为了探讨微生物多糖剂量效应在水产动物对不良环境刺激反应中的作用,本研究利用从南极冰海水中分离得到的南极细菌Pseudoalteromonas sp.3-3-1-2产生的胞外多糖,采用注射不同浓度的胞外多糖溶液的方式处理刺参,定期记录其生长状况并测定体腔液免疫相关指标的变化。结果表明,南极细菌胞外多糖处理后的刺参保护率升高,即较高浓度(4mg/mL)的细菌多糖可以有效降低刺参的死亡率。细菌多糖可以降低体腔液中MDA含量,虽然体腔液ACP、CAT和SOD活性在处理前期有所受到抑制,但是试验后期细菌多糖处理明显增强了上述免疫酶活性,以保护细胞免受氧自由基诱导的氧化损伤。处理过程中刺参体腔液的溶菌酶活性降低,但是细菌多糖的处理减弱了溶菌酶活性下降的幅度。此外,与2mg/mL处理组相比,4mg/mL处理组对刺参体腔液免疫酶的促进作用稍弱,但无显著差异。研究结果表明,极地细菌胞外多糖可对刺参产生一定的免疫保护作用,从而提高刺参逆境下的生长活力。  相似文献

为了探讨胞外多糖在南极微生物低温适应性中的作用与机制,克隆了南极菌Pseudoalteromonas sp.S-15-13的引导糖基转移酶(GTF)核心片段,并采用Real-Time PCR的方法研究了温度、冻融循环、盐浓度、pH等条件对该糖基转移酶基因(gtf)表达的影响。结果表明,低温有利于gtf的表达,短时的低温刺激(2℃)1 h后,gtf的表达量即上调为对照的1.5倍;而该菌经4和10℃培养24 h后gtf的表达量约为20℃时的8~12倍;经过冻融循环gtf的表达量上调,在第2个冻融循环后gtf 的表达量较对照提高了3.667倍;盐浓度对gtf的影响表现为低促高抑,即NaCl含量为6.0 %时gtf 的表达量是对照(3.0%)的3.59倍,当NaCl含量达9.0 %时gtf 表达量则显著下调;在一定范围内(pH5.0~8.0),pH的改变会促进gtf 的表达,当pH为6.0时gtf 表达量约为pH7.0时的2倍。该结果为探讨胞外多糖在南极微生物环境适应性中的作用与机制提供了科学依据。  相似文献

采用苯酚硫酸法和醇沉淀法,从600余株南极微生物中筛选得到10株胞外多糖产量较高的菌株.分子鉴定与系统发育分析表明,9株菌株属于假交替单胞菌属(Pseudoalteromonas),1株属于嗜冷杆菌属(Psych robacter).对10株菌株进行发酵培养,发酵液经醇沉、除蛋白、冷冻干燥后得到粗胞外多糖.其中菌株3-3-1-2的胞外多糖产量达0.6 g/L.对粗多糖产量较高的4株菌所产的胞外多糖进行免疫活性试验,粗胞外多糖20#3在注射3d时,实验组AKP活性比对照组增加了39%,CAT活性比对照组增加了24%,二者均差异显著(P<0.05),注射5d时,ACP活性比对照组增加了20%,差异显著(P<0.05),总超氧化物歧化酶(T-SOD)活性比对照组增加了14%,差异极显著(P<0.01);粗胞外多糖33-1-2在注射3d时,实验组AKP活性比对照组增加了12%,差异不显著(P>0.05),注射5d时,酸性磷酸酶(ACP)活性比对照组增加了24%,过氧化氢酶(CAT)活性比阳性对照组增加了5%,二者均差异显著(P<0.05).研究结果表明,菌株20#3与3-3-12产生的胞外多糖对大菱鲆(Scophthalmus maxoimus)具有显著的免疫促进作用.由此可知,筛选出的极地细菌胞外多糖20#3和3-31-2可显著提高大菱鲆非特异性免疫力,可作为水产饲料添加剂应用于大菱鲆的生产养殖.  相似文献

鳗弧菌(Vibrio anguillarum)是一种水产动物的重要致病菌,鳗弧菌分泌的胞外蛋白酶是该菌的致病因子之一.研究了环境因子对鳗弧菌W-1生长及胞外蛋白酶表达的影响.结果表明,鳗弧菌的最适生长温度为28℃,菌体生长量在21 h达到最高,胞外蛋白酶的活力在24 h最高.在37℃培养时,菌体生长和酶活力显著降低;添加葡萄糖、蔗糖、麦芽糖等碳源物质均能显著促进菌体的生长,但对于胞外蛋白酶表达有明显抑制作用;1 mmol/L EGTA对茵体的生长和胞外蛋白酶表达没有明显影响,而1 mmol/L的EDTA强烈抑制鳗弧菌的生长及其胞外蛋白酶的表达.添加不同浓度的氨苄青霉素对菌体的生长及其胞外蛋白酶表达有明显的抑制作用.  相似文献

