本实验采用群体感应抑制(quorum sensing inhibitory, QSI)活性导向法,对分离自山东威海近海底泥的真菌 WH7-2开展代谢产物研究。综合菌落形态和转录间隔区(Internal Transcribed Spacer, ITS)全序列分析,菌株 WH7-2 鉴定为腐皮镰刀菌 Fusarium solani。综合运用多种色谱方法,从该真菌大米发酵产物的活性部位中分离得到 11 个化合物。分别鉴定为(2E, 5E)-3, 5, 7-三甲基-2, 5-辛二烯酸(1)、不饱和脂肪酸酯混合物(2 4)、镰红菌素-3-甲醚(5)、脱水镰红菌素(6)、(22E)-5α, 8α-过氧化麦角甾-6, 22-二烯-3β-醇(7)、(22E)-5α, 8α-过氧化麦角甾-6, 9 (11), 22-三烯-3β-醇(8)、3β-羟基-胆甾-5-烯-7-酮(9)、6β-羟基-胆甾-4-烯-3-酮(10)和(22E)-胆甾-5, 22-二烯-3β-醇(11)。其中不饱和脂肪酸酯混合物(2 4)具有 QSI 活性,其他化合物无 QSI 活性。除 5 和 6 外,其他化合物均为首次从腐皮镰刀菌 F. solani 中发现。  相似文献

文章旨在研究南海冷泉来源真菌Talaromyces helicus SCSIO41311中聚酮类次生代谢产物。主要采用正相硅胶柱层析、反相硅胶柱层析、Sephadex LH-20、高效液相色谱(high performance liquid chromatography, HPLC)及薄层色谱(thin layer chromatography, TLC)等多种方法进行分离纯化; 通过核磁共振波谱(nuclear magnetic resonance, NMR)、质谱(mass spectrometry, MS)等现代波谱分析方法及理化性质进行结构鉴定。从中分离得到7个聚酮类化合物, 分别为: desmethylsulochrin (1)、sulochrin (2)、monomethylsulochrin (3)、circinophoric acid (4)、dimethyl 2,3-dimethylosoate (5)、endocrocin (6)和1-methyl emodin (7)。所有化合物均为首次从深海冷泉微生物中得到, 其中化合物1为首次报道其核磁数据。  相似文献

The soluble organic matters were extracted from marine sediments in the sea area of Southern Ocean by using organic solvents. The hydrocarbon and carboxylic acid fractions were separated with the thin layer chromatography. The organic compounds of 170(H), 21α(H)- bishomomoretene, cholest - 5 - ene, 24 - methyl - cholestene, 24 - ethylcholestene, hop-17(21)-ene, neohop - 13(18)-ene, fern-8-ene, fern-7-ene, 170(H), 210 (H) - hop - 22(29)-ene (diplotene), steranes, β, β-hopanes C29 -C31 and unsaturated carboxylic acids C18:2Δ9.12 , C18:1Δ9,C18:1Δ11 , have been identi-fied in the hydrocarbon and carboxylic acid fractions by means of gas chromatography and gas chromatography - mass spec-trometry - data system. The distributions of 3, a - bishomomoretene, β,β- hopene and β,β- hopane are characteristic of immature organic matter. 从松节藻中分离的含卤素化合物表现出显著PTP1B抑制活性。其中BPN效果最佳。为研究BPN侧链基团极性对其抑制活性的影响,本文以香兰素(1)为起始原料,经过溴代、氧化、还原、傅克烷基化以及酯化等反应,成功合成了新化合物2,3-二溴-1-(2-溴-3,4-二甲氧基-6-((4-硝基苯氧基)甲基)苯甲基)-4,5-二甲氧基苯(10)、4-((3-溴-2-(2,3-二溴-4,5-二甲氧基苄基)-4,5-二甲氧基苄基)醚)苯胺(11)、4-((3-溴-2-(2,3-二溴-4,5-二甲氧基苄基)-4,5-二甲氧基苄基)醚)-4-乙酰乙酸(12)和3-溴-2-(2,3-二溴-4,5-二甲氧基苄基)-4,5-二甲氧基苯甲醛肟(14)。通过1 H-NMR,13C-NMR等方法对目标产物进行了结构表征。并对化合物进行了PTP1B酶抑制活性的测定,结果表明化合物侧链极性对PTP1B酶抑制活性有显著影响。  相似文献

