A fragment of a large sub-unit ribosomal DNA (LrDNA) of 12 strains of Prorocentrum species was amplified by polymerase chain reaction (PCR). The PCR products were digested by 3 restriction endonucleases (Cfo Ⅰ, Hae Ⅲ, and RSA Ⅰ) and then resolved in agarose gels. Results show that different species had different RFLP patterns, except for P arcuatum (ME 131), which had the same pattern to P. micans (ME 160 and 04).The same fragment of 19 strains of the genus was also amplified and subjected to denaturing gradient gel electrophoresis (DGGE). 11 different patterns were resolved. Different cultures of a same species had the same pattern. The results of RFLP and DGGE analyses showed that eight newly isolated epibenthic Prorocentrum species were different from each other, and also from other cultured ones examined in this study. P arcuatum(MEI32) could not be differentiated from P. micans (MEI60 and 04), it was probably mis-identified, since they are quite different morphologically. P. redfieldii (MEI38) could also not be distinguished form P. triestinium(MEI32), it should be regarded as a synonym ofP. triestinium. 以中国生态系统研究网络CERN长武王东沟实验站的坡耕地和塬耕地为供试土壤,研究了黄土高原沟壑塬区旱耕地原状土壤氮素迁移和时空变异。结果表明:耕作土壤NO3--N为23.1~33.8 mg/kg,NH4+-N为0.23~0.50mg/kg,DOC为9.17~13.38 mg/kg,P为0.18~0.23 mg/kg;坡耕地和塬耕地的NO3--N和NH4+-N主要集中在0~20 cm的上层土壤,中下层土壤NO3--N和NH4+-N起源于上层淋溶迁移和累积;高含量时养分淋溶溶出呈指数衰减型;NO3--N、NH4+-N、DOC向下层的迁移塬耕地慢于坡耕地。  相似文献

Sediments and surface water were sampled in a tide flat in the Huiquan Bay, Qingdao, China in January 2004 to simulate the exchange of NH4-N/NO3-N/ PO 43- between sediments and surface water. A working system was designed with which samples were shaken at 60, 120 and 150 revolutions per minute (r/min). Experiment results show that NH4-N concentration in water at shaking rate of 60r/min decreased gradually, while at 120r/min increased gradually. In resuspension, fine-grained sediments contributed most NH4-N to the seawater, followed by medium-grained and coarse-grained sediments. The NO3-N concentration in water had a negative relation with the shaking rate; the medium-grained sediments contributed more NO3-N to seawater than the coarse-and fine-grained sediments. The PO 43- concentration is positively related with the shaking rate, the fine-grained sediments were the main N and P contributor to the seawater, followed by medium-and coarse-grained sediments.  相似文献

在实验室条件下研究半叶马尾藻对不同浓度N、P以及不同化合态N的吸收速率。结果显示:不同浓度N、P下,半叶马尾藻对N、P的吸收速率差异有统计学意义(P0.05);半叶马尾藻对N、P的吸收速率随N、P浓度的升高而增加,最大吸收速率出现在N为200μmol/L、P为20μmol/L时,分别为2.397和0.229μmol·g-1·h-1;不同氮磷比条件下,半叶马尾藻对N、P的最大吸收速率出现在氮磷比为10时,在氮磷比为1~30范围内,氮磷比对半叶马尾藻吸收N、P速率的影响有统计学意义(P0.05);不同化合态N对半叶马尾藻吸收N、P速率的影响有统计学意义(P0.05),当NH4+-N和NO3--N共存时,半叶马尾藻对这2种化合态氮可同时吸收利用,各处理组间,对总无机N的吸收速率差异有统计学意义(P0.05),在c(NO3--N)/c(NH4+-N)为2∶1和1∶2处有两个峰值,分别为2.036、1.862μmol·g-1·h-1,对P的最大吸收速率均出现在c(NO3--N)/c(NH4+-N)值为2∶1时,为0.206μmol·g-1·h-1。因此,在粤西沿海半叶马尾藻在消除养殖水域营养盐、防治海水富营养化方面有很大优势。  相似文献

