盐藻胞浆hsp70 cDNA的克隆及其mRNA的诱导表达   总被引:2,自引:0,他引:2  
用cDNA末端快速扩增方法克隆得到了一个2652bp的盐藻(Dunaliella salina)胞浆hsp70 cDNA全长,编码着650个氨基酸残基的多肽。推导的氨基酸序列有2个ATP酶结合位点和一个多肽结合区。由克隆得到的盐藻cDNA推导的氨基酸序列,经GenBank blast后与数据库中胞浆hsp70序列一致。与莱茵衣藻(Chlamydomonas reinhardtii)、人、小麦(Triticum astivum)和啤酒酵母(Saccharamyces.cerevisiae)胞浆hsp70的序列一致性分剂为83%、80%、81.5%和80.5%。Northern印迹显示,90min后,mRNA量在40℃热休克时是光诱导时的3倍。光诱导则使hsp70 mRNA积累相对迟缓。结果表明,所克隆得到的hsp70基因蛋白产物定位在盐藻细胞浆中,光诱导和热休克均可以使hsp70 mRNA水平明显增高,但以热休克为显著。  相似文献

为深入分析热休克蛋白响应胁迫的分子机制,实验以宽体沙鳅(Botia reevesae)为研究对象,利用同源克隆和cDNA 末端快速扩增(rapid amplification of cDNA ends,RACE) 技术克隆得到宽体沙鳅热休克蛋白70(BR-HSP70) 的cDNA 全长。结果发现,BR-SP70 cDNA全长为2,371bp,包含1,947 bp的开放阅读框(opening reading frame,ORF),102 bp 5’-非编码区(untranslated region,UTR)和322 bp 3’-UTR等。通过序列同源性比对发现,BR-HSP70 cDNA与团头鲂(Megalobrama amblycephala)和猪(Sus scrofa)的同源性分别为98%及83%,且ORF编码的649个氨基酸中含有HSP70家族的家族信号标签、N-糖基化位点及EEVD等能位点等保守序列。上述结果表明,本研究所获的基因为宽体沙鳅 HSP70基因。实时荧光定量PCR 分析发现,氨氮胁迫和嗜水气单胞菌(Aeromonas hydrophila)侵染均会显著上调宽体沙鳅鳃、肝脏及肾脏HSP70 mRNA的表达,表明HSP70基因在宽体沙鳅应对环境胁迫中发挥了重要的抗应激作用。  相似文献

利用RT-PCR和快速扩增cDNA末端(rapid amplification of cDNA ends,RACE)技术首次克隆了宽体沙鳅(Botia reevesae)-肌动蛋白基因的cDNA全序列,该序列全长为1795bp,由长100bp的5非翻译区(untranslated region,UTR)、570bp的3非翻译区和1125bp的开放阅读框(open reading frame,ORF)组成,编码375个氨基酸。宽体沙鳅-actin氨基酸序列包含1个糖基化位点、9个N-豆寇酰化位点、3个actin信号位点等主要结构区域。PSI-BLAST比对表明,宽体沙鳅-actin氨基酸与真鲷、罗非鱼、虹鳟等鱼类同源性达99%。NJ法系统进化分析显示宽体沙鳅-actin首先与鲢聚在一起,然后与、尖头等鱼类聚在一起。荧光定量PCR检测-actin基因在宽体沙鳅脑、鳃、心脏、肝、胃等12个组织的表达无显著差异(P<0.05),具有良好的稳定性。  相似文献

采用RT-PCR及RACE法从斜带石斑鱼Epinephelus coioides肝胰脏克隆得到胰蛋白酶原(trypsinogen,TRY)与淀粉酶(amylase,AMY)基因cDNA全序列.斜带石斑鱼肝胰脏TRY基因cDNA全长911 bp,其中5非翻译区(5'-UTR)为55bp,3'-UTR为127bp,开放阅读框(ORF)为729bp,编码242个氨基酸,包含所有丝氨酸蛋白酶中共有的高度保守的催化活性中心.序列一致性分析发现,斜带石斑鱼与牙鲆Paralichthys olivaceus、金头鲷Sparus aurata、鳎Solea senegalensis、石鲽Pleuronectes bicoloratus的TRY序列相似性高达86.8%-89.70%,与人Homo sapiens小鼠Mus musculus、斑马鱼Danio rerio的TRY相似性较低为59.9%-64.5%.斜带石斑鱼AMY基因cDNA全长1657bp,其中5'-UTR为41bp,3'-UTR为77 bp,ORF为1539bp,编码512个氨基酸,包含与哺乳动物α-AMY二级结构相似的8个α旋和8个β折叠.序列一致性分析发现,斜带石斑鱼与澳洲肺鱼Neoceratodus forsteri、美洲拟鲽Limanda americanus、大西洋鲑Salmo salar、斑马鱼AMY基因序列相似性高达82.4%-91.8%,与人、小鼠、鸡G.Gallus的AMY基因相似性较低为70.1%-72.3%.斜带石斑鱼TRY和AMY基因cDNA全序列的成功克隆为进一步研究其表达调控机理及研发有效提高其表达水平的饲料添加剂奠定基础.  相似文献

