对一株分泌热稳定κ-卡拉胶酶印尼热泉菌进行了种属鉴定, 并采用响应面法对该菌发酵产酶条件进行了优化。鉴定结果表明, 该菌株属于芽孢杆菌属(Bacillus), 命名为Bacillus sp. Car19(GeneBank: KT865196)。发酵条件优化结果显示, 9个环境因子影响Bacillus sp. Car19产酶量。其中影响Bacillus sp. Car19产酶量的三个主要因素分别为培养温度、培养基中Cu2+浓度和培养基中NaCl浓度。综合次要因素对Bacillus sp. Car19产酶影响, Bacillus sp. Car19最佳产酶发酵条件为: 培养温度52.31℃、Cu2+浓度6.93 mmol/L、NaCl浓度37.03 g/L, 培养基pH为6, 接种量1%, 培养时间36h, 半乳糖浓度0.3 g/L,硝酸铵浓度7g/L, 卡拉胶浓度0.5g/L。优化后发酵上清液酶活力达到15.21 U/mL, 与优化前相比提高了1.5 倍。  相似文献

采用体外测定细菌浓度、胞外产物(ECP)蛋白含量和蛋白酶活力的方法,进行了鳗弧菌M3菌株在2216E培养基中的培养条件研究。结果表明,该菌株用固体培养基培养至24h左右,可得到较高的菌体浓度、ECP蛋白含量和蛋白酶活力。采用响应面分析方法设计实验,用SAS统计软件分析数据,得到NaCl浓度、pH值和温度对菌体生长及蛋白酶产量影响的回归模型。在2216E培养基的基础上,添加不同氮源、碳源物质以及不同浓度蛋白胨进行生长研究。结果表明,胰大豆蛋白胨能促进菌体生长及ECP蛋白分泌;NH4Cl与酪蛋白水解物可抑制蛋白酶的产生;牙鲆肌肉匀浆对菌体、ECP蛋白产量和蛋白酶产生有不同程度的促进作用;培养基中蛋白胨浓度为4%时菌体量与ECP蛋白含量达最高值,在蛋白胨浓度为2%时蛋白酶分泌量已稳定;1%的葡萄糖、蔗糖、甘油均能显著地提高菌体及ECP蛋白产量,却抑制了蛋白酶的产生。  相似文献

采用响应面法对海洋微生物Pseudozymasp.SD301发酵产角鲨烯培养条件进行优化。在单因素实验结果的基础上,通过Design-Expert软件对葡萄糖浓度、接种量、培养温度、装液量进行4因素响应面实验设计,以角鲨烯产量为响应值,优化菌株Pseudozymasp.SD301的培养条件,并验证响应面预测值与实测值的一致性。结果表明,在葡萄糖浓度为67.2g·L-1、接种量为1.5%、培养温度为26℃、装液量在41mL/250mL的最优培养条件下,角鲨烯产量为1.152g·L-1,比出发条件提高了3.62倍,实测值与响应面预测值吻合良好。  相似文献

通过对降解甲壳素菌株 HD001 的发酵条件研究,确定了其产甲壳素酶最适培养基组分为 ( w/v ):( NH4 ) 2SO4 2.0%, NaCl 0.5%, K2HPO4 0.07%, KH2P O4 0.03%, MgSO4·7H2O 0.05% 以及葡萄糖 0.1%、酵母粉 0.3% 和胶体甲壳素 1.5%;最适产酶培养条件为:培养基起始 pH 值 6.0,培养温度 30 ℃,培养时间 120 h,发酵液酶活力达 376.2 U/mL.采用硫酸铵分级盐析、DEAE-纤维素离子交换层析、Sephacryl S-100 凝胶过滤层析对该酶进行纯化后,SDS-PAGE电泳显示单一条带,其相对分子质量约为 85.7 kDa.  相似文献

本研究的目的是利用聚苯胺修饰双室微生物燃料电池(MFC),从海水养殖废水中分离筛选阳极海洋产电菌,并利用响应面Box-Behnken设计优化各产电因子,提高产电性能。从海水养殖池塘的废水和污泥中分离出一株兼性厌氧海洋菌株,利用16S rRNA基因序列和API 20NE生化分析进行鉴定,采用扫描电镜进行菌株形态的观察。采用循环伏安法(CV)和电化学阻抗谱法(EIS)研究了不同阳极电极在MFC中的性能。采用Box-Behnken设计(BBD)对复合模式进行优化。结果显示:分离到的阳极海洋产电菌株鉴定为希瓦氏菌Shewanella sp.N3B_R,聚苯胺修饰阳极可以减少MFC的电荷转移电阻,输出电压和功率密度比未经修改的碳布电极分别提高了3倍和10倍。BBD结果表明,最适温度为35℃,NaCl和苯胺浓度分别为5 g/L和0.15 mol/L,得到的功率密度最高产率为522.1 mW/m2。  相似文献