对采自广西涠州岛近海多管藻(Poiysiphonia urceolata)分离到的一株内生真菌EN-22的次生代谢产物的化学成分进行了研究.利用正相硅胶柱层析、葡聚糖凝胶Sephadex LH-20柱层析、制备薄层层析(PTLC)以及重结晶等分离手段从该菌发酵液中分离得到15个化合物,通过一维、二维核磁共振技术、质谱技术等鉴定了所有化合物的结构,分别为:4-(4-喹啉)-4-羟基-2-丁酮(1),3-羟基-3-(2-氧丙基)吲哚-2-酮(2),3-吲哚乙醇(3),3-吲哚甲酸(4),2-羟基-3-吲哚丙酸(5),2-酮-3-吲哚乙醇(6),3,3-二吲哚-2-羟基-丙醇(7),β-咔啉(8),尿嘧啶(9),环-(S-脯氨酸-S-异亮氨酸)(10),(22E,24R)-3β,5α,9α-三羟基麦角甾-7,22-二烯-6-酮(11),(22E,24R)-5α,6α-环氧麦角甾-8,22-二烯-3β,7α-二醇(12),5α,6α-环氧-24(R)-甲基胆甾-7,22-二烯-3β-醇(13),胡萝卜苷(14),过氧化麦角甾醇(15);其中化合物1和2为新天然产物,并首次报道化合物1的碳谱数据.  相似文献

研究南海礁栖腔节藻Coelarthrum sp.共附生真菌Pestalotiopsis neglecta SCSIO41403的次级代谢产物。采用中压正相柱层析色谱法和半制备高效液相色谱法, 对真菌发酵产物进行分离纯化, 制备获得6个次级代谢产物单体化合物, 使用核磁共振、质谱等波谱学方法, 结合其理化性质以及文献数据对比, 分别鉴定为: gamahorin (1)、microsporaline C (2)、7-hydroxy-5-methoxy-4,6-dimethylphthalidc (3)、demethyl nectriapyrone A (4)、4-hydroxybenzaldehyde (5)和4-hydroxyphenethyl acetate (6)。这6个化合物均为首次从该菌株发现, 其中化合物1、2、5和6是首次从海洋动植物共附生真菌中发现。抗菌活性筛选显示其中化合物3对白色念珠菌有明显抑制活性。  相似文献

运用硅胶柱层析、Sephadex LH-20凝胶柱层析和HPLC等技术,采用活性追踪分离的方法,对南海短指软珊瑚Sinularia sp.的化学成分进行了研究.从乙醇提取物中分离获得单体化合物,运用NMR,MS等波谱技术鉴定了8个化合物的结构,分别为:3β-羟基-24-亚甲基胆甾-5-烯-7-酮(1),孕甾烯醇酮(2),cembrene A(3),epoxycembrane A(4),4,8,12-trimethyl-1-(1-methylethenyl)-3,7-cyclotetradecadien-10-one(5),(3E,7E,11E)-11,12-dihydroxy-1-isopropyl-4,8,12-trimethyicyclotetradeca-1,3,7-triene(6),邻苯二甲酸二丁酯(7)和十九烷-2-酮(8).化合物5和8为首次从珊瑚动物中分离得到,6为首次从短指软珊瑚属中分离得到.化合物3和4显示较强的卤虫致死活性.西松烷二萜类化合物是该短指软珊瑚中的重要活性成分.  相似文献