Interactions between Skeletonema costatum (S. costatum) and Prorocentrum donghaiense (P. donghaiiense) were inves-tigated using bi-algal cultures at different concentrations of phosphate (PO4-P) and nitrate/phosphate (N/P) ratios. Experiments were conducted under P-limited conditions and the Lotka-Volterra mathematical model was used to simulate the growth of S. costatum and P. donghaiense in the bi-algal cultures. Both of these two species were inhibited significantly in bi-algal culture. The results of the simulation showed that the inhibitory degree of S. costaum by P. donghaiense was high when the concentration of PO4-P was low (0.1μmolL-1/2 d), but that of P. donghaiense by S. costaum was high with increased PO4-P supply (0.6μmolL-1/2 d). At low concen-tration of PO4-P (0.1μmolL-1/2 d), or high concentration of PO4-P (0.6μmolL-1/2 d) with high N/P ratio (160), the interactions be-tween S. costatum and P. donghaiense were dependent on the initial cell densities of both species. At high concentration of PO4-P (0.6μmolL-1/2 d) with low N/P ratio (25 or 80), S. costatum exhibited a survival strategy superior to that of P. donghaiense. The de-gree of inhibition of P. donghaiense by S. costaum increased with elevated N/P ratio when the medium was supplemented with con-centration 0.1μmolL-1/2 d of PO4-P. The degree of inhibition to P. donghaiense by S. costaum increased with elevated N/P ratio at low concentration of PO4-P (0.1 μmolL-1/2 d). This trend was conversed at high concentration of PO4-P (0.6μmolL-1/2 d). However, the degree of inhibition of S. costaum by P. donghaiense increased with the increased N/P ratio at different PO4-P concentrations (0.1μmolL-1/2 d and 0.6μmolL-1/2 d). These results suggested that both phosphate concentration and N/P ratio affected the competition between S. costaum and P. donghaiense: P. donghaiense is more competitive in environments with low phosphate or high N/P ratio and the influence of N/P ratio on the competition was more significant with lower phosphate concentration.  相似文献

The ratio of nitrogen/phosphorus (N/P) is known to affect cell proliferation of some marine micro algae. We evaluated the effect of N/P ratios on the proliferation and succession of phytoplankton using five marine micro algae species. Study on the coagulation of four species of red tide organisms(Nitzschia pungens,Skeletonema costatum,Prorocentrum minimum and Noctiluca scintillans)with montmorillonite,and the effect ofmontmorillonite pretreatment on the coagulation shows that the capability for montmorillonite to coagu-late with them is in the order:N.pungens>S.costatum>P.minimum>N.scintillans.Thecoagulation is discussed from the aspects of the structure,shape,size,movement,habit,etc.of differ-ent species and the results are explained theoretically.The experimental results also indicate that the treat-ment of montmorillonite with acid can enhance its coagulating capability.This is due to the fact that Al(OH_2)_6~(+3),exchanged from the clay lattice by hydrogen ion H~+,forms hydroxy-aluminum polymers on thesurface of the montmorillonite.The hydroxy-aluminum polymers positively charge and increase the positivecharacteristic of the clay surface,and also serves as a bridge between adjacent surfaces of particles.Thesetwo functions enhance  相似文献