4种海洋浮游硅藻(牟勒氏角毛藻、海链藻、三角褐指藻和新月菱形藻)培养在改进的f/2培养基中,研究了不同氮、磷和硅营养水平对它们胞外多糖产量的影响.结果表明,硅藻胞外多糖的生产和释放具有种间特异性,角毛藻和海链藻胞外多糖的生产和释放主要在静止期,而三角褐指藻和新月菱形藻在指数生长期前期和静止期都能生产和分泌较高的胞外多糖;培养液中低浓度磷减少了4种硅藻在静止期胞外多糖的产量,但增加了角毛藻在生长期前期胞外多糖的产量;氮浓度的降低增加了三角褐指藻在指数生长前期胞外多糖的产量,但减少了其他3种藻类胞外多糖的产量;硅浓度的降低对4种硅藻胞外多糖的产量影响不大,在一定程度上还促进了静止期胞外多糖的生产.本研究表明,营养盐水平对硅藻胞外多糖生产的影响因种类和细胞所处生长期不同而存在着很大的差异.  相似文献

B-3胞外多糖是从一株南极嗜冷菌Psychrobacter sp.B-3发酵液中提取的胞外多糖,前期研究发现该多糖能够激活巨噬细胞并影响其免疫调控活动。为探明其免疫调控途径,采用转录组学分析对经B-3多糖刺激的RAW264.7巨噬细胞系进行了差异表达基因分析。研究结果表明B-3多糖的刺激引起了420个基因的差异表达,其中上调178个、下调242个。差异表达基因与细胞功能存在诸多关联,但主要集中在代谢和免疫两个通路上:其中在多个免疫相关通路上,富集最显著的是抗原处理和表达途径;另一个明显富集的则是代谢通路,B-3多糖处理引起了大量参与代谢的酶类的表达变化,包括氨基酸,碳水化合物,脂质和核苷酸代谢相关基因均发生了一定程度的改变。因此,B-3胞外多糖作为一种免疫激活剂直接影响了RAW264.7巨噬细胞系的免疫功能,同时也对其胞内的代谢途径产生了影响。本论文首次开展了南极嗜冷细菌胞外多糖对RAW264.7巨噬细胞系的影响,该研究为不同来源的活性多糖对巨噬细胞免疫激活研究提供了科学依据。  相似文献

研究南极树粉孢属(Oidiodendron truncatum)真菌所产胞外多糖的理化性质和结构特征。采用阴离子交换柱层析和凝胶柱层析对南极树粉孢属(Oidiodendron truncatum)真菌胞外多糖进行了系统的分离纯化,从中得到4个均一的组分(AFW1,AFW2,AFS1和AFS2),并结合红外、气质及核磁等技术对它们的结构进行了解析。结果表明:AFW1和AFW2为中性多糖,AFS1和AFS2中含有少量的蛋白;AFW1和AFS1是由甘露糖、葡萄糖和半乳糖3种单糖组成的结构复杂的杂多糖;AFW2和AFS2则仅由葡萄糖组成,其中AFW2属线性(-1,6-葡聚糖,而AFS2属于C-3位有分支的α-1,6-葡聚糖。  相似文献

以具有明显海洋特性的、产胞外免疫活性多糖的链霉菌2305菌株作为发酵菌种,对其胞外多糖的发酵工艺及其条件进行了研究,考察了不同碳源、氮源、水质、培养基初始pH值、发酵温度、通气量等因素对发酵菌种的生物量及胞外多糖产量等的影响,建立和优化了胞外多糖摇瓶发酵培养基配方、发酵工艺及发酵条件,胞外多糖产量达到4.58g/L,转化率达到6.90%。研究了发酵过程中菌体生物量、底物糖残余量、pH值、产物多糖积累量等的动态变化,分析讨论了2305菌株胞外多糖发酵的动力学类型。  相似文献

紫球藻胞外多糖抗氧化和免疫调节活性的研究   总被引:1,自引:0,他引:1  
采用密闭微波法降解紫球藻胞外多糖。通过测定多糖对羟自由基OH·、超氧阴离子O2-·和二苯代苦味酰基自由基DPPH·的清除效果及对小鼠单核-巨噬细胞RAW264.7和淋巴细胞的增殖效果,考察低分子量紫球藻多糖的体外抗氧化活性和免疫调节活性。结果表明,在一定的浓度范围内,降解后的紫球藻多糖对OH·、O2-·和DPPH·的清除能力与浓度呈正相关性,它们的半抑制率IC50分别为0.619,0.114和0.015mg/mL,表明紫球藻胞外多糖具有体外抗氧化活性。溶液浓度在50~200μg/mL的多糖都表现出体外免疫促进作用,与对照组相比,100μg/mL的多糖对巨噬细胞RAW264.7的促进作用最大,其增值指数为1.72。200μg/mL的多糖对脾淋巴细胞的增值指数为2.11。  相似文献