采用围隔实验生态学方法研究了对虾综合养殖生态系有机碳的平衡,结果表明:(1)研究期间各实验围隔有机碳的总收入量为4847.46~6154.67 g,平均为(5646.94±551.09)g.有机碳总收入量与浮游植物生产量、附生藻类生产量及颗粒饲料(含饵料鱼)等各组分碳收入量的比例平均为1:0.73:0.11:0.16.(2)研究期问各实验围隔有机碳的总支出量为3310.28~3974.79g,平均为(3644.21±281.44)g.有机碳总支出量与浮游生物群落呼吸量、附生生物群落呼吸量、底栖群落呼吸量、养殖生物生产量、养殖生物呼吸量等各组分碳支出量的比例平均为1:0.53:0.19:0.15:0.04:0.09.(3)各实验围隔有机碳积累量为1383.45~2707.31 g,平均为(2002.73±546.76)g,占收入量的(35±7)%(26%~44%).不同养殖模式实验围隔依积累量大小顺序为:Y5(对虾-扇贝)、Y7(对虾-罗非鱼)、Y4(单养对虾)、T5(对虾-罗非鱼-缢蛏)、Y6(对虾-缢蛏).(4)围隔收入的有机碳对对虾的转化率为(1.77±0.62)%(0.76%~2.28%),转化率大小顺序为Y6,T5,Y4,Y5,Y7,收入的有机碳对全部养殖动物的转化率为(2.18±0.79)%(1.07%~3.12%),转化率大小顺序为T5,Y6,Y5,Y4,Y7.  相似文献

In order to clarify the chemical forms of minor metallic elements occurring in the ocean, a new method of separation of organic compounds of metals in sea water by using XAD-2 resin was contrived. By applying the new method of determination, it is found that, (1) More than 80 % of V, Fe, Cd and Cu dissolved in sea water are present in organic forms regardless of the depth; (2) A most part of Co and Pb are present in organic forms near the surface, but they change into inorganic forms in the deep layer up to 50 to 60%; (3) Of the total amounts of dissolved Al, Ni, Zn, Ag, Mo, and U, less than 30 % are present in organic forms in sea water anywhere in the ocean; (4) Up to 45% of Se is in organic forms.  相似文献

Quantitative information on the abundance and biomass of metazoan meiofauna was obtained from samples collected at 15 deep-sea stations in the Eastern Mediterranean Sea (533–2400m). Meiofaunal abundance was compared to bacterial biomass and other environmental factors such as the total sedimentary organic matter content, the concentrations of the main biochemical classes of organic compounds (i.e. proteins, carbohydrates and lipids) and to ATP. To estimate the sedimentation potential of primary organic matter, sediment bound chloroplastic pigment equivalents (CPE) were assayed. Meiofaunal density was very low ranging from 4 ind.10cm−2 (Station A4, 1658m depth) to 290 ind.10cm−2 (Station A12, 636m depth). Nematodes were the numerically dominant taxon (68% of total meiofauna) and were usually confined to the top 6cm of the sediments. Total meiofaunal biomass ranged from 2.78μgC 10cm−2 (Station A4) to 598.34μgC 10cm−2 (Station 15A). There was a significant decrease in the density of metazoan meiofauna with water depth. Bacterial biomass largely dominated the total biomass (as the sum of bacterial and meiofaunal biomass) with an average of 73.2% and accounted for 35.8% of the living biomass (as ATP carbon) whereas meiofaunal biomass accounted only for 6.56%. Bacterial biomass was significantly related to the DNA concentrations of the sediment. A significant correlation between ATP concentration and CPE content was also found. No correlations were found between meiofauna, ATP and CPE, or between meiofauna and bacterial parameters. The significant relationship between meiofaunal density and the ratio of labile organic matter/total organic matter indicates that deep-sea meiofauna inhabiting an extremely oligotrophic environment (such as the Eastern Mediterranean) may be more nutritionally dependent upon the quality than on the quantity of sedimentary organic matter.  相似文献

The characteristic low-frequency oscillation of the sea surface temperature anomaly (SSTA) of ENSO related regions, Nino 1 + 2, Nino 3, Nino 4 and Nino West, and the Southern Oscillation index (SOI) is analyzed with the method of maximum entropy spectrum. Antarctic sea ice is divided into 4 regions, i. e. East Antarctic is Region Ⅰ (0°-120° E), the region dominated by Ross Sea ice is Region Ⅱ (120° E-120° W), the region dominated by Ross Sea ice is Region Ⅲ (120° W-0°), and the whole Antarctic sea ice area is Region Ⅳ. Also, the month-to-month correlation series of the sea ice with ENSO from contemporary to 5-years lag is calculated. The optimum correlation period is selected from the series. The characteristics and the rules obtained are as follows.1. There are a common 4-years main period of the SSTA of Ninos 1 + 2,3 and 4, a rather strong 4-years secondary period and a quasi-8-years main period of that of Nino West. A linear decrease in dissolved organic carbon and nitrogen with increasing salinity offshore from the Georgia coast suggests that organic nitrogen compounds contributed to coastal waters by rivers are stable during the period (2–3 months) of their transfer over the continental shelf. While the C/N ratio decreased with distance from shore, total dissolved organic nitrogen (DON), total amino nitrogen, and primary amino nitrogen showed similar relative decreases, suggesting that nitrogen is associated with refractory organic compounds. Measured amino nitrogen accounted for about 20% of the total DON, leaving about 80% of the organic nitrogen undefined.  相似文献

