The New Zealand Waters Pollution Regulations 1963, referring to recreational water to which the public can gain access, states that the “coliform bacteria content shall not consistently exceed 1,000/100 ml”. This standard is now under revision, and the suggested new standard is a median faecal coliform index of 200/100 ml with a maximum value not exceeding 400/100 ml.

Recent literature supports the use of faecal coliform sub‐group bacteria as the best indicator of faecal pollution and reference is made to the membership of this sub‐group. Inherent inaccuracies of the Most Probable Number (MPN) method of assessing bacterial populations are pointed out, such as experimental variations with different cultural media, and possible errors in interpretation of results. In North America, there is evidence that an MPN for the total coliform group of 1000/100 ml indicates a faecal coliform sub‐group MPN of 200/100 ml, and epidemiological experience supports these values as standards.

We tabulate and discuss evidence that the usual ratio of faecal coliforms : coliform group bacteria is typically 1 : 1 in fresh and marine waters of the Wellington region, and that a more attainable standard would be 500 faecal coliforms/100 ml. In ratios quoted in this paper, ‘coliform group’ means the remainder of the coliform group excluding faecal coliforms. The World Health Organisation (1963) definition, of the entire group of coliforms, is not intended. The ratio is different in other parts of New Zealand; in Auckland, for example, the ratio is close to that found in the United States.  相似文献

Antibiotic resistant bacteria were studied as bio-indicators of marine polluted effluents during egg-laying in green turtles. A non-invasive procedure for sampling oviductal fluid was used to test for exposure of turtles to pollution in Ras Al-Hadd, Oman, which is one of the most important nesting beaches in the world. Each sample was obtained by inserting a 15 cm sterile swab gently into the cloacal vent as the sphincter muscle is relaxed and the cloacal lining is unfolded to the outside. Forty turtles were sampled. A hundred and thirty-two species of bacteria from 7 genera were isolated. The dominant isolate was Citrobacter. Among the isolates 60.6% were multiple resistant to 15 tested antibiotics. The dominant resistance to antibiotics was ampicillin followed by streptomycin and sulphamethoxazole. Sampling oviductal fluid for resistant bacteria to antibiotics is valuable way to assess exposure to polluted effluents during feeding and migratory in turtles. Polluted effluents using bacteria as bio-indicator may influence reproductive potential in this endangered species.  相似文献

本研究针对高温时期(6月、9月)广东沿海地区的香港牡蛎(Crassostrea hongkongensis)体内异养细菌总数和弧菌总数进行了深入的调查,并由添加单种抗生素(分别是:庆大霉素、呋喃唑酮、利福平、环丙沙星、恩诺沙星、氯霉素、复方新诺明)的培养基中分离得到310个菌株。进一步采用纸片扩散法(kirby-bauer法)针对常见的抗生素对此310个菌株进行药敏测试,了解不同来源菌株的耐药状况。结果表明310个菌株分属48个不同种属,主要是肠杆菌(Enterobacter)、弧菌(Vibrio)、芽孢杆菌(Bacillus)、肺炎克雷伯菌(Klebsiella)、希瓦氏菌(Shewanella)、不动杆菌(Acinetobacter)、假单胞菌(Pseudomonas)、短波单胞菌(Brevundimonas)、发光杆菌(Photobacterium)、黄杆菌(Tenacibaculum)、盐单胞菌(Halomonas)等。在9月份从牡蛎体内分离的异养细菌比6月份分离的异养细菌高出1~2个数量级(除台山外)。6月份从台山地区牡蛎分离的异养细菌数(2.6×10~6~5.8×10~6 cfu/g)最多,高出其他地区1~3个数量级。药敏测试结果显示牡蛎体内异养细菌对20种抗生素普遍存在抗性,尤其对青霉素、卡拉霉素、呋喃唑酮、阿莫西林、克林霉素及万古霉素表现出较高的耐药率。高温时期(6月、9月)分离的异养菌大多数为多重耐药细菌,其比例分别为84.18%和91.72%。本研究将为牡蛎养殖业的疾病控制以及水产养殖中细菌的耐药状况提供重要参考依据。  相似文献