Polar regions are poorly understood components of global biogeochemical cycles. This paper discusses the influences of nitrate and ammonium concentrations on nitrate uptake (and hence new production), particularly with regard to data collected within marginal ice zones in the Arctic and Antarctic. Subsurface ammonium maxima in waters over 150 m are frequently encountered in the Arctic and occasionally in the Antarctic. Such maxima result from the heterotrophic remineralization of organic matter, and because stratified environments occur more frequently in the Arctic, significant concentrations of ammonium accumulate as a result of lower diffusive losses. Causal agents (bacteria vs. Zooplankton) may also be different in the Arctic from those in the Antarctic. Elevated ammonium concentrations significantly reduce nitrate uptake, and it is suggested that this nutrient interaction may play a significant role in controlling new production, particularly in open water regions. The new production of the Southern Ocean is estimated, and it is suggested that the production and flux from the surface layer is significant relative to the world's oceans.  相似文献

南极大磷虾(Euphausia superba)为南大洋生态系统中的关键种,也是南极生态系统食物网中的重要枢纽。该种秋冬季转换期的营养信息对于理解其知之甚少的越冬机制非常重要。但关于此方面的少数研究在时空变化上仍存在着差异。为此,我们调查了南极半岛秋季(4-5月)和冬季(6月)磷虾成体δ13C和δ15N值的个体、月份及区域性差异。我们的目标旨在检验该期间磷虾的营养变化以及磷虾与其在南极海洋生态系统中的摄食环境之间的关系。结果如下:(1)磷虾δ13C值与体长之间无显著关系,但δ15N值与体长之间则存在显著相关性;(2)秋季磷虾δ13C值呈现增长趋势,但冬初季节并无显著变化,此期间δ15N值无显著不同;(3)布兰斯菲尔德与南设得兰群岛之间的δ15N平均值显著不同。我们的数据表明南极半岛秋至初冬转换期间磷虾成体营养呈现个体、季节性及区域性变化。  相似文献

基于中国第30次南极科学考察在南极半岛(60°~63°S)近岸海域获取的调查资料,分析了该海域生物化学要素中溶解有机碳(DOC)、总氮(TN)和总磷(TP)分布特征并讨论地形和水团对其的影响。结果表明:2014年夏季南极半岛近岸海域水体DOC浓度变化范围为40.5~78.1μmol/L,平均浓度为66.3μmol/L;TN浓度变化范围为4.2~29.5μmol/L,平均浓度为14.9μmol/L;TP浓度变化范围为0.8~2.9μmol/L,平均浓度为2.0μmol/L。表层DOC呈现研究海域西北部D1断面和东南部D5断面浓度较高,中部DOC浓度较低;表层TN与TP浓度高值区出现在研究海域西部D1断面北部以及南部,中部和东部浓度较低;DOC,TN和TP浓度的垂直分布与海底地形和水团交汇密切相关,水团运动受阻于地形致使生物化学要素在垂直方向再分布。DOC,TN和TP空间分布反映了南极半岛近岸海域生物化学要素复杂的流通,将为合理开发和利用南极资源及环境影响评价提供科学依据。  相似文献

南极海冰的生消冻融与全球气候变化息息相关,多年以来受到国际社会的高度关注。目前已有诸多关于南极海冰范围变化的研究,但大部分是基于卫星遥感影像来展开分析。Argo观测网遍布全球各大洋,为海冰范围研究提供了一种新的思路,即根据浮标GPS点的南部边界推算南极海冰边界,由浮标的年、月累积数据得到南极海冰范围的年际、月际变化规律。对于这种研究思路提出了三种实现方法:(1)绘制专题图,可以清晰直观地看到南极海冰在2—9月间的生消变化情况;(2)利用南极附近浮标GPS点数量占全球比例的变化情况来分析海冰变化规律,在月际变化趋势上与影像数据一致,在年际变化上稍显不足;(3)使用点密度分析方法估计海冰边界,建立基于浮标GPS点密度的海冰-海水分界模型,可得到南极海冰范围变化规律的定量分析结果。浮标数据与影像数据互为补充,可为全球气候变化研究提供更多参考。  相似文献