为探讨专一性地降解甲壳质或壳聚糖的甲壳质酶或壳聚糖酶 ,从采集的土样中分离到 4株产壳聚糖酶能力较强的菌株 ,经摇瓶复筛 ,菌株YJ0 2产酶能力最强。对其发酵产酶条件的研究结果表明其产酶最适培养基组分为 (% ,w/v) :粉末壳聚糖 2 .0 ,葡萄糖 0 .1,NH4NO3 1.0 ,酵母提取物 0 .5 ,K2 HPO40 .0 7,KH2 PO40 .0 3 ,NaCl0 .5 ,MgSO4·7H2 O 0 .0 5 ,起始 pH 6.0。最适产酶培养条件是 :5 0 0mL三角烧瓶装瓶量为 15 0mL ,2 .0 % (v/v)接种量 ,3 0℃ ,15 0r/min培养 72h。在最适产酶培养条件下 ,72h时菌株YJ0 2发酵液中壳聚糖酶活力可达到 17.0 4U/mL。  相似文献

为寻找新型α-葡萄糖苷酶抑制(α-glucosidase inhibition,α-GI)活性化合物来源,针对前期研究发现的具有α-GI活性的海绵附生微生物坚强芽孢杆菌(Bacillus firmus),对其发酵培养条件进行优化,并对其活性组分中主要成分进行初步分析.以α-GI活性为主要检测对象,选出蛋白胨、酵母膏、Na Cl和p H值共4个因素进行正交优化试验.结合菌株生长情况和代谢产物产量指标,发现在MB培养基的基础上,蛋白胨质量浓度3 g/dm3、酵母膏质量浓度3 g/dm3、Na Cl质量浓度19.5 g/dm3、p H值为7.5的组合为最优培养条件.在优化后的培养条件下,规模化发酵该菌株,提取其代谢产物粗提物,经活性追踪下的二次色谱柱分离,获得一个高抑制活性组分,抑制率达82.4%.该组分经GC-MS分析后发现,活性组分中主要成分为环二肽类及哌啶酮类物质.本研究结果为寻找新型α-GI活性化合物提供了一个新的、可扩大生产的来源.  相似文献

经紫外诱变,筛选出在碳酸氢铵浓度为400 mg/L条件下可以生存的突变株。通过培养基中氮、磷、铁、温度、光照及pH值对雨生红球藻诱变株生长的影响进行比较,对以上3种营养盐及3种环境因子分别进行三因素三水平正交实验,以优化培养条件。进而对雨生红球藻野生型及诱变株的生长状况及色素含量进行比较。实验结果表明,雨生红球藻抗铵品系在培养基中NaNO30.1 g/L,KH2PO40.01 g/L,FeCl3.6H2O 1.0 mg/L,在pH值为8.0,光照强度100μmol.m-2.s-1,温度为18℃的培养条件下,雨生红球藻的生物量得到有效提高。  相似文献

对分离自腌鱼的高产菌红素(bacterioruberin)的菌株r-fish进行了形态特征、生理生化特性与细胞化学组分的分析以及基于16SrDNA序列的系统发育分析。形态特征、生理生化特性及细胞化学组分的分析结果表明菌株r-fish为球形的嗜盐古菌。基于16SrDNA序列的系统发育结果表明,r-fish与Halobacterium trapanicum NCIMB784菌株的相似性为99.8%,归入Na-trinema属。用TLC法对r-fish的色素进行了分析,r-fish的色素组分包括菌红素(bacteriorubefin)、脱水菌红素(anhydrobacteriorubefin)、双脱水菌红素(bisanhydrobacteriorubeftn)和一未知色素。与Halobacteriumhalobium和Halococcus morrhuae的色素组分相似。  相似文献

Response surface methodology was used to optimize a medium for a red-pigmented marine bacterium S-9801 strain (Flavobocterium sp.). In the first optimization step the influence of yeast extract, peptone, glucose and sodium chloride on pigment production was evaluated using a fractional factorial design. Pigment production was positively influenced by glucose and sodium chloride while other components had no significant effect. In the second step the path of steepest ascent was used to approach the optimal region of the medium composition. In the third step the optimal concentration of glucose and sodium chloride was determined by a central composite design and response analysis. The optimized medium allowed pigment production (A 535-650) to be increased from 0.137 to 0.559, being 320% higher than the original medium.  相似文献

于1993年8月,从山东省昌邑县下营镇患败血病的中国对虾血淋巴内分离到多株细菌,其中2株人工感染证实为病原菌;经50多项形态,生理,生化特性测试鉴定为产气弧菌Vibiogazogenes。其主要特性为:革兰氏阴性,弧状,极生单鞭毛,产生红色素,氧化酶呈阳性,精氨酸双水解酶,赖氨酸脱羧酶,鸟氨酸脱羧酶均为阴性,淀粉酶,明胶酶为阳性,硝酸盐还原阴性,利用木糖,水杨苷,山梨醇发酵葡萄糖产气,利用柠檬酸盐  相似文献