采用同源克隆和RACE-PCR(Rap id amp lification of DNA ends-PCR)技术,从黄鳍鲷Acanthopgrus latus中克隆了白细胞介素1β(interleuk in-1,βIL-1β)基因的全长cDNA序列。黄鳍鲷IL-1β的cDNA全长共1 242 bp,5端非编码区(UTR)为121 bp,3端非编码区(UTR)为342 bp,开放阅读框(open read ing fram e,ORF)为762 bp,编码一条253个氨基酸残基的多肽,分子量约为28.5kDa,理论等电点为5.55,含有2个潜在的糖基化位点。同源性分析表明与其它鱼类和脊椎动物的IL-1β序列具有较高的同源性。利用软件S ignalP分析表明它不存在信号肽序列,氨基酸序列中不存在IL-1β转换酶(interleuk in-1βconverting enzym e,ICE)剪切位点,推导可能成熟肽为从第85位氨基酸开始共169个氨基酸残基的多肽,分子量约为18.9kDa。将此成熟肽序列克隆入pQE30质粒构建表达载体pQE30-IL1β,并在大肠杆菌Escherichia coliM15(pREP4)中进行诱导表达,获得了分子量约为21 kDa的特异性融合蛋白。  相似文献

采用同源克隆和cDNA末端快速扩增技术(RACE)克隆了浙江枝吻纽虫热休克蛋白70(DzHSP70)基因全长cDNA序列,并利用荧光定量PCR方法,分析了该基因在重金属胁迫下的表达特征。结果表明,DzHSP70cDNA全长2827bp,包括294bp的5’UTR、628bp的3’UTR和编码634个氨基酸残基的1905bp的开放阅读框。HSP70家族的3个签名序列以及细胞质特异性调控基序EEVD等特征元件在DzHSP70中高度保守。重金属离子Fe3+、Pb2+和Cd2+均可上调该基因的表达,其中Pb2+的毒理效应最强。研究结果表明DzHSP70可能参与了机体对于重金属的解毒过程。  相似文献

热休克蛋白70(HSP70)是生物体内一种重要的机体与细胞保护性蛋白。本文利用RT-PCR以及RACE技术首次克隆获得太平洋真宽水蚤(Eurytemora pacifica)HSP70简称Ep.HSP70 c DNA的全长序列,序列全长为2252bp(KY807149),开放阅读框(ORF)长1947bp,编码649个氨基酸,5′端99bp,3’端206bp;预测蛋白分子量为70.81k Da,等电点为5.16,为一种亲水性蛋白,不存在信号肽及跨膜区,含有丰富的α螺旋结构(37.60%),β折叠(18.80%)。同源氨基酸序列比对发现,与其他甲壳动物的同源基因保守性较高,尤其是HSP70家族典型的结构位点序列在甲壳类动物中具有高度保守性。系统进化分析表明,太平洋真宽水蚤和安氏伪镖水蚤(Pseudodiaptomus annandalei)进化关系最近;桡足类种内同源性要高于虾蟹类,与虾类同源性高于蟹类。荧光定量数据分析表明,不同浓度铜、镉、锌胁迫下太平洋真宽水蚤HSP70基因表达水平具有显著的时间效应与浓度效应的特征,三种金属对Ep.HSP70抑制效应呈现CuCdZn的趋势。Ep.HSP70基因的成功克隆及金属胁迫下的表达分析为深入研究HSP70蛋白生物学功能具有重要意义。  相似文献