Due to their particular physiology and life history traits, bryophytes are critical in regulating biogeochemical cycles and functions in alpine ecosystem. Hence, it is crucial to investigate their nutrient utilization strategies in comparison with vascular plants and understand their responses to the variation of growing season caused by climate change. Firstly, this study testified whether or not bryophytes can absorb nitrogen(N) directly from soil through spiking three chemical forms of 15N stable isotope tracer. Secondly, with stronger ability of carbohydrates assimilation and photosynthesis, it is supposed that N utilization efficiency of vascular plants is significantly higher than that of bryophytes. However, the recovery of soil N by bryophytes can still compete with vascular plants due to their greater phytomass. Thirdly, resource acquisition may be varied from the change of growing season, during which N pulse can be manipulated with 15N tracer addition at different time. Both of bryophytes and vascular plants contain more N in a longer growing season, and prefer inorganic over organic N. Bryophytes assimilate more NH4+ than NO3– and amino acid, which can be indicated from the greater shoot excess 15N of bryophytes. However, vascular plants prefer to absorb NO3– for their developed root systems and vascular tissue. Concerning the uptake of three forms N by bryophytes, there is significant difference between two manipulated lengths of growing season. Furthermore, the capacity of bryophytes to tolerate N-pollution may be lower than currently appreciated, which indicates the effect of climate change on asynchronous variation of soil N pools with plant requirements.  相似文献

A new planktonic dinoflagellate, Prorocentrum donghaiense I_a sp. nov., is described in the present paper. The water sample was collected from the Changjiang Estuary, the East China Sea. The species identification is based on shape, size, surface micro-morphology, ornamentation of thecal plates and the architecture of the pel‘iflagellar area and the intercalary bands as seen by fight and scanning electron microscope. Prorocentrum donghaiense Lu sp. nov. is compared with other prorocentrum species with respect to morphological characteristics and bloom behavior. It is not known whether Prorocentrum donghaiense Lu sp. nov produces phycotoxins like some other Prorocentrum species. Four other red tide species in the family Prorocentraceae (Dinophyceae), namely P. balticum, P. minimum, P.micans, P. triestinum, were examined and identified by light and scanning electron microscope. They have been recorded as bloom-forming species. Some aggregates of Prorocentrum are observed at the end of blooms. An event of strong discoloration caused by P. donghaiense could be detected by satellite sensor in the East China Sea in the late spring, of 1995.  相似文献

The dinoflagellates Alexandrium pacificum and Prorocentrum donghaiense are two wellknown harmful algal blooms(HABs)-forming species,both were usually found in the same sea areas in form of HABs in China.To date,there is no mechanistic model that can fully explain the occurrence of P.donghaiense blooms with A.pacificum.We found that different strains of P.donghaiense had different intrinsic growth rates of 0.107-0.215/d,and these strains exhibited different responses to the allelopathic action from A.pacificum.Some strains of P.donghaiense could grow well despite some degrees of inhibition in a short period,suggesting the two algal species P.donghaiense and A.pacificum could coexist,even if A.pacificum was allelopathic.Our findings may advance the understanding of phenotypes in P.donghaiense and provide a potential mechanism involved in the coexistence of P.donghaiense and A.pacificum in the same area.  相似文献

Benthic nutrient recycling in shallow coastal waters of the Bohai Sea   总被引:3,自引:0,他引:3  
Sediment-water fluxes of N and P species in the Bohai Sea were investigated in September-October 1998 and April-May 1999. The benthic fluxes of nutrient species were determined by incubating sediment core samples with bottom seawater bubbled with air or nitrogen. NO^-2,NH4, dissolved organic nitrogen (DON) and phosphorus (DOP), total dissolved nitrogen (TDN) and phosphorus (TDP), and PO4^3- showed a net exchange flux from seawater to sediment, while NO^-3, dissolved inorganic nitrogen (DIN) and SiO3^2- were released from sediment to seawater in the Bohai Sea. Sediment-water nutrient exchange increases DIN and reduces the phosphorus load in the Bohai Sea. The release of silicate from sediment to overlying seawater reduces potential silicate limitation of primary production resulted from decrease of riverine discharge.The exchange flux of nutrients showed no obvious seasonal variation. The present study showed that the concentrations and composition of nutrients in the water column were affected by suspended sediment, and that not all the exchangeable phosphate in sediment could be released via sediment resuspension.  相似文献