不同氮源对微小亚历山大藻生长和毒素产生的影响   总被引:12,自引:0,他引:12  
通过尿素、氯化铵、酵母浸出粉和硝酸钠等氮源对微小亚历山大藻(Alexandrium minutum)生长及毒素产生的影响研究,分析了微小亚历山大藻对不同氮源利用状况的差异.结果表明,在氮饥饿条件下,加入硝酸钠和酵母浸出粉能显著促进微小亚历山大藻的生长;高浓度的氯化铵在加入后对微小亚历山大藻有一定的毒性效应,表现为生长停滞,但毒性效应在5 d后消失,并得到与添加硝酸钠及酵母浸出粉相似的增长速率0.21 d-1;添加尿素对微小亚历山大藻的生长没有显著促进作用.在四种氮源中,尿素对微小亚历山大藻毒素产生的刺激作用也最弱,在稳定期每个细胞藻细胞毒素含量维持在6.00~8.00 fmol;添加硝酸钠、氯化铵和酵母浸出粉的藻细胞在稳定期毒素含量分别达到11.85,12.86和14.64 fmol.硝酸钠和氯化铵刺激藻毒素产生的效果比酵母浸出粉更为直接.四种含氮营养盐对微小亚历山大藻毒素组成的影响都很小.  相似文献

Abstract

Marine soft clay with a high salt concentration is widely distributed in coastal areas. In this study, cement-based composites consisting of cement, silica fume, plant ash and NaOH were used as a substitute for ordinary Portland cement, and the effect of salt (sodium chloride) on the strength development of clay was investigated by unconfined compressive strength (UCS) testing and scanning electron microscopy (SEM). With the addition of sodium chloride (NaCl), the amount of cementitious materials decreased, and the salt (sodium chloride) was considered to consume the cement-based composites. The consumption effect could be quantitatively evaluated by the consumption index of salt (CIS) and the clay-water/cement ratio hypothesis. The relationship between the CIS and curing period and an UCS prediction model of clay stabilized with cement-based composites with different salt contents and curing times were established. The CIS gradually decreased with increasing curing time and cement-based composites content. The accuracy of the prediction model was evaluated by a comparative analysis between the measured strengths and predicted strengths; the deviation was mostly within 10%. SEM analyses were employed to describe the changes in the microstructure of the specimens and the influencing mechanism of salt on clay stabilized with cement-based composites.  相似文献

Steel bar in concrete structures under harsh environmental conditions, such as chlorine corrosion, seriously affects its service life. Bidirectional electromigration rehabilitation (BIEM) is a new method of repair technology for reinforced concrete structures in such chloride corrosion environments. By applying the BIEM, chloride ions can be removed from the concrete and the migrating corrosion inhibit can be moved to the steel surface. In conventional engineering, the concrete structure is often configured with a multi-layer steel mesh. However, the effect of the BIEM in such structures has not yet been investigated. In this paper, the relevant simulation test is carried out to study the migration law of chloride ions and the migrating corrosion inhibitor in a concrete specimen with complex steel mesh under different energizing modes. The results show that the efficiency of the BIEM increases 50% in both the monolayer steel mesh and the double-layer steel mesh. By using the single-sided BIEM, 87% of the chloride ions are removed from the steel surface. The different step modes can affect the chloride ion removal. The chloride ions within the range of the reinforcement protective cover are easier to be removed than those in the concrete between the two layers of steel mesh. However, the amount of migrating corrosion inhibitor is larger in the latter circumstances.  相似文献

研究了饲料中添加氯化钠(0,0.05%,0.10%,0.20%,0.40%,0.80%,1.60%,3.20%,6.40%和12.80%)对体湿质量为1.041~1.104g凡纳滨对虾(Litopenaeusvannamei)的存活、生长和能量收支的影响。实验周期30d。结果表明:饲料中添加氯化钠显著地影响对虾的存活率、特定生长率和饲料转换效率,而对摄食量和吸收效率的影响不显著;在0.80%氯化钠水平,凡纳滨对虾的特定生长率显著高于0,3.20%,6.40%和12.80%氯化钠水平,但与其它处理差异不显著。  相似文献

The spatial distribution of the phytoplankton productivity was investigated in the Atlantic Sector of the Southern Ocean in the austral summer of 2009?C2010 and the spring of 2010 on the basis of field measurements. In October?CNovember, the average integrated primary production and the concentration of surface and integrated (photosynthetic layer) chlorophyll ??a?? in the Subantarctic waters of the Drake Passage exceeded the similar values along the Greenwich meridian by 3, 2, and 1.5 times, respectively. Similar primary production was observed in December?CJanuary for the water column in the eastern and western sectors of the studied area. The chlorophyll ??a?? concentration in the surface water layer was higher by 1.7 times for the Greenwich meridian area compared to the deep layers, but the concentrations of this pigment in the deeper layers did not differ. During the spring, the average primary production in the water column (the chlorophyll ??a?? concentration in the surface and in the photosynthetic layer) differed in the Drake Passage and along the Greenwich meridian by 2.3, 1.6, and 1.7 times, respectively. The opposite pattern was observed during the summer period, when the parameters listed above were higher for the Greenwich meridian area by 1.9, 2.5, and 1.7 times, respectively. Therefore, the western Antarctic areas in the spring are characterized by higher production than the eastern ones, and an opposite pattern is observed in the summer. The possible reasons for the meridional zonation of the chlorophyll ??a?? and the primary production are discussed in regard to different seasons.  相似文献