Some experiments were carried out to explain thein situ phenomena that the number of coliform organisms decreased rapidly from estuaries to offshore, and also at deeper layer, and that the appearance of coliform types varied.In natural seawater, experimental results did not show thatEscherichia coli was extinct by self-purification or anti-biosis action of seawater, but it showed that this organism decreased mainly because of their starvation caused by lack of nourishment. Although the decreasing rate of bacterial density was delayed in enriched seawater, addition of nutrient even at the time of bacterial extinction promoted the appearance of variated form of this bacteria with floc formation.Flocculation of bacterial cells was influenced by quality and quantity of added nutrients. Temperature was shown to have an effect on the floc formation, but appearance of variated form in flocculated cells ofE. coli was not affected by temperature. Flocculated particles of coliform bacteria were adsorbed on suspended particles in seawater and precipitated rapidly. This phenomenon seems to be a cause of the rapid disappearance of coliform bacteria in coastal waters.In bottom deposits the coliform bacteria probably survive longer as physiologically variated forms when suitable nutrients were supplied.  相似文献

牡蛎体内及其养殖水体中细菌耐药性研究   总被引:1,自引:0,他引:1  
为进一步了解牡蛎体内外细菌的耐药性,对分离自阳江的香港牡蛎(Crassostrea hongkongensis)养殖水体及其内脏团的637个异养细菌进行了10种抗生素的药敏试验。采用纸片扩散法(Kirby-Bauer,简称K-B),参照NCCLS抗生素敏感试验操作标准,研究细菌的耐药概况。结果显示:健康牡蛎体内细菌对恩诺沙星和庆大霉素耐药率较低,发病牡蛎体内细菌对庆大霉素耐药率最低,次之为复合磺胺和恩诺沙星;水体细菌对氯霉素、恩诺沙星、环丙沙星耐药率较低,水体及体内细菌对呋喃唑酮耐药率最高,而对卡那霉素及青霉素耐药率则波动较大。结果还显示,不同来源的受试菌株多重耐药状况严重,来源于健康和病牡蛎的菌株在7月份的多重耐药率达到峰值,分别为66.7%和64.3%,养殖水体细菌多重耐药率峰值出现在4月份,最高达76.7%。总体来看,源于牡蛎养殖环境水体及其牡蛎体内的异养菌多重耐药比例较高,值得引起注意。而受试菌株对不同药物的抗性存在较大差异,且异养细菌对呋喃唑酮的耐药率明显高于对其它受测抗生素的耐药率。从时间动态上看,水体菌耐药率峰值出现于4月份,而牡蛎体内菌群耐药率峰值出现于7月份。  相似文献

In this study, we examined the bacterial accumulation in the filter feeder polychaete Branchiomma luctuosumGrube (Sabellidae). Analyses were performed on worm homogenates from 'unstarved' and 'starved' individuals, and seawater from the same sampling site (Gulf of Taranto, Western Mediterranean, Italy). Densities of culturable heterotrophic bacteria (22 degrees C), total culturable bacteria at 37 degrees C and halophilic vibrios at 22 and 35 degrees C were measured on Marine Agar 2216, Plate Count Agar and thiosulphate-citrate-bile-salt-agar (TCBS) plus 2% NaCl, respectively. Total and faecal coliforms as well as faecal streptococci were determined by the Most Probable Number method. Results showed that B. luctuosum is able to accumulate all the considered six microbiological groups with a higher efficiency for autochthonous bacteria. The analysis also indicated that bacterial groups differ in their resistance to digestion by B. luctuosum. Our data suggest the potential role of B. luctuosum as an useful bioindicator to assess low levels of microbiological water pollution as well as bioremediator of microbial polluted waters.  相似文献