选择蒽、9-甲基蒽和9,10-二甲基蒽作为模型化合物,以李村河口区沉积物为培养基质,进行实验室好氧微生态培养,通过超声萃取-柱层析-气相色谱法测定目标污染物浓度随培养时间的变化,研究了它们在自然菌群作用下的生物降解行为。测定方法的基质加标回收率在66.20%~82.95%之间,相对标准偏差在3.34%~6.19%之间。6个月的好氧生物降解实验表明,沉积物中本底蒽由32.88 ng.g-1降到32.32 ng.g-1,降解率仅为1.70%;而在添加了蒽的沉积物样品中其浓度由170.40 ng.g-1降到151.36 ng.g-1,降解率高达11.17%;加入的9-甲基蒽和9,10-二甲基蒽的浓度也分别由133.84 ng.g-1和144.26 ng.g-1减少至122.52 ng.g-1和135.13 ng.g-1,降解率分别达到8.46%和6.33%。相同条件下三种有机污染物的不同降解速率表明,9,10位取代使得蒽在沉积环境中更加持久,自然降解速率更慢  相似文献

The present study investigates the differences between nutrient fluxes and particulate organic matter within an artificial reef system (AR) deployed in August 2002 off Faro (Algarve, Southern Portugal) and in a non-reef area (NRA), and how fluxes and suspended material may be affected by the hydrodynamic regime. Surveys to collect sediment cores, suspended/settled particles and overlying water samples were carried out by divers, from March (2006) to October (2007) in AR and NRA. Sediment cores and settled particles were collected to determine grain size, organic and inorganic carbon, nitrogen and phosphorus content. Overlying water and pore water samples were analysed for ammonium, nitrite, nitrate, phosphate, silicate, dissolved organic nitrogen, dissolved organic phosphorus and chlorophyll a. Results from the period studied showed that: (1) the benthic export of dissolved N, P and Si was 2–3 times higher at AR; (2) the particulate organic carbon (POC), nitrogen (PON) and phosphorus (POP) in suspended/settled particles were about 1.5 times higher at AR; (3) at both AR and NRA, the benthic export of dissolved N, P and Si, during a calm weather period, was 2–4 times higher than during or immediately after a storm event; and (4) at both sites, particulate organic compounds (POC, PON and POP) increased about 20 times during a storm event. These findings suggest that both the nutrients transport from sediment to water column and the quantity/quality of suspended/settled particles were highly dependent on the existence of reef structures and on the hydrodynamic regime.  相似文献

以安徽宣城官塘村第四纪红土剖面为例,研究了不同前处理方法对南方红土粒度测量的影响。结果表明:(1)南方红土中所含的有机质对粒度测量结果会产生明显影响,在进行粒度测量前处理时,必须加入双氧水去除有机质;(2)南方红土因淋溶强烈,碳酸钙含量极低,对粒度测量结果无影响,无需加入盐酸进行去钙处理。加入盐酸反而使得所测粒径偏粗;(3)与六偏磷酸钠相比,氢氧化钠作为分散剂因其强碱性产生的复杂化学反应,会导致所测得的粒度频率曲线峰态出现异常,且<1μm的黏粒组分含量减少,故不宜用作南方红土粒度测量的分散剂;(4)在对南方红土进行粒度测量时,建议使用H2O2+(NaPO3)6的前处理方法,即先使用双氧水去除有机质,再使用六偏磷酸钠作为分散剂,并用超声波清洗仪震荡后上机测试。  相似文献
