为了研究不同温度和N、Fe3+质量浓度对微拟球藻脂肪酸组成的影响,采用气相色谱-质谱联用仪,测定了不同处理条件下微拟球藻脂肪酸的组成。并利用荧光定量PCR技术,监测了在不同处理条件下微拟球藻硬脂酰辅酶A去饱和酶(SCD)基因的表达差异。结果表明:在缺N的条件下,微拟球藻中的不饱和脂肪酸的比例如C18∶2、C18∶1和C20∶4明显降低;在富N条件下C18∶2、C20∶4和C20∶5等不饱和脂肪酸的比例明显提高;在高温处理下,微拟球藻中C16∶1和C18∶1等单不饱和脂肪酸占总脂肪酸的比例有显著提高,而C20∶4和C20∶5等多不饱和脂肪酸则有明显的降低趋势;在低温处理组中,多不饱和脂肪酸如C18∶2和C20∶4在微拟球藻总脂中的比例明显提高;在富Fe条件下,多不饱和脂肪酸尤其是C20∶4和C20∶5发生积累;在缺Fe条件下,不饱和脂肪酸如C18∶1、C20∶5等占总脂肪酸的比例明显下降。与此同时,在富N、低温、高温和富Fe的条件下,SCD基因的表达量有显著的提高;在缺N和缺Fe的条件下,SCD基因的表达量下降。这揭示了SCD基因在不饱和脂肪酸形成途径中所起的重要作用,同时也证明了基因的表达与产物的合成有着密切的联系。  相似文献

为研究日本囊对虾(Marsupenaeusjaponicus)耐高亚硝酸盐的分子调控机制,利用新一代高通量Illumina测序技术,对日本囊对虾在高亚硝酸盐胁迫下的肝胰腺进行转录组测序,通过对高质量序列的拼接组装以及功能基因的注释和分类,发掘与耐高亚硝酸盐相关性状的候选基因。实验结果表明:①10个文库共获得920785608个净读本,数据量为6.48G~7.34G。②Q30>93.07%。利用 Trinity软件组装后获得 46308条单基因簇,N50为1833bp。③与对照组相比,高亚硝酸盐组分别识别出593、606、1089、497和988个差异表达基因。④对 DEGs进行功能注释,得到与免疫和代谢相关的通路和基因。⑤采用 qPCR 对随机选择的9个差异基因(DPD、ABCH、ProPOb、ACADL、CYP2J、PAT4、BHMT、CLEC 和PEPCK)在高亚硝酸盐胁迫下的表达情况进行检测,其表达量与转录组测序技术(RNASequencing,RNA-seq)趋势一致。本研究结果丰富了对虾cDNA 数据库的信息,为日本囊对虾在高亚硝酸盐胁迫下的分子机制研究奠定了基础。  相似文献

海洋微藻的无菌化处理及对其生长特性和生化组成的影响   总被引:2,自引:0,他引:2  
利用常用抗生素(氨苄青霉素、链霉素、氯霉素和卡那霉素)对4株海洋微藻(1株金藻和3株小球藻)进行了无菌化处理,并对这4株微藻在带菌和无菌培养时的生长特征和生化组成进行了比较。结果表明:中高浓度(≥100 mg/dm3)的单种抗生素或抗生素组合可抑制微藻的生长,个别低浓度(50 mg/dm3)的抗生素或抗生素组合可促进微藻的生长;氨苄青霉素、链霉素和卡那霉素各200 mg/dm3联合使用处理球等鞭金藻,氯霉素100, 50, 50 mg/dm3单独使用分别处理小球藻C95, CV和C146,可获得这4株微藻的无菌藻系;无菌培养的4株海洋微藻的稳定期持续时间由除菌前的4~5 d延长到15~20 d,藻液悬浮性增强,藻细胞不易老化;除菌后微藻的主要生化成分发生明显变化,球等鞭金藻的叶绿素A、叶黄素和总脂含量显著增加,小球藻C95和CV的总蛋白含量显著增加,3株小球藻的脂肪酸组成如EPA, PUFA等的含量发生显著变化。  相似文献

为初步了解长江口邻近海域可降解合成聚酯菌株的多样性,本研究采用含聚己内酯(polycaprolactone,PCL)和甘油三丁酸酯(Tributyrin)的固体培养基,利用透明圈法从长江口邻近海域不同时间表层沉积物的菌种库中筛选具有降解PCL和Tributyrin活性的菌株。本研究共获得480株具有PCL或Tributyrin降解活性的菌株,其中能同时降解PCL和Tributyrin的445株,仅能降解Tributyrin的34株,仅能降解PCL的1株,分别占全部菌株数的92.71%、7.08%、0.22%。分类研究表明,这些菌株分属于14个属50个种,2、5、7、9月获得的可降解PCL和Tributyrin菌株的优势属分别为Psychrobacter、Bacillus、Vibrio、Vibrio。其中弧菌属在5、7、9月均有出现,尤其是在7月占比较大。多样性指数分析表明长江口近海表层沉积物中合成聚酯降解菌生物多样性5月2月9月7月;Tributyrin降解菌生物多样性高于PCL降解菌生物多样性。研究结果表明,中国长江口临近海域表层沉积物中PCL、Tributyrin降解菌种类丰富,在该区域塑料生物降解中发挥重要作用。  相似文献

In most bacteria,plants and algae,fatty acid biosynthesis is catalyzed by a group of freely dissociable proteins known as the type II fatty acid synthase(FAS II) system.In the FAS II system,enoylacyl carrier protein reductase(ENR) acts as a determinant for completing the cycles of fatty acid elongation.In this study,the cDNA sequence of ENR,designated as IgENR,was isolated from the microalga Isochrysis galbana CCMM5001.RACE(rapid amplification of cDNA ends) was used to isolate the full-length cDNA of IgENR(1 503 bp),which contains an open reading frame(ORF) of 1 044 bp and encodes a protein of 347 amino acids.The genomic DNA sequence of IgENR is interrupted by four introns.The putative amino acid sequence is homologous to the ENRs of seed plants and algae,and they contain common coenzymebinding sites and active site motifs.Under different stress conditions,real-time quantitative polymerase chain reaction(RT-qPCR) showed the expression of IgENR was upregulated by high temperature(35℃),and downregulated by depleted nitrogen(0 mol/L).To clarify the mechanism of lipids accumulating lipids,other genes involved in lipids accumulation should be studied.  相似文献

Defensin is one of preserved ancient host defensive materials formed in biological evolution. As a regulator and effector molecule, it is very important in animals' acquired immune system. This paper reports the defensin gene from the mixed liver and kidney cDNA library of abalone Haliotis discus hannai Ino. Sequence analysis shows that the gene sequence of full-length cDNA encodes 42 mature peptides (including six Cys), molecular weight of 4 323 Da, and pI of 8.02. Amino acid sequence homology analysis shows that the peptides are highly similar (70% in common) to other insects defensin. Because of a typical insect-defensin structural character of mature peptide in the secondary structure, the polypeptide named Haliotis discus defensin (hd-def), a novel of antimicrobial peptides, belongs to insects defensin subfamily. The RT-PCR result of Haliotis discus defensin shows that the gene can be expressed only in the hepatopancreas by Gram-negative and positive bacteria stimulation, which is ascribed to inducible expression. Therefore, it is revealed that the Haliotis discus defensin gene expression was related to the antibacterial infection of Haliotis discus hannai Ino.  相似文献