为了探究北部湾养殖区域香港牡蛎(Crassostrea hongkongensis)体内的异养细菌和弧菌数量及其耐药概况变化, 对不同养殖场牡蛎体内的异养细菌进行分离培养, 并统计其数量, 通过药敏纸片扩散等方法研究了细菌的耐药状况。结果显示: 牡蛎在高死亡率养殖环境中体内的异养细菌[(8.6±0.4)×106CFU·g-1]和弧菌[(9.5±0.4)×105CFU·g-1]数量较高, 在中死亡率环境中体内的异养细菌[(6.9±0.2)×106CFU·g-1]和弧菌[(4.5±0.6)×105CFU·g-1]数量次之, 在低死亡率养殖环境中体内的异养细菌[(3.3±0.1)×106CFU·g-1]和弧菌[(2.5±0.6)×105CFU·g-1]数量最低。耐药细菌主要为革兰氏阴性菌, 对β-内酰胺类(青霉素)、糖肽类(万古霉素)的耐药率较高, 对四环素类(四环素、多西环素)的耐药率次之, 对氨基糖苷类(链霉素、庆大霉素、妥布霉素、新霉素)、大环内酯类(红霉素)、喹诺酮类(诺氟沙星、环丙沙星、氧氟沙星、恩诺沙星)的耐药率较低。在高死亡率环境中牡蛎体内的多重耐药菌占79.7%, 其耐药谱型(48种)较广; 在中度死亡率环境中牡蛎体内的多重耐药菌占66.2%, 其耐药谱型为30种; 在低死亡率环境中牡蛎体内的多重耐药菌占58.4%, 其耐药谱型为17种。本文探究了牡蛎死亡率与其体内异养细菌数量和细菌耐药性的关系, 结果显示牡蛎在高死亡率环境中体内的耐药细菌数量多、耐药谱型较广, 低死亡率环境中牡蛎体内的耐药细菌数量较少, 异养细菌数量与牡蛎死亡率呈正相关关系, 两者相关系数为0.996。  相似文献

The aim of this study was to test whether silver nanoparticles (Ag-NPs) released into estuarine environments result in increased antibiotic resistance amongst the natural bacterial population in estuarine sediments. A 50-day microcosm exposure experiment was carried out to investigate the effects of Ag-NPs (50 nm average diameter) on the antibiotic resistance of bacteria in sediments from an estuary in southwest England. Experimental microcosms were constructed using 3.5 kg sediment cores with 20 l of overlaying seawater treated with (final) Ag-NPs concentrations of 0, 50 or 2000 μg l−1 (n = 3). Sediment samples were screened at the end of the exposure period for the presence of bacteria resistant to eight different antibiotics. Multivariate statistical analyses showed that there was no increase in antibiotic resistance amongst the bacterial population in the sediment due to the dosing of the microcosms with Ag-NPs. This study indicates that, under the tested conditions, Ag-NPs released into the coastal marine environment do not increase antibiotic resistance among naturally occurring bacteria in estuarine sediments. These results contrast previous findings where antimicrobial effects of Ag-NPs on key bacterial species in laboratory experiments have been demonstrated, and reasons for this are discussed. The negligible effects demonstrated on bacterial populations under the selected estuarine conditions, provide important information on no observed effect concentrations (NOECs) for environmental regulation.  相似文献

本文对秦山核电站邻近水域表层海水中大肠菌群和粪大肠菌群的分布作了春、夏、秋、冬四季的初步调查分析。结果表明,大肠菌群和粪大肠菌群的分布范围为0~3500个/dm~3,并在一年四季中都能检出大肠菌群和粪大肠菌群。其分布趋势为沿岸高于外侧,退潮大于涨潮,南部多于北部。细菌的数量随水温、盐度、潮汐等环境因子的变化而变化。  相似文献

A baseline groundwater quality survey of 120 household wells in an unsewered area at Yaldhurst, near Christchurch, was carried out between September and December, 1976. Ammonia‐N levels were below the detection limit (0.02 g m‐3) in all samples. Nitrate‐N, conductivity, and chloride levels tended to vary according to well depth and location, being lower in the deeper wells and in wells located in an area of shallow, stony soils. Approximately 33% of the wells contained coliform bacteria, faecal coliform bacteria or faecal streptococci. A subset of 25 wells was sampled fortnightly from January to August 1977. From January to June, mean nitrate‐N and conductivity levels remained stable at approximately 1.5 g m‐3 and 11.2 mS m‐1 respectively. An increase in these levels to 3.3 g m‐3 and 15.8 mS m‐1 in July‐August corresponded with a rise in the water table. Leaching losses from soils probably accounted for most of the nitrate entering the Yaldhurst aquifers, the contribution by septic tank systems being estimated at 20–30%. No direct evidence of contamination of the wells by septic tank effluent was found, although 23 of the 25 wells exhibited intermittent contamination by indicator bacteria throughout the 8 month sampling programme. Although a localised health hazard may exist, septic tank systems in the area were not considered likely to constitute a health threat to the confined aquifers underlying Christchurch city.  相似文献