Algal allelopathy is an ecological/physiological phenomenon that has focused attention on the interactions among algae and the production of algal toxins. We investigated the allelopathic interactions between the dinoflagellate genus Prorocentrum micans and diatom genus Skeletonema costatum and between P. micans and dinoflagellate genus Karenia mikimotoi using bi-algal cultures. Because the effects were species-specific and size-dependent, we evaluated the effect of different initial densities. At low densities of P. micans and high densities of S. costatum inoculated into the same medium, the growth of P. micans was weakly restrained, whereas the growth of S. costatum was significantly suppressed. S. costatum and K. mikimotoi were strongly inhibited by P. micans, in both the bi-algal cultures and enriched filtrates. Direct cell-to-cell contact was not necessary to gain a competitive advantage, thus, our results suggest that P. micans inhibited the growth of S. costatum and K. mikimotoi by the release of allelochemical(s). Last, a mathematical model was used to simulate growth and interactions between P. micans and S. costatum and between P. micans and K. mikimotoi in bi-algal cultures.  相似文献

Microalgal lipids are regarded as main future feedstock of biofuels for its higher efficiency of accumulation and sustainable production.In order to investigate the effect of various nitrogen to phosphorus ratios on cells growth,chlorophyll content and accumulation of lipids in Dunaliella tertiolecta,experiments were carried out in modified microalgal medium with inorganic nitrogen(nitrate-nitrogen) or organic nitrogen(urea-nitrogen) as the sole nitrogen source at initial N:P ratios ranging from 1:1 to 32:1.The favorable N:P of 16:1 in the nitrate-N or urea-N medium yielded the maximum cell density and specific growth rate.Decrease in chlorophyll content were observed at the N:P of 4:1 in both nitrate-N and urea-N cultures.It was also observed that the maximum lipids concentration was obtained at the N:P of 4:1 in both nitrate and urea nutrient medium.The lipid productivity and lipid content of cultures in the urea-N medium at the N:P of 4:1were markedly higher than those from cultures with other N:P ratios(p 0.05).The results of this work illustrate the possibility that higher ratios of nitrogen to phosphorus have enhancing effect on cells growth of D.tertiolecta.Conversely,higher lipid accumulation is associated with a decrease in chlorophyll content under lower ratios of nitrogen to phosphorus.The results confirm the hypothesis of this study that a larger metabolic flux has been channeled to lipid accumulation in D.tertiolecta cells when the ratios of nitrogen to phosphorus drop below a critical level.  相似文献

This study on dynamic changes of culture color,astaxanthin and chlorophylls,inorganic N including N-NO3^-,N-NO2^- and N-NH4^ in batch culture of Haematococcus pluvialis exposed to different additive nitrate concentration showed(1)ast/chl ratio was over 0.8 for brown and red algae,but was usually less than 0.5 for green and yellow algae;(2)N-NO3^-,in general,was unstable and decreased,except for a small unexpected increase in nitrate enriched treatment groups;(3)measurable amounts of N-NO2^- and N-NH4^ were observed respectively with three change modes although no extemal nitrite and ammonia were added into the culture;(4)a non-linear correlation between ast/chl ratio(or color)changes and the levels of N-NO3^-,N-NO2^-,N-NH4^ in H.pluvialis culture;(5)up and down variation of the ast/chl ratio occurred simultaneously with a perceptible color change from yellow to brown(or red)when N-NO3^-,N-NO2^- and N-NH4^ fluctuated around 30,5,5μmol/L respectively;(6)existence of three dynamic modes of N-NO3^-,N-NO2^- and N-NH4^ changes,obviously associated with initial extemal nitrate;(7)the key level of total inorganic N concentration regulating the above physiological changes during indoor cultivation was about 50 μmol/L;and(8)0.5-10mmol/L of nitrate was theoretically conducive to cell growth in batch culture.  相似文献