本研究利用RACE-PCR技术获得企鹅珍珠贝(Pteria penguin)一个Dmrt2基因cDNA的全长序列,通过荧光定量PCR分析Dmrt2基因在各组织中的表达特征,以及在早期卵巢、成熟期卵巢、早期精巢、成熟期精巢和排放期精巢中的表达变化。结果表明,Dmrt2基因全长1 257bp,其中开放阅读框(open reading frame,ORF)为951 bp,编码316个氨基酸,5′非编码区(untranslated region,UTR)为52 bp,3′UTR为254 bp,第20位到第73位氨基酸为DM结构域。预测其分子质量为36.61ku,等电点为9.80。氨基酸序列比对显示该企鹅珍珠贝Dmrt2基因与黑蝶真珠蛤(Pinctada margaritifera)和马氏珠母贝(Pinctada martensii)的Dmrt2基因同源性(identity)最高,分别为46.0%和45.7%。其中在DM结构域高度同源。荧光定量PCR分析组织表达特征显示,Dmrt2在企鹅珍珠贝的外套膜、鳃、消化盲囊、足、精巢和卵巢均有表达,其中在精巢中表达量最高(P0.05),足为其次,在闭壳肌中没有检测到Dmrt2表达。对性腺发育不同时期的表达量分析发现,Dmrt2在发育早期和成熟期卵巢中表达量都很低,在发育早期精巢中表达量较高,在成熟期精巢检测到最大表达量(P0.05),到精巢排放期表达显著下降,推测Dmrt2可能与企鹅珍珠贝精巢的发育有关,可能参与了企鹅珍珠贝雄性性别分化和性腺发育这一生理过程。  相似文献

CREB(cAMP response element binding protein)是一种真核生物中广泛存在的调控因子。为了探讨企鹅珍珠贝(Pteria penguin)Creb基因的序列和表达特征,为进一步阐述Creb的生理功能提供科学依据,本研究利用RACE-PCR技术克隆到企鹅珍珠贝一个Creb基因的全长序列,分析其理化性质和进化地位;利用荧光定量PCR分析Creb基因在各组织中的表达特征。结果表明,该企鹅珍珠贝Creb基因的c DNA全长为1484 bp,其中开放阅读框(ORF)为1071 bp,编码356个氨基酸,5′非编码区为218 bp,3′非编码区为195 bp,C端包含一个碱性亮氨酸拉链结构(basic region leucin zipper,b ZIP)。预测其分子质量为39.49 k D,等电点为4.43。序列比对显示该企鹅珍珠贝Creb基因与欧洲平牡蛎(Ostrea edulis)和太平洋牡蛎(Crassostrea gigas)的Creb2同源性(identity)最高,分别为46.3%和46.1%,故命名为pp Creb2;系统进化树分析结果与传统形态学分类结果相吻合。荧光定量PCR分析显示,pp Creb2在企鹅珍珠贝的各个组织中组成性表达,其中在足中表达量最高,鳃中其次。  相似文献

企鹅珍珠贝(Pteria penguin)分布在我国广西合浦,广东湛江、惠阳,台湾和海南,主产地为海南,日本、菲律宾等地也有出产;它利用足丝固着在低潮线以下的浅海岸石或珊瑚礁上生活。其成体壳高18cm,大者可达25cm。贝壳近斜方形,略似斧状,背缘约平直,腹缘弧形,前缘和腹缘连成一大弧状,后缘向后腹端倾斜延伸。前耳小,后耳长,左壳平,右壳凸。壳表黑色,鳞片细密。壳内面有虹彩色珍珠光泽。足丝发达,成体足丝呈树枝状[1]。因其具有生长快、个体大、抗病力强等优点,是养殖大型附壳珍珠的理想母贝。目前这一…  相似文献

The body condition of altricial seabird chicks is directly related to the amount of food received from their parents, which depends on feeding conditions at sea. An index of body condition is therefore a useful tool for assessing variation in food availability between different breeding colonies. Quantile regression techniques were used to develop a body condition index for African penguin Spheniscus demersus chicks, based on maximum and minimum growth relative to structural size. This is considered the most appropriate index for comparing the body condition of groups of chicks, varying in size and of unknown age, between breeding colonies. Based on this study, recommendations are made with regard to morphometric measurements, sample size and the sampling interval required to compare effectively the condition of African penguin chicks between breeding colonies throughout their distributional range.  相似文献

本文通过企鹅珍珠贝游离珠插核过程中的一系列对比实验,首次探讨了施术贝死亡过高的原因.发现术前处理时间太长造成的贝体虚弱会增加死亡率;施术伤口对贝体造成的生理不适如能及时得以恢复,不会因此发生大规模的死亡;偏小的核位、较厚的核位皮层肌肉以及由之产生的较强的排异反应会严重影响企鹅珍珠贝施术贝的成活率;休养时提倡先室内休养10d左右再移至海区,且不必添加昂贵的抗生素.  相似文献

