spectroscopy. A gradient of mercury concentration in both sediments and H. diversicolor tissues was found. Moreover, significant differences in concentration of mercury were observed among and within sites. Allozyme electrophoresis was used to test the occurrence of genetic differentiation among H. diversicolor samples. A total of 19 putative gene loci were analysed. For six gene loci, significant variability in genotypic and allelic frequencies was detected. Patterns of differentiation, which could be related to the contamination levels, were found at loci LDH, PGI, SDH. Other loci (ALD, FH, HBDH) showed a significant genetic structuring among samples, not related to concentrations of mercury.  相似文献

The present investigation focuses on population genetic structure analysis of the endangered giant clam species Tridacna maxima across part of the Red Sea,with the main aim of assessing the influence of postulated potential barriers to gene flow(i.e.,particular oceanographic features and marked environmental heterogeneity)on genetic connectivity among populations of this poorly dispersive bivalve species.For this purpose,a total of 44 specimens of T.maxima were collected from five sampling locations along the Saudi Arabian coast and examined for genetic variability at the considerably variable mitochondrial gene cytochrome c oxidase I(COI).Our results revealed lack of population subdivision and phylogeographic structure across the surveyed geographic spectrum,suggesting that neither the short pelagic larval dispersal nor the various postulated barriers to gene flow in the Red Sea can trigger the onset of marked genetic differentiation in T.maxima.Furthermore,the discerned shallow COI haplotype genealogy(exhibiting high haplotype diversity and low nucleotide diversity),associated with recent demographic and spatial expansion events,can be considered as residual effect of a recent evolutionary history of the species in the Red Sea.  相似文献

采用垂直板型聚丙烯酰胺凝胶电泳测定了三种不同生境的桐花树(Aegiceras cor niculatum)种群的遗传多样性和遗传分化。桐花树和种群水平都维持有较高的遗传变异性,观察杂合度分别为0.252和0.244。期望杂合度分别为0.266、0.240。种群间遗传分化系数为0.127,表明总的遗传变异中有12.7%来自种群间,种群间遗传距离和遗传一致度均值分别为0.060和0.964,基因流畅,Nm=2.187。  相似文献

4种石斑鱼的分子遗传多样性和亲缘关系的RAPD分析   总被引:4,自引:0,他引:4  
采用25个引物,对海南养殖的点带石斑鱼、广东养殖的斜带石斑鱼、南海西沙野生鲑点石斑鱼和蜂巢石斑鱼的基因组DNA进行了RAPD分析,并用UPGMA法对4种石斑鱼进行聚类分析.研究结果表明:(1)野生鲑点石斑鱼和蜂巢石斑鱼群体内遗传多样性较高,多态位点比率分别为58.07%和55.65%,平均遗传杂合度分别为0.1793和0.1622,表明南海野生石斑鱼遗传变异水平较高,种质资源状况良好;而养殖的点带石斑鱼和斜带石斑鱼遗传多样性偏低,多态位点比率分别为49.70%和40.38%,平均遗传杂合度分别为0.1349和0.1135,表明石斑鱼养殖群体遗传多样性水平有所下降,应及时采取适当的保护措施.(2)4种石斑鱼种间遗传距离及聚类分析表明,点带石斑鱼与斜带石斑鱼遗传距离最近(D=0.2570);鲑点石斑鱼与蜂巢石斑鱼遗传距离次之(D=0.5146);斜带石斑鱼与蜂巢石斑鱼的亲缘关系最远(D=0.5810).(3)在8个引物中,有区分4种石斑鱼的特异性片段,可用于这些石斑鱼的鉴定.  相似文献

花斑蛇鲻(Saurida undosquamis)是一种重要的底层经济鱼类, 本研究利用线粒体控制区(D-loop区)序列分析了中国近海分布的花斑蛇鲻的遗传结构和遗传多样性, 一共测定了6个地理群体129尾样本的D-loop区全序列。结果显示, 全长为921bp的序列包含71个多态性位点, 共检测到101个单倍型。花斑蛇鲻总体呈现很高的单倍型多样性(0.9873 ± 0.0048)和较低的核苷酸多样性(0.0132 ± 0.0067)的特征。基于邻接法构建的单倍型系统发育树的拓扑结构很浅, 没有形成分化明显的支系。单倍型在各个地理群体中的分布呈分散交叉状态, 表明地理群体间没有显著的遗传分化。6个群体的分子方差分析显示, 花斑蛇鲻绝大部分的遗传变异(99.87%)来源于群体内的个体之间, 而群体间的变异仅占0.13%。大部分地理群体之间的遗传分化指数(FST)均很小, 揭示了群体间的基因交流很频繁, 存在高度的遗传同质性。这些研究结果表明中国近海的花斑蛇鲻遗传多样性丰富, 没有明显的遗传分化, 是一个随机交配群, 可以作为同一个渔业单元来管理。  相似文献

The genetic diversity and variation of three pseudo‐species of the commercially important Indian conch Turbinella pyrum from the southeast coast of India were studied. A total of 88 individuals was genotyped at 12 loci and differences in the genetic diversity among genetic groups were correlated with known population histories. The study identified 535 alleles and all the studied loci were highly polymorphic. The number of alleles ranged between four and 29 with a global mean of 14.861. The global means of the observed and expected heterozygosities were 0.774 and 0.891, respectively. The estimated within‐population inbreeding value (FIS = 0.131) indicated a shortfall of heterozygosity in the populations. An analysis of molecular variance showed that 5% of the total variation was due to differences between genetic groups. The Bayesian approach revealed strong distinctiveness in the three genetic groups studied. Overall microsatellite analysis revealed less genetic diversity in all studied genetic groups and significant genetic variation among the three pseudo‐species. The study highlights the significant distinctiveness of the pseudo‐species of T. pyrum and the need for appropriate conservation efforts in order to prevent further losses of their genetic diversity.  相似文献

密斑马面鲀Thamnaconustessellatus是在我国南海海域较为常见的一种马面鲀属鱼类。本研究基于线粒体DNA控制区序列对南沙群岛密斑马面鲀3个群体的遗传结构及其遗传多样性进行了分析。研究结果显示,密斑马面鲀的控制区序列变异程度较大,86尾个体序列的变异位点数为38个,共定义了28个单倍型;3个密斑马面鲀群体的单倍型多样度和核苷酸多样度均较高,且相差不大;密斑马面鲀群体间未检测到显著的群体遗传结构,分子方差分析表明,99.67%的遗传差异来自于群体内,群体间遗传差异仅为0.33%。贝叶斯树和最大似然树均显示出密斑马面鲀单倍型间松散的分布,未检测到显著的谱系结构。群体历史动态分析结果表明密斑马面鲀的群体经历了更新世的群体扩张事件。  相似文献