As one of the most common and dominant species in the Southern Ocean, Antarctic krill(Euphausia superba)play a significant role in food web structure and the process of energy flow. The diet of Antarctic krill in the Prydz Bay during austral summer of 2012/2013 was investigated and the ontogenetic shift in krill diet was evaluated using the stable isotope method. The nitrogen stable isotope values(δ~(15) N) of adults((2.78±0.58)‰) were much higher than those of juveniles((1.69±0.70)‰), whereas the carbon stable isotope values(δ~(13) C) of adults(–(28.26±1.08)‰) were slightly lower than those of juveniles(–(27.48±1.35)‰). Particulate organic matter(POM)from 0, 25, and 50 m depth combined(0/25/50 m) represented phytoplankton food items. The results showed that phytoplankton food items in surface water and mesozooplankton were two essential food items for Antarctic krill in the Prydz Bay during summer. POM(0/25/50 m) contributes 56%–69% and 26%–34% to the diet of juvenile and adult krill, respectively, whereas mesozooplankton composes 13%–34% and 58%–71% of the diet of juvenile and adult krill, respectively. Thus, an ontogenetic diet shift from POM(0/25/50 m), which consists mainly of phytoplankton, to a higher trophic level diet containing mesozooplankton, was detected. The capacity for adults to consume more zooplankton food items may minimize their food competition with juveniles, which rely mostly on phytoplankton food items. This suggests "diet shift with ontogeny" which may somehow help krill keep their dietary energy budget balanced and well adapted to the Antarctic marine ecosystem as a dominant species.  相似文献

The location of the GAVDOS facility is under a crossing point of the original ground-tracks of TOPEX/Poseidon and the present ones for Jason-1, and adjacent to an ENVISAT pass, about 50 km south of Crete, Greece. Ground observations and altimetry comparisons over cycles 70 to 90, indicate that a preliminary estimate of the absolute measurement bias for the Jason-1 altimeter is 144.7 ± 15 mm. Comparison of Jason microwave radiometer data from cycles 37 and 62, with locally collected water vapor radiometer and solar spectrometer observations indicate a 1-2 mm agreement.  相似文献

An unusual region of high meso-scale turbulence has been identified in the Indian Ocean sector of the Southern Ocean. It has been shown that this is the result of eddy shedding from the Antarctic Polar Front. These eddies may dramatically affect the local distribution of marine organisms. To investigate this, the euphausiid community structure and species composition in the region of a cold eddy within the Antarctic Polar Frontal Zone (APFZ) was investigated during April 2005. Water masses within the core of the eddy were typically Antarctic, showing they had come from south of the Antarctic Polar Front. Results of numerical analyses indicate that the euphausiid community within the survey area consisted of three distinct groups: those in APFZ waters, those at the edge of the eddy and those in the core of the eddy. These results indicate that eddies generated by the interaction of the Antarctic Circumpolar Current with the South-West Indian Ridge play an important role in transporting Antarctic euphausiid species equatorward, thus contributing to the spatial heterogeneity of the zooplankton community within the region.  相似文献

An oxygen minimum zone (OMZ) currently exists at intermediate water depths on the northern Japanese margin in the northwestern Pacific. The OMZ results largely from a combination of high surface–water productivity and poor ventilation of intermediate waters. We investigated the late Quaternary history (last 27 kyr) of the intensity of this OMZ using changes in benthic foraminiferal carbon isotopes and assemblages in a sediment core taken on the continental slope off Shimokita Peninsula, northern Japan, at a water depth of 975 m. The core was located well within the region of the present-day OMZ and high surface–water productivity. The benthic foraminiferal δ13C values, which indicate millennial-scale fluctuations of nutrient contents at the sediment–water interface, were 0.48‰ lower during the last glacial maximum (LGM) than during the late Holocene. These results do not indicate the formation of glacial intermediate waters of subarctic Pacific origin, but rather the large contribution of high-nutrient water masses such as the Antarctic Intermediate Water, implying that the regional circulation pattern during the LGM was similar to that of modern times. Benthic foraminiferal assemblages underwent major changes in response to changes in dissolved oxygen concentrations in ocean floor sediments. The lowest oxygen and highest nutrient conditions, marked by dysoxic taxa and negative values of benthic foraminiferal δ13C, occurred during the Bølling/Allerød (B/A) and Pre-Boreal warming events. Dysoxic conditions in this region during these intervals were possibly caused by high surface–water productivity at times of reduced intermediate–water ventilation in the northwestern Pacific. The benthic assemblages show dysoxic events on approx. 100- to 200-year cycles during the B/A, reflecting centennial-scale productivity changes related to freshwater cycles and surface–water circulation in the North Pacific.  相似文献