在雷州流沙港湛江海洋大学珍珠研究室实验基地分别于春、夏和秋季进行了企鹅珍珠贝游离珠植核实验。其中,春季插核的死亡率最低,为15.1%--16.2%,留核率最高,为10%左右:而夏季插核的死亡率最高,为63.56%,留核率也仅为2.22%:秋季插核的留核率最低,仅收珠1颗,留核率为0.35%,死亡率也较高,为54.4%。结果表明,春季是企鹅珍珠贝较适宜的插核季节。  相似文献

The jackass penguin, endemic to southern Africa, is the only member of the Sphenisciformes now breeding in Africa. Its breeding distribution extends from Sylvia Hill, South West Africa (Namibia), to Bird Island, Algoa Bay, South Africa. The non-breeding range extends from Sette Cama, Gabon, to Inhaca Island, Moçambique. Jackass penguins do not usually occur far off shore. The minimum total breeding population in 1978/79 was estimated to be approximately 134 000 birds. Numbers halved between 1956 and 1978 alone as a result of collapses in the South and South West African pilchard populations. Numerical decreases at Possession and Dassen Islands were particularly severe. Prior to 1956 numbers had also decreased, primarily because of large collections of eggs. Sealing activities and guano harvests may also have influenced trends. Only colonies at the periphery of the breeding distribution have increased numerically in recent times, and such increases appear to be related to the availability of suitable prey. Although the species breeds in captivity and is unlikely to become extinct, its continued survival in the wild probably relies on the continued good state of the pelagic fish resources.  相似文献

Nearly 36,000 vertical temperature profiles collected by 15 king penguins are used to map oceanographic fronts south of New Zealand. There is good correspondence between Antarctic Circumpolar Current (ACC) front locations derived from temperatures sampled in the upper 150 m along the penguin tracks and front positions inferred using maps of sea surface height (SSH). Mesoscale features detected in the SSH maps from this eddy-rich region are also reproduced in the individual temperature sections based on dive data. The foraging strategy of Macquarie Island king penguins appears to be influenced strongly by oceanographic structure: almost all the penguin dives are confined to the region close to and between the northern and southern branches of the Polar Front. Surface chlorophyll distributions also reflect the influence of the ACC fronts, with the northern branch of the Polar Front marking a boundary between low surface chlorophyll to the north and elevated values to the south.  相似文献

借鉴马氏珠母贝插核技术培育企鹅珍珠贝(Pteria(Magnaxicula)pengui Roding)游离珠时,死亡率为23%,留核率为11.1%,死亡高峰出现在施术后的第2~7天,脱核高峰出现在施术后第2~10天,生理状态在施术后17d左右进入稳定期。70~90d后基本恢复,可结束休养进入珍珠育成。另外,为提高成珠率和成活率,植核用贝要经人工养殖驯化,采用开放式与笼养相结合的养殖方式;术后可采用室内与海区相结合的休养方式。  相似文献

The interdependence between the seismo-acoustic properties of a marine sediment and its geotechnical/physical parameters has been known for many years, and it has been postulated that this should allow the extraction of geotechnical information from seismic data. Though in the literature many correlations have been published for the surficial layer, there is a lack of information for greater sediment depths. In this article, a desktop study on a synthetic seafloor model illustrates how the application of published near-surface prediction equations to subsurface sediments (up to several tens of meters burial depth) can lead to spurious predictions. To test this further, acoustic and geotechnical properties were measured on a number of sediment core samples, some of which were subjected to loading in acoustically-equipped consolidation cells (oedometers) to simulate greater burial depth conditions. For low effective pressures (representing small burial depths extending to around 10 meters subsurface), the general applicability of established relationships was confirmed: the prediction of porosity, bulk density, and mean grain size from acoustic velocity and impedance appears generally possible for the investigated sedimentary environments. As effective pressure increases through, the observed relationships deviate more and more from the established ones for the near-surface area. For the samples tested in this study, in some instances increasing pressure even resulted in decreasing velocities. There are several possible explanations for this abnormal behavior, including the presence of gas, overconsolidation, or bimodal grain size distribution. The results indicate that an appropriate depth correction must be introduced into the published prediction equations in order to obtain reliable estimates of physical sediment properties for greater subsurface depths.  相似文献

Specific properties of the interannual sea level variations and annual tides in the Northwestern Pacific were studied. Several tide stations were monitored. The monthly mean sea level for the year of 1995 was analyzed at each tide station. A seismic event in 1995, some tectonic activity around the subject area, and the Kuroshio (the oceanic western boundary current) may possibly contaminate results which would have occurred from the astronomical annual tide alone.  相似文献