Distribution of coliform bacteria was surveyed monthly during July-1968 through December-1969 in Shimizu Harbor and Orido Bay. Among three significant rivers flowing into the coastal region, the water of Tomoe River was most microbiologically polluted. Throughout the period of investigation horizontal distribution of the bacterial density was highest at the estuary of the Tomoe River, and its density decreased toward Shimizu Harbor and Orido Bay.It was shown that the bacterial density decreased from surface to deeper layer. High density of bacteria was observed in bottom deposits at winter. The bacterial distribution might be disturbed by current caused by geographical features.Various types of coliform group were isolated in the surveyed areas. All types ofE. coli, E. freundii, A. aerogenes and Irregular appeared in the area of estuaries, and only limited types occurred in offshore.Bacterial number increased from upper stream toward the mouth of each river. The bacterial density differed from river to river. In the rivers flowing through the city the bacterial density changed with water temperature, whereas in the river running through agricultual district it might be affected with the rainfall.  相似文献

Faecal coliform levels were determined for sea water and shellfish flesh from farms of the green mussel, Perna canaliculus, in Pelorus Sound, Marlborough Sounds, New Zealand. Simple linear models were developed to relate logarithmically transformed data on the faecal coliform levels to measures of fresh water entering the mussel beds. These models support guidelines to restrict harvesting following periods of heavy rain in order to comply with shellfish sanitation standards. Alternative guidelines are suggested based on the volume of river water entering the sound or salinity.  相似文献

为了解海水浴场中大肠杆菌耐药性状况及其耐药基因分布,以大连星海浴场近岸海水样品为研究对象,分离鉴定得到41株大肠杆菌,采用K-B药敏纸片法检测其对9种常用抗生素的耐药性,并对耐药菌株中的耐药基因进行检测。结果表明:41株受试大肠杆菌菌株对四环素的耐药率最高(46%),而对氨曲南的耐药率最低(5%),大肠杆菌的多重耐药率达到34%。采用PCR方法对41株分离菌株进行四环素耐药基因检测,结果显示,仅四环素耐药基因tetA和tetB被检出,其他4种类型四环素耐药基因未被检出。大肠杆菌基因组与质粒DNA中tetA的检出率分别为34%和29%;tetB的检出率分别为2%和12%。相关性分析结果表明大肠杆菌的四环素耐药表型与四环素耐药基因的阳性检出率显著相关。  相似文献

抗生素的滥用是当前水产养殖业发展及食品安全面临的重大威胁。贻贝体内含有丰富的附生菌群,但目前尚不清楚贻贝附生菌群在抗生素残留背景下的动态变化和耐受性特征。为此进行了厚壳贻贝中抗生素耐受细菌多样性研究。将厚壳贻贝暴露于含有青霉素、链霉素和卡那霉素的养殖水体中,之后采用超声法获取含贻贝软体部组织表面附着菌群的超声液,经Zobell 2216E液体培养基培养后,将组织超声液和培养液一并经16S rDNA高通量测序来分析厚壳贻贝微生物的群落组成。结果表明,超声处理后获得的微生物主要为拟杆菌门(Bacteroidetes)、变形菌门(Proteobacteria)和疣微菌门(Verrucomicrobia)。多重抗生素的处理可以显著下降贻贝体内微生物的OTU数,对微生物群落结构有明显影响。经264h恢复养殖后,在门水平上观察到贻贝体内微生物群落可以恢复重建,表明其具有一定的韧性。研究结果将为今后进一步探索宿主相关微生物群落构建机制及特殊抗性微生物奠定基础。  相似文献

Tsunami, the natural disaster, which occurred on December 26, 2004 in the Indian Ocean, caused severe damage to mankind in the coastal areas. Total loss of life and economic loss because of this disaster have been estimated by various agencies but its effect on microbial density has not been probed. With our previous results on microbial populations in four locations of the Chennai coast of the Bay of Bengal, India in the pre‐tsunami period, the change in microbial populations was studied after the tsunami at regular intervals in the same locations. Coastal sediment and seawater samples were collected from four different locations after 5–10 h and thereafter at intervals of every 7 days up to 28 days post‐tsunami. Bacteria, fungi and actinomycetes were isolated from the marine samples by serial dilution on respective media. Before the tsunami, the bacterial population was higher in seawater samples than the sediments, whereas fungi and actinomycetes were recorded only in the sediments. The microbial population remarkably increased 5–10 h post‐tsunami in all the marine samples irrespective of the location. However, it slowly declined in the subsequent days and became similar to that of the population recorded before the tsunami. The population of gram‐positive bacteria increased whereas the gram‐negative bacterial population decreased after the tsunami. Further, populations of pathogenic bacteria such as coliform and vibrios did not increase after the tsunami. It has been observed that the increase in populations of bacteria and actinomycetes even after 28 days of tsunami may be due to the introduction of foreign microorganisms that developed the ability to survive in the extreme environment by exhibiting special characteristics such as pigmentation and production of exopolysaccharides.  相似文献

磺胺类、四环素、喹诺酮类和多粘菌素抗生素是医学与养殖业中的常用抗生素,其中多粘菌素在畜牧养殖业中常作为促长剂添加。调查分析了这4类抗生素的常见耐药基因在东海温州近海水域的分布情况。利用抗性平板筛选温州近海水域对磺胺类、四环素类及喹诺酮类抗生素耐药的可培养细菌分离株,对耐药分离株通过PCR方法进行目标耐药基因检测;对目标耐药基因阳性的耐药分离株进行多粘菌素耐药基因MCR检测以及采用微量二倍稀释法测定对上述4类抗生素的耐药性。结果表明,利用抗性平板筛选获得可培养耐药分离株1 605株,从中筛选到目标耐药基因阳性的51株,检出率为3.18%,包括含磺胺类耐药基因35株、含四环素耐药基因36株、含喹诺酮耐药基因17株、含多粘菌素耐药基因2株; 51株耐药菌归于7个菌属,其中气单胞菌属(Aeromonas sp.) 43株,占比84.31%,枸橼酸杆菌属(Citrobacter sp.)和希瓦氏菌属(Shewanella sp.)各2株,埃希氏菌属(Escherichia sp.)、类芽孢杆菌属(Paenibacillus sp.)、Thalassobius sp.及Amphritea sp.各1株;上述2株非常见分离株不统计在内,常见的49株均对四环素耐药, 41株对多粘菌素耐药,对磺胺类和喹诺酮类耐药的分离株均为34株;同时,发现23株对4类抗生素均产生耐药, 16株对多达3种抗生素耐药,多重耐药现象普遍存在。同时,鉴定出MCR-3阳性的条件致病菌维氏气单胞菌和豚鼠气单胞菌各1株。综上,温州市近海水域中含有常见耐药基因的耐药分离株检出率较低,但同一耐药基因的细菌分布多样,对磺胺类、四环素类、喹诺酮类及多粘菌素类等4类抗生素表现出较强的耐药性,多重耐药现象明显。研究结果将为评估温州近海水域的细菌耐药性风险,规范抗生素的使用提供科学依据。  相似文献

为解决人口、资源、环境等人类面临的重大问题,海洋作为地球上尚未充分开发的重要疆域而备受重视。中国以海水养殖业为基础的“蓝色农业”在缓解人口增长对陆地农业的压力,促进沿海地区经济的发展中正发挥着重要作用。然而,近10年来,世界范围内海水微生物流行性和暴发性病害相继发生,严重制约了海水养殖业的健康发展(相建海,2003)。为了防治细菌性病害,大量的抗生素药类被应用于养殖过程的各个阶段,巨大的选择压力导致了海水养殖水域耐药细菌的种类和数量的显著增加,养殖生物耐药病原菌病害的发生也越来越频繁,微生物病害防治的难度越来越大。更严重的问题是这样培养的水产养殖品往往携带耐药细菌,通过食物链传递,将对人类健康和生命安全直接构成威胁(Sorum et al.,2002)。一些海产生物病原菌本身也是人类病原菌,如一些海洋弧菌(French et al.,1989;Wong et al.,2000),这些水陆两栖人畜共患病原菌耐药性的获得及这种病害的发生和流行将会给人类带来灾难性的后果。另外,由于大多耐药因子的可传播性,海水养殖环境中正常菌群的耐药性同样是一个不可忽视的严峻问题(Sorum et al.,2002)。大多数抗性决定子(耐药基因)是在可移动和传递的遗传载体,如质粒或接合转座子上,可在不同菌株甚至是不同种属细菌间相互传递(Ferber,1998;OBrien,2002)。再加上养殖水体的半封闭性(池养中的定期或不定期的换水)或全开放性(滩涂或近海的网箱或筏式养殖),耐药细菌有可能被潮汐和海流带入近岸的非养殖区及海滨旅游区,通过菌间遗传物质的传递,耐药基因有可能被传入环境中的人体病原菌,大大增加了人体耐药病原菌病害发生和流行的可能性及危害程度(Miranda et al.,2001;Thayumanavan et al.,2003)。大量抗生素的使用还改变了养殖海域及临近自然海区细菌群落的结构及种类和功能多样性,破坏了生态平衡和海洋正常的生命过程(Chelossi et al.,2003)。养殖水域作为可能的抗药因子的孳生地、储藏库和扩散源(Miranda et al.,2002;Rhodes et al.,2000),已引起了全球性的关注。世界上一些水产养殖国家相继开展了养殖水体耐药细菌的调查研究,并有一些国家已对养殖中抗生素的种类、使用方法、使用剂量、残存量及检测标准进行了行政化的规范和管理(李爱华,2002)。目前我国对海水养殖水体细菌耐药危害的认识明显不足,许多需要进行的研究尚未开展,为更有效地防治耐药细菌病害,避免耐药基因在海洋环境中的广泛传播扩散,保护海洋生态环境及资源,提高养殖产品卫生质量,保护海滨旅游环境及人民群众身体健康,在我国开展海水养殖环境耐药细菌及其抗性机理的调查研究已是刻不容缓,这项工作的重点应在于查清耐药因子的源及其传播途径和动态,并在此基础上寻找治理的科学办法,以达到防患于未然的目的。20世纪滥采乱捕导致的近海渔业资源的严重衰竭是一个非常深刻的教训,希望这种急功近利、涸泽而渔的状况不会再度在海产养殖业中出现。我们应该把海洋细菌耐药性的研究提升到直接关系到人类生存和发展的环境和资源问题的高度来对待,避免海洋成为耐药因子孳生、繁衍、传播和扩散的潘多拉魔盒。  相似文献

Mangroves are coastal ecosystems, found in tropical and subtropical regions around the world. They are found in the transitional zones between land, sea, and rivers. Petroleum hydrocarbons are the most common environmental pollutants, and oil spills pose a great hazard to mangroves forests. This research was focused on the isolation and characterization of crude oil‐degrading bacteria from mangrove ecosystems at the Persian Gulf. Sixty‐one crude oil‐degrading bacteria were isolated from mangrove samples (plant, sediment, and seawater) that enriched in ONR7a medium with crude oil as only carbon source. Some screening tests such as growth at high concentration of crude oil, bioemulsifier production, and surface hydrophobicity were done to select the most efficient strains for crude oil degradation. Molecular identification of strains was carried out by amplification of the 16S rRNA gene by PCR. The results of this study were indicated that the quantity of crude oil‐degrading bacteria was higher in the root of mangrove plants compare to other mangrove samples (sediment and seawater). Also, identification results confirmed that these isolated strains belong to Vibrio sp. strain NW4, Idiomarina sp. strain BW32, Kangiella sp. strain DP40, Marinobacter sp. strain DW44, Halomonas sp. strain BS53, and Vibrio sp. strain DS35. The application of bioremediation strategies with these bacteria can reduce crude oil pollution in this important marine environment.  相似文献
